Yellow fever beyond the Amazon: surveillance challenges and the role of Callithrix spp. in sylvatic cycles in Brazil.

Yellow fever virus (YFV) remains a major public health concern in tropical and subtropical regions of the Americas and Africa. Since late 2016, sylvatic yellow fever (YF) outbreaks and sporadic cases affecting both humans and non-human primates (NHP) have been recorded in Southern Brazil. These events raised concerns about viral persistence beyond the Amazon Basin, where YFV is endemic, and the potential reemergence of urban transmission. Neotropical NHPs are central to the ecology of sylvatic YF, but many aspects of their infection dynamics and species-specific roles remain poorly understood.

Entomo-virological investigation during the epizootic outbreak of sylvatic yellow fever in Rio Grande do Sul, Brazil, between 2021 and 2022.

BACKGROUND: Yellow fever virus (YFV) re-emerged among non-human primates (NHPs) in Rio Grande do Sul in early 2021, more than a decade after its last detection in the state. The spread of the virus was accompanied by increased mortality among NHPs. OBJECTIVES: To conduct entomological surveillance and molecular detection of YFV and other Orthoflavivirus species in mosquito samples collected from affected and potentially receptive areas. METHODS: Mosquitoes were collected during epizootics using human landing catches, BG-Pro traps, and ovitraps. Virus detection was performed using reverse tra

Evolution and spillover dynamics of yellow fever at the forest-urban interface in Brazil.

Yellow fever virus (YFV) continues to threaten human and wildlife populations in the Americas, yet its transmission at the forest-urban interface remains unclear. Here we integrate ground- and canopy-level mosquito surveillance, systematic monitoring of non-human primate carcasses and viral metagenomics to describe the dynamics of a sylvatic YFV outbreak in a 186-hectare Atlantic Forest fragment embedded within metropolitan São Paulo, Brazil, between 2017 and 2018. Our analyses reveal that transmission was primarily driven by a single genetic cluster introduced during a period of high abundanc

Assessing the Sylvatic Yellow Fever Vectors in Southern Brazil.

Yellow fever (YF) is an infectious disease caused by the yellow fever virus (YFV), an arbovirus of the Flaviviridae family. It is transmitted through the bite of infected mosquitoes of the Culicidae family and affects both humans and non-human primates (NHPs). This study aimed to investigate the sylvatic Culicidae fauna and the occurrence of natural YFV infection in a microregion of southern Santa Catarina, Brazil, an area recently affected by a sylvatic YF outbreak. Entomological collections were conducted between January and February 2023 in five municipalities with confirmed viral circulati

Mosquito fauna (Diptera: Culicidae) in an Atlantic Forest remnant urban park and report of Chikungunya virus RNA detection in sylvatic mosquitoes.

Patches of Atlantic Forest within urban areas in Brazil may provide suitable conditions for the establishment of transmission cycles of emerging alphaviruses and orthoflaviviruses due to the presence of potential hosts, such as non-human primates, and competent vectors. This study aimed to characterize the diurnal mosquito fauna and detect arboviruses in an Environmental Protection Area in the city of Aracaju, Northeast Brazil, where dengue, zika, and chikungunya are endemic. Entomological surveys and molecular assays were conducted to detect viral infection in adult mosquitoes. Ovitraps were

Drivers of sylvatic yellow fever transmission in southeast Brazil: a joint epidemiological and phylodynamic inference.

BACKGROUND: Yellow fever virus (YFV) has caused substantial human disease in Brazil, driven by spillover from outbreaks in nearby non-human primates (NHPs). As disease surveillance in NHPs is challenging, the impact of environmental conditions on YFV outbreaks in NHPs remains poorly understood. In this study, we aimed to use joint inference modelling to overcome these obstacles by integrating diverse data streams and to estimate YFV transmission dynamics in NHPs to enhance understanding of yellow fever disease ecology. METHODS: We applied a multistage analysis using epidemiological, phylogene

Group size and composition of brown howler monkeys (Alouatta guariba) in three fragments of Araucaria Pine Forest in southern Brazil.

Brown howler monkeys (Alouatta guariba) are increasingly threatened by habitat loss, sylvatic yellow fever, and negative human interactions. In southern Brazil, brown howlers still inhabit some fragments of the endangered Araucaria Pine Forest (APF), but basic information on their social organization remains scarce. We surveyed brown howler monkey groups to assess their group size and composition in three nearby similarly sized peri-urban APF fragments in Curitiba, southern Brazil. Using Generalized Linear Mixed Models, we investigated whether the number of males, females and immatures could b

Species Composition and Ecological Aspects of Immature Mosquitoes (Diptera: Culicidae) in Phytotelmata in Cantareira State Park, São Paulo, Brazil.

Phytotelmata are aquatic microenvironments formed by the accumulation of water and organic matter in cavities of plants. These microenvironments serve as breeding sites for various species of mosquitoes, including some of epidemiological importance. Our objective was to identify the mosquito fauna in these microenvironments and to analyze variations in mosquito fauna diversity between bromeliads, tree holes, and bamboo internodes in Cantareira State Park, São Paulo (CSP), Brazil, where there have been reports of yellow-fever epizootics in non-human primates and circulation of plasmodia. Uncovering Hidden Transmission: Active Surveillance Reveals Cryptic Circulation of Yellow Fever Virus in Urban Marmosets in Belo Horizonte, Brazil, 2024.

Between 2016 and 2018, the state of Minas Gerais, Brazil, experienced its most significant yellow fever (YF) outbreak in 80 years. Yellow fever virus (YFV) circulation persisted afterward, with continued non-human primate (NHP) epizootics and, recently, human cases. In June 2024, YFV RNA was detected in a dead marmoset (Callithrix penicillata) in an urban square in Belo Horizonte (BH), prompting a field investigation in an adjacent park to assess infection in potential mosquito vectors and NHPs. A total of 250 mosquitoes representing nine species were collected at ground and canopy level, of w

First report on the diversity and geographical distribution of mosquitoes of the genera Haemagogus and Sabethes (Diptera: Culicidae), the main vectors of jungle yellow fever in Paraguay.

Sylvatic yellow fever virus is transmitted by mosquitoes Haemagogus and Sabethes genera, which predominantly inhabit forested areas but can also be found near human dwellings in rural zones. In Paraguay, outbreaks of the disease were reported between 2007 and 2008, prompting the initiation of systematic sampling starting from those years to monitor the vectors transmitting the yellow fever virus. This study aims to identify the primary vector species and determine their geographic distribution in Paraguay, based on administrative divisions and ecoregions. Sampling was conducted in 17 departmen

Evaluation of humoral immune response after yellow fever infection: an observational study on patients from the 2017-2018 sylvatic outbreak in Brazil.

Between 2016 and 2018, Brazil experienced major sylvatic yellow fever (YF) outbreaks that caused hundreds of casualties, with Minas Gerais (MG) being the most affected state. These outbreaks provided a unique opportunity to assess the immune response triggered by the wild-type (WT) yellow fever virus (YFV) in humans. The plaque reduction neutralization test (PRNT) is currently the standard method to assess the humoral immune response to YFV by measuring neutralizing antibodies (nAbs). The present study aimed to evaluate the humoral immune response of patients from the 2017-2018 sylvatic YF out

Development and validation of RT-LAMP for detecting yellow fever virus in non-human primates samples from Brazil.

Monitoring yellow fever in non-human primates (NHPs) is an early warning system for sylvatic yellow fever outbreaks, aiding in preventing human cases. However, current diagnostic tests for this disease, primarily relying on RT-qPCR, are complex and costly. Therefore, there is a critical need for simpler and more cost-effective methods to detect yellow fever virus (YFV) infection in NHPs, enabling early identification of viral circulation. In this study, an RT-LAMP assay for detecting YFV in NHP samples was developed and validated. Two sets of RT-LAMP primers targeting the YFV NS5 and E genes w

Ecological drivers of sustained enzootic yellow fever virus transmission in Brazil, 2017-2021.

Beginning December 2016, sylvatic yellow fever (YF) outbreaks spread into southeastern Brazil, and Minas Gerais state experienced two sylvatic YF waves (2017 and 2018). Following these massive YF waves, we screened 187 free-living non-human primate (NHPs) carcasses collected throughout the state between January 2019 and June 2021 for YF virus (YFV) using RTqPCR. One sample belonging to a Callithrix, collected in June 2020, was positive for YFV. The viral strain belonged to the same lineage associated with 2017-2018 outbreaks, showing the continued enzootic circulation of YFV in the state. Next
