One-step detection of Chlamydia psittaci using recombinase polymerase amplification and CRISPR/Cas12b.

The development of rapid, sensitive, and user-friendly diagnostic methods for Chlamydia psittaci (C. psittaci) is critical for early diagnosis and effective control of infections. Current techniques, such as qPCR and next-generation sequencing, face limitations in accessibility and practicality for point-of-care testing (POCT). In this study, we present the WPTTS platform (one-step recombinase polymerase amplification (RPA)-CRISPR/Cas12b assay using weak protospacer adjacent motif (PAM) recognition with two-temperature shifting), which leverages the temperature-dependent PAM stringency of Alic

Lawsonia intracellularis infection induces changes in microbial community function and composition associated with reduced pig growth and feed efficiency.

BACKGROUND: Lawsonia intracellularis and its resulting disease remains a troubling pathogen for pork producers worldwide. In the current experiment, we aimed to characterize the microbiome of pigs challenged with L. intracellularis through peak disease impact to better understand microbial community function and how microbial changes may contribute to disease and resulting decreased growth. Twenty-four L. intracellularis negative barrows were assigned to either L. intracellularis negative (NC) or L. intracellularis challenged (PC) treatment groups (n = 12 pigs/treatment). On days post-inoculat

Polymicrobial PID Presenting as Primary Peritonitis in a Young Immunocompetent Patient-Case Report and Disease Perspectives.

Background and Clinical Significance: Pelvic inflammatory disease represents a multifaceted sexually transmitted disease affecting women of reproductive age, beginning in adolescence. Clinical presentation ranges from asymptomatic patients to acute abdominal pain in the setting of tubo-ovarian abscesses; however, presentation as primary peritonitis with seemingly intact fallopian tubes is exceptional. Primary peritonitis in the absence of other comorbid conditions (e.g., liver cirrhosis and nephrotic syndrome) in healthy, immunocompetent women is rare and typically occurs without an identifiab

Immunohistochemical Characterization of the Immune Response in Chronic Endometritis Caused by Chlamydia trachomatis.

In 2020, 128.5 million new chlamydia infections were reported worldwide in adults aged 15-49 years. Notably, the prevalence of Chlamydia trachomatis infection in pregnant women varies between 2% and 35%, correlating with increased risks of low birth weight, preterm birth, and neonatal death. C. trachomatis is a leading preventable cause of miscarriage. Recurrent first-trimester pregnancy loss can be induced by asymptomatic chlamydia infection through the immune response. In this study, we performed immunohistochemical characterization of the immune response in endometrial tissue biopsies from

Family psittacosis cluster diagnosed by metagenomic next-generation sequencing in Hangzhou City, Eastern China: A case series.

RATIONALE: Psittacosis, a human infection caused by Chlamydia psittaci (C psittaci), is often underdiagnosed due to its nonspecific presentation and the limitations of conventional diagnostic methods. This diagnostic challenge can lead to missed outbreaks and delays in appropriate treatment. This report aims to highlight the critical role of metagenomic next-generation sequencing (mNGS) in rapidly identifying C psittaci and facilitating the investigation of a family cluster, thereby providing a clearer rationale for its application in similar scenarios. PATIENT CONCERNS: In this study, we rep

Aconite Alkaloids From Gymnaconitum gymnandrum.

Two previously uncharacterized C19-diterpenoid alkaloids (1 and 2) and seven known ones (3-9) were isolated from Gymnaconitum gymnandrum. Their structures were elucidated based on spectroscopic data, including one- and two-dimensional nuclear magnetic resonance, infrared, high-resolution electrospray ionization mass spectrometry, and single-crystal X-ray diffraction analysis. These compounds were tested for their insecticidal activity and anti-chlamydial activity. The results show that compound 1 has the most insecticidal activity, with a median lethal concentration value of 0.73 ± 0.08 mg/mL;

National and international guidelines for preconception care and comparison with Australian guidelines: a systematic review.

BACKGROUND: Guidelines for preconception care are resources that are systematically arranged based on evidence and local contexts to enhance the delivery of high-quality and consistent healthcare. The Royal Australian College of General Practitioners Guidelines (RACGP) for preventive activities in general practice (the Red Book) contains specific components of preconception care items, yet this guideline primarily focuses on general preventive healthcare and may not include a comprehensive list of preconception items based on more recent evidence. This review aimed to compare the recent RACGP

Identification of Chlamydia pneumoniae and NLRP3 inflammasome activation in Alzheimer's disease retina.

Chlamydia pneumoniae is an intracellular bacterium implicated in Alzheimer's disease (AD), but its role in retinal pathology and disease progression is unclear. Here we identify Chlamydia pneumoniae inclusions in the retina, showing higher burden in AD retina and brain, increasing with APOEε4, disease stage, and cognitive deficit. Retinal and cortical proteomics reveal bacterial-infection and related NLRP3-inflammasome pathways. Retinal NLRP3 is elevated in mild cognitive impairment and activated in AD dementia, evidenced by increased caspase-1, cleaved interleukin-1β, and cleaved N-terminal g

Chlamydia psittaci inclusion membrane protein CPSIT_0844 elicits inflammatory IL-6 and IL-8 production in human monocytes via TLR2/TLR4 signaling pathways.

The respiratory disease caused by Chlamydia psittaci (C. psittaci) infection is often characterized by significant inflammation. In the present study, we demonstrated that the C. psittaci inclusion membrane protein CPSIT_0844 induced THP-1 cells to express IL-6 and IL-8. However, silencing of the Toll-like receptor 2 (TLR2) and Toll-like receptor 4 (TLR4) genes by using small interfering RNA (siRNA) and transfection with the dominant negative plasmid encoding MyD88 (pDeNy-hMyD88) were found to reduce the expression of IL-6 and IL-8 in THP-1 cells stimulated with CPSIT_0844. Furthermore, CPSIT_

Metagenomic Profiling of Fecal and Cecal Microbiota and Their Antimicrobial Resistance Genes in Indigenous Backyard Poultry.

Indigenous backyard poultry is the predominant type of poultry in developing countries. Rural smallholder farmers in these regions usually adopt the free-range (backyard) production system, which exposes the poultry to diverse environments and a broad spectrum of microorganisms that influence their diet and gut microbiota. In this cross-sectional purposive study, we evaluated the microbial community profiles of indigenous backyard poultry and their antimicrobial resistance genes (ARGs) using both cecal samples, which provide a more accurate representation of the core gut microbiota, and fecal

Anti-Chlamydia trachomatis Host Defence Arsenal Within the Cervicovaginal Environment.

Chlamydia trachomatis has a significant impact on public health, especially among adolescents and young women; it primarily affects urogenital epithelial cells, leading to cervicitis and urethritis, with >90% of cases showing no symptoms. Consequently, chlamydial infections are commonly misdiagnosed, and, if untreated, they may result in severe reproductive sequelae including infertility. A better understanding of C. trachomatis cell biology and bacterial-host cell interactions may be helpful to identify strategies able to counter its transmission among the population, as well as its dissemina

Effect of Hantavirus Infection on the Rodent Lung Microbiome: Specific Regulatory Roles of Host Species and Virus Types.

The lung-targeting characteristic of Hantavirus infection and the unclear mechanism underlying its interaction with the lung microbiome hampers the development of effective prevention and control strategies. In this study, lung tissues from Apodemus agrarius and Rattus norvegicus were collected at Hantavirus surveillance sites in Hunan Province. Metagenomic sequencing was subsequently applied to compare microbiome diversity, community structure, and function between infected and uninfected groups. Then the linear discriminant analysis effect size (LEfSe) was employed to identify key biomarkers
