BACKGROUND: Campylobacter jejuni is a zoonotic pathogen that infects the human gut through the food chain mainly by consumption of undercooked chicken meat, raw chicken cross-contaminated ready-to-eat food or by raw milk. In the last decades, C. jejuni has increasingly become the most common bacterial cause for food-born infections in high income countries, costing public health systems billions of euros each year. Currently, different whole genome sequencing techniques such as short-read bridge amplification and long-read single molecule real-time sequencing techniques are applied for in-dept

Multiple myeloma (MM) is the second most common haematological malignancy characterised by clonal proliferation of plasma cells within the bone marrow. Despite advances in treatment options, myeloma remains incurable. Relapsed MM poses significant challenges due to clonal evolution, drug resistance, patient comorbidities and therefore complexities of therapeutic decision-making. A critical question in managing relapsed MM is whether to switch drug classes or retreat with agents within the same class. The question is present with the recent addition of three new drug classes (XPO1 inhibitors, B

There are no approved treatments for Lassa fever, which is estimated to cause 100,000 to 300,000 infections and 5,000 deaths annually in West Africa1,2. Recently, it was shown that 4'-fluorouridine (also known as EIDD-2749), an orally available ribonucleoside analogue, protected guinea pigs from lethal challenge with the lineage IV prototype Josiah strain of Lassa virus when treatment was delayed beyond the onset of clinical signs of disease3. Here we assessed the therapeutic efficacy of 4'-fluorouridine in an African green monkey model of Lassa fever using the contemporary and apparently more

BACKGROUND: Lassa virus (LASV) is a persistent threat to public health in west Africa and beyond. LASV is endemic in west Africa and each year it is responsible for an estimated 2·7 million infections, 23 700 hospitalisations, and 5000 deaths. With over 32 reported cases of Lassa fever imported into non-endemic countries-one-third of which were fatal-the importance of enhanced detection and management of Lassa fever extends beyond west Africa.
METHODS: The prevalence, pathogenesis, and persistence (PREPARE) study was a prospective cohort study among patients admitted to two hospitals in a hyp

BACKGROUND: Lassa fever is an emerging zoonotic disease endemic to west Africa. Several vaccines aimed at preventing Lassa fever are currently under development, creating a need to assess how best to administer them once licensed for human use. We aimed to project the health-economic burden of Lassa fever from 2025 to 2037 across age and sex groups in subnational administrative divisions of west Africa with endemic Lassa mammarenavirus transmission and to estimate the cost-effectiveness of targeting Lassa vaccination to different risk groups.
METHODS: In this vaccine-impact modelling study, w

BACKGROUND: Marburg virus disease (MVD) poses an emerging threat to Nigeria, particularly following the 2022 outbreak in neighbouring Ghana. With Nigeria already managing Lassa fever and Mpox outbreaks, assessing healthcare workers' preparedness at viral haemorrhagic disease reference centres is crucial for effective outbreak response.
OBJECTIVES: This study aimed to assess healthcare workers' knowledge, attitudes and preparedness regarding MVD at Nigeria's primary viral haemorrhagic fever reference centre.
METHOD: A cross-sectional study was conducted at Irrua Specialist Teaching Hospital,

The continued emergence of pathogens, whether novel, re-emerging, or engineered, poses a persistent global biosecurity and public health challenge. Recent outbreaks, including COVID-19, Lassa fever, Marburg virus, mpox, and avian influenza, underscore the urgent need for robust systems that enable rapid surveillance, early diagnosis, and timely countermeasures before widespread human transmission occurs. In this article, we focus on early detection technologies and systematically evaluate current diagnostic and sensing modalities. We highlight sequencing and spectroscopy as two complementary a

Lassa virus (LASV) is the causative agent of Lassa fever, which causes thousands of deaths every year within the endemic regions of West Africa. Despite its discovery over half a century ago, our knowledge regarding basic steps in the LASV life cycle remains limited. In order to simplify studying basic principles of the LASV life cycle, we developed a novel transcription- and replication-competent virus-like particle (trVLP) system that can model the whole virus life cycle using only authentic LASV components but without the need for a maximum containment laboratory. As a proof-of-concept we u

Lassa virus (LASV), a zoonotic, bi-segmented arenavirus endemic to West Africa, causes seasonal epidemics with substantial morbidity and mortality. Increasing frequency of exported cases emphasizes the need for near real-time genomic surveillance to support outbreak response and clinical decision-making. We developed a suite of open-access resources on the Nextstrain and Nextclade platforms tailored to LASV. These include live phylogenetic and phylogeographic visualization, as well as Nextclade builds for rapid mutation detection and lineage assignment based on the L and S segments and the gly

Lassa fever is a zoonotic disease endemic to West Africa, primarily transmitted through contact with Mastomys natalensis, its rodent reservoir. Existing risk models often rely on regression-based approaches, which may overlook interactions with human settlement and environmental conditions. This study introduces a Self-Organizing Maps (SOM) classification approach to improve Lassa fever risk prediction. SOM included high-dimensional environmental data, outbreak zones based on rodent suitability, and human settlement density to improve classification accuracy. Google Earth Engine was used to ex

BACKGROUND: Lassa fever is one of the most important viral haemorrhagic fevers, yet post-discharge sequelae remain inadequately characterised. Previous studies have been limited by small sample sizes and unsystematic assessments. We aimed to describe post-discharge sequelae in Lassa fever survivors and explore the effect of disease severity on sequelae patterns.
METHODS: LASCOPE was a prospective study of patients with PCR-confirmed Lassa fever hospitalised at Federal Medical Centre Owo, Owo, Nigeria, between April 23, 2018, and Feb 17, 2023. All patients who provided informed consent were in

BACKGROUND: Lassa fever is a recurrent public health threat in Nigeria, particularly in endemic regions such as Kogi State. Its transmission is closely tied to environmental and seasonal dynamics, especially those influencing rodent populations. Understanding these climatic influences is essential for strengthening disease surveillance and implementing timely interventions. METHODS: A descriptive cross-sectional study was conducted using retrospective epidemiological and climatic data from 2019 to 2024. Confirmed Lassa fever case records and meteorological variables (temperature, rainfall, and

A high-risk infectious disease or a Category A pathogen, Lassa virus (LASV), requires strict containment, classified as biosafety level 4 (BSL-4) conditions, which restricts research on the virus due to the scarcity of BSL-4 facilities. Thus, replication-defective pseudotyped retroviral vectors have been widely used as safe materials for neutralizing activity assays of drugs and antibodies in BSL-2. Here, we established a novel retroviral vector system encoding LASV glycoprotein complex (GPC) that can exclusively replicate in cells expressing the Gag-Pol protein of murine leukemia virus (MLV)

UNLABELLED: The Lassa virus (LV) is a hemorrhagic virus that causes Lassa fever (LF). Despite its high mortality rate in Africa, no FDA-approved mRNA vaccine currently exists. Considering its epidemic potential, a new multi-epitope mRNA vaccine (LF-mVax) candidate is designed by immunoinformatics, targeting the GP, NP, and ZP through CD8+, CD4+, and B cell epitopes. LF-mVax was introduced with antigenic, non-allergic, and non-toxic epitopes that were derived from consensus sequences. The combined CD8 + and CD4 + epitopes provided 99.98% coverage of the global population, with robust immunity.

Lassa fever is a high-risk viral disease endemic to West Africa for which rapid and reliable diagnosis is essential for outbreak control. Here, we describe the development of a lateral flow-based rapid diagnostic test (RDT) targeting the Lassa virus nucleoprotein using newly generated monoclonal antibody pairs. The analytical performance of the assay was evaluated using human serum and plasma specimens as well as inactivated Lassa virus strains and compared with a commercially available antigen test. The final RDT (Pair 2; 1P-44-7/9B-28) demonstrated high analytical sensitivity and 100% specif

Post-acute sequelae are symptoms that persist or arise after the acute phase of an infection, but their frequency following outbreaks remains poorly understood. Recurrent Lassa fever outbreaks pose a significant public health threat in West Africa and may have long-term health effects. This study systematically reviewed the prevalence, incidence, duration, and characteristics of post-acute sequelae in survivors of Lassa virus infection. We searched PubMed and Web of Science up to November 17, 2025. Two reviewers screened and extracted data independently. We included six articles in the review.
