Computational Exploration of Potential Pharmacological Inhibitors Targeting the Envelope Protein of the Kyasanur Forest Disease Virus.

The limitations of the current vaccination strategy for the Kyasanur Forest Disease virus (KFDV) underscore the critical need for effective antiviral treatments, highlighting the crucial importance of exploring novel therapeutic approaches through in silico drug design. Kyasanur Forest Disease, caused by KFDV, is a tick-borne disease with a mortality of 3-5% and an annual incidence of 400 to 500 cases. In the early stage of infection, the envelope protein plays a crucial role by facilitating host-virus interactions. The objective of this research is to develop effective antivirals targeting th

Kyasanur Forest Disease: A Comprehensive Review.

Vector-borne microbial diseases are ubiquitous, and their management remains elusive. Such diseases with zoonotic potential result in public health challenges requiring additional control and preventive measures. Despite their cosmopolitan presence, vector-borne infections are neglected due to their endemicity in specified geographical regions. The Kyasanur forest disease (KFD) caused by the Kyasanur forest disease virus (KFDV) is among such diseases transmitted through ticks and localized to India. Despite its prevalence, high transmissibility, and potential to cause fatalities, KFDV has not

Oral Candidiasis in Acute Flaviviral Infection.

Kyasanur forest disease (KFD) is an endemic flaviviral hemorrhagic fever in south India. We report seven cases of oral candidiasis in patients from Karnataka, India, who presented in March and April 2024 with the first phase of KFD. Except for lymphopenia, no other risk factors for oral candidiasis were identified in these patients. All patients were managed with antifungals and did well at the 28-day follow-up. We report this case series to highlight the possibility of transient immunosuppression and the occurrence of opportunistic infections in flaviviral infections such as KFD.

Insights into the virome of Hyalomma marginatum in the Danube Delta: a major vector of Crimean-Congo hemorrhagic fever virus in Eastern Europe.

BACKGROUND: Ticks are significant vectors of pathogens, including viruses, bacteria, and protozoa. With approximately 900 tick species worldwide, many are expanding their geographical range due to changing socioeconomic and climate factors. The Danube Delta, one of Europe's largest wetlands, is an ecosystem that, despite its ecological importance, remains understudied concerning the risk of introducing new tick-borne viruses. This region serves as a critical habitat for migratory birds, which can carry ticks over long distances, potentially introducing exotic tick species and their pathogens i

Alkhumra Hemorrhagic Fever Virus (AHFV): A Concise Overview.

Alkhumra fever is a viral disease caused by the Alkhumra hemorrhagic fever virus (AHFV). It belongs to family Flaviviridae, genus Flavivirus. AHFV is primarily transmitted to humans through the bite of infected ticks, for example, Hyalomma. This disease was first identified in the Kingdom of Saudi Arabia (KSA) in 1995 and then reported in other countries of the Arabian Peninsula and the Middle East. The AHFV genome consists of a positive-sense, single-stranded RNA molecule of approximately 10.2 kilobases (kb) in length. The Open Reading Frame (ORF) encodes a polyprotein precursor that is proce

A simple improved method for extracting DNA from ethanol-preserved hard ticks and its applications.

Hard tick exoskeletons, composed primarily of chitin, pose a significant challenge for researchers attempting to extract genetic material. This study presents a simple modified, alternative method for extracting DNA from ethanol-preserved hard ticks. The extracted DNA was further used for PCR amplification of phylogenetic markers for population genetics studies. The study also improvises the DNA extraction methods from commercial kits. We have used four DNA extraction methods: Modified Simple Alkaline Lysis, and other commercial kit-based methods (Kit X, Kit Y & Kit Z). The modified method for

Prevalence of Crimean-Congo Haemorrhagic Fever virus and Ganjam virus among livestock & ticks in Wayanad, Kerala.

The Western Ghat region of Kerala has abundant flora and domestic and wild animal species that provide an ideal ecological niche for the ticks. Kyasanur Forest Disease virus (KFDV), a zoonotic tick-borne virus causing Haemorrhagic fever, commonly known as monkey fever is prevalent in the area. The tick infestations have been documented in the majority of household animals in Wayanad and risks are further amplified as the majority of the animals reared, graze in the adjoining forest areas and these animals are often housed in and around farmer's dwellings. Therefore, the farmers are at high ris

Development of a clinical scoring system to make a presumptive diagnosis of Kyasanur Forest Disease: a case-control study from South India.

INTRODUCTION: Kyasanur Forest Disease (KFD) is a viral haemorrhagic fever endemic in South India. Based on clinical presentation alone, it is challenging to distinguish KFD from other febrile illnesses in the region. The study aimed to develop a clinical scoring system for early presumptive diagnosis of KFD. PATIENTS AND METHODS: This retrospective case-control study included microbiologically diagnosed KFD patients (n=186) with other undifferentiated febrile illnesses as controls (n=203). The clinical and laboratory features between cases and controls were compared. A logistic regression ana

Cellular Immune Responses Against γ-Inactivated Antigen in the Recovered Cases of Kyasanur Forest Disease.

Kyasanur Forest Disease Virus (KFDV) is a tick-borne flavivirus that causes life-threatening hemorrhagic fever in humans with case fatality rates of 3-5%. Relatively little is known about the mechanism of its pathogenesis or host immune responses to KFDV infection. Here, we investigated KFDV-specific cellular immune responses in the recovered cases of Kyasanur Forest Disease (KFD). Peripheral blood mononuclear cells of the recovered KFD cases and healthy controls were exposed to γ-inactivated KFDV antigen ex vivo. The proliferation index was determined using an enzyme-linked immunosorbent assa

Studies on the sequential pathology of Kyasanur Forest Disease (KFD) in Mouse brain: KFD virus induces apoptosis of neurons in cerebrum and hippocampus.

The sequential pathology of Kyasanur forest disease (KFD) in mouse brain was assessed in this study. Kyasanur forest disease virus (KFDV) strain P9605 used in this study was confirmed by real-time reverse transcriptase-polymerase chain reaction targeting the NS5 gene. Mouse Lethal Dose 50 (MLD50) of the virus was determined by in-vivo mice inoculation test. One MLD50 of the KFDV was inoculated intra-cerebrally into 36 mice aged 2-3 weeks. Another group of 36 age-matched mice that served as control group were inoculated with plain media. Six mice each from infected and control groups were eutha

Predicting immune response targets in orthoflaviviruses through sequence homology and computational analysis.

CONTEXT: Flaviviruses cause severe encephalitic or hemorrhagic diseases in humans. Its members, Kyasanur forest disease virus (KFDV) and Alkhumra hemorrhagic fever virus (ALKV), cause hemorrhagic fever and are prevalent in India and Saudi Arabia, respectively, while the tick-borne encephalitis virus (TBEV) causes a dangerous encephalitic infection in Europe and Asia. However, little information is available about the targets of immune responses for these deadly viruses. Here, we predict potential antigenic peptide epitopes of viral envelope protein for inducing a cell-mediated and humoral immu

[Laboratory Diagnosis of Omsk Hemorrhagic Fever Virus and Kyasanur Forest Disease Virus].

The Omsk hemorrhagic fever virus and Kyasanur Forest disease virus, which belong to the genus Orthoflavivirus in the family Flaviviridae, are tick-borne viruses. These viruses, which were previously endemic to certain countries and regions, are now at increasing risk of spreading to new areas owing to global climate change and increased human mobility. Despite advances in real-time reverse transcription-polymerase chain reaction and serological assays, there are challenges in distinguishing these viruses from other tick-borne flavivirus infections. There is still a need for accurate and rapid

Land system governance shapes tick-related public and animal health risks.

Land cover and land use have established effects on hazard and exposure to vector-borne diseases. While our understanding of the proximate and distant causes and consequences of land use decisions has evolved, the focus on the proximate effects of landscape on disease ecology remains dominant. We argue that land use governance, viewed through a land system lens, affects tick-borne disease risk. Governance affects land use trajectories and potentially shapes landscapes favourable to ticks or increases contact with ticks by structuring human-land interactions. We illustrate the role of land use

Does the rise in cases of Kyasanur forest disease call for the implementation of One Health in India?

The viral hemorrhagic illness known as Kyasanur forest disease (KFD), also referred to as monkey fever, is transmitted by ticks. The etiological agent, which was formerly isolated from monkeys, is Kyasanur forest disease virus (KFDV), an RNA virus belonging to the family Flaviviridae. Since 1957, India has reported 400-500 cases annually, with a case fatality rate of 1-3%. Shiroma, Chikkamagalore, Uttara Kannada, Dakshina Kannada, and Udupi are the five regions in Karnataka, India where KFD is highly prevalent, with around 3263 notified cases reported between 2003 and 2012, of which 823 cases

The role of social vulnerability in improving interventions for neglected zoonotic diseases: The example of Kyasanur Forest Disease in India.

Forest-based communities manage many risks to health and socio-economic welfare including the increasing threat of emerging zoonoses that are expected to disproportionately affect poor and marginalised groups, and further impair their precarious livelihoods, particularly in Low-and-Middle Income (LMIC) settings. Yet, there is a relative dearth of empirical research on the vulnerability and adaptation pathways of poor and marginalised groups facing emerging zoonoses. Drawing on a survey of 229 households and a series of key-informant interviews in the Western Ghats, we examine the factors affec
