BACKGROUND: Cisplatin is a widely used chemotherapeutic agent whose clinical utility is limited by dose-dependent hepatotoxicity, primarily driven by oxidative stress and inflammation. Agmatine, an endogenous polyamine, has demonstrated antioxidant and anti-inflammatory properties in various models; however, its hepatoprotective effects in cisplatin-induced liver injury remain underexplored.
METHODS: This experimental study investigated the protective effects of agmatine against cisplatin-induced hepatotoxicity in 28 male Sprague Dawley rats. The animals were randomized into four groups: Sham

Previous studies have indicated that Valsartan (Val) modulates peroxisome proliferator-activated receptor α (PPARα) expression in the liver; however, its precise roles in liver injury, both pathological and physiological, remain unclear, necessitating further investigation. This study hypothesises that FGF21 mediates Val's hepatoprotective effects in acetaminophen (APAP)-induced acute liver injury. The results demonstrate that fibroblast growth factor 21 (FGF21) is a key mediator of Val-induced hepatoprotection, acting through the activation of the peroxisome proliferator-activated receptor-γ

The current research work examined the protective effects of some green plant extracts compared with N-acetylcysteine against liver toxicity caused by CCL4 in hepatic Alpha Mouse Liver 12 (AML-12) cells. Using standard biochemical assays, oxidative stress indicators, inflammatory cytokines, and redox status were measured. Plant extracts boosted the survival rate to reach 86.05% (moringa), 83.64% (arugula), 79.45% (broccoli), 78.19% (mint), and 77.21% (celery). Moringa demonstrated the strongest hepatoprotection, with enzyme levels such as alanine aminotransferase (ALT), aspartate aminotransfer

Paclitaxel (PAX) is an effective chemotherapeutic agent for treating various cancers but is associated with side effects such as hepatotoxicity, neurotoxicity, and myelosuppression. To mitigate these toxicities, adjunct therapies like salicylic acid (SA), a plant-derived compound with anti-inflammatory, antioxidant, and cytoprotective properties, have been explored. This study investigates SA's potential to counteract PAX's cytotoxic effects, focusing on hepatic tissue. Thirty-five BALB/cJ mice were treated with PAX and SA, either individually or in combination (low and high doses), for 14 day

Acute liver failure (ALF) is characterized by extensive hepatocyte necrosis and limited treatment options. Adipose-derived stem cells (ADSCs) show potential for liver regeneration, yet the mechanisms regulating their hepatic differentiation remain unclear. Human ADSCs were infected with lentiviruses to overexpress or silence miR-204-5p, Murine double minute 2 (MDM2), E2F transcription factor 8 (E2F8), or bone morphogenetic protein 4 (BMP4). qPCR, immunofluorescence, and western blot detected mRNA and protein levels of hepatic markers-including hepatocyte nuclear factor 4 alpha (HNF4α), albumin

Acetaminophen (i.e. paracetamol; APAP) overdose is the most common cause of acute liver injury in the United States, yet outcomes range from recovery to acute liver failure (ALF), defined by coagulopathy and hepatic encephalopathy (HE). Clinical decision-making is difficult because routine indices of hepatocellular injury (e.g. ALT/AST) do not reliably predict which patients will recover versus progress to ALF. This underscores the need to define mechanisms that drive progression to ALF and to discover biomarkers that better predict clinical trajectory. A major barrier is the lack of an experi

Chronic ethanol exposure activates inflammatory signaling pathways and inflicts hepatocellular damage, leading to alcohol-associated liver diseases (ALDs). ALD is one of the major causes of global burden, yet there are no FDA-approved treatment options available. This study evaluates the hepatoprotective effects of short-chain fatty acids (SCFAs), mainly sodium acetate (NaA) and sodium butyrate (NaB), against ethanol-induced inflammation and oxidative stress in both in vitro (Buffalo Rat Liver-3A [BRL3A]) and in vivo (male Wistar rats) models. The treatment of NaA and NaB and their combination

Although doxorubicin (DOX) is a first-line antineoplastic drug, its clinical use is limited due to its widespread side effects. Hepatic and renal metabolic clearance causes DOX accumulation in these organ systems, ultimately leading to oxidative-inflammatory damage. This study investigated the potential impact of syringic acid (SA), a natural antioxidant phenolic compound, against in vivo DOX-evoked hepatorenal damage. SA was orally administered to rats at 100 mg/kg body weight for 10 consecutive days. On the 8th day of the study, DOX was administered at 20 mg/kg by single-dose intraperitoneal

Lead acetate (PbAc) is an environmental toxicant known to induce liver injury through oxidative stress, inflammation, and apoptosis. This study aimed to investigate the potential hepatoprotective effects of β-caryophyllene (BCP), a natural sesquiterpene with antioxidant and anti-inflammatory properties, against PbAc-induced hepatotoxicity and to explore the underlying molecular mechanisms. Sixty male Sprague-Dawley rats were randomly assigned to five groups: control, BCP200, PbAc, PbAc + BCP100, and PbAc + BCP200. PbAc was used to induce hepatotoxicity, while BCP was administered at doses of 1

BACKGROUND: T-2 toxin, a highly toxic trichothecene mycotoxin contaminating cereal grains, poses serious health threats. However, its specific effects on hepatic drug-metabolizing enzymes (DMEs) remain poorly characterized. This study aimed to investigate the impact of T-2 toxin on key cytochrome P450 (CYP) and carboxylesterase (CES) enzymes in mouse liver.
METHODS: Male C57BL/6 mice were randomly divided into control and T-2 toxin-treated groups, receiving either normal saline or T-2 toxin (0.2 mg/kg) by oral gavage daily for four weeks. Histopathological and ultrastructural alterations were

Poly rC binding proteins (PCBPs) 1 and 2 bind iron and ssDNA/RNA, acting as iron chaperones for enzyme metalation, ferritin storage, and iron toxicity prevention. Previously, we demonstrated that liver-specific PCBP1 deletion in mice increased unchaperoned iron, causing hepatic oxidative damage and steatosis. Little is known about the functions of PCBP2 in vivo. To define the roles of PCBP1 and PCBP2 in murine hepatic iron metabolism, we generated conditional (Alb-Cre) and vector-mediated (AAV8-TBG-iCre) deletion as models of chronic and acute PCBP deficiency, respectively. Conditional deletio

The environmental chemical rotenone, which is employed as a natural insecticide, is hazardous to many organs. Thus, this study aimed to elucidate the protective efficacy of an oral administration of pramipexole (PPX) at a dose of 0.5 mg/kg/d against rotenone-induced hepatocellular damage for 60 days. Rotenone administration caused significant hepatic dysfunction, as evidenced by elevated serum AST, ALT, GGT, and bilirubin levels, along with reduced total protein and albumin, indicating impaired liver function. PPX co-administration markedly attenuated these changes, restoring liver enzyme acti

Drug-induced hepatic insult is the most frequent antecedent of liver dysfunction. The folic acid antagonist, Methotrexate (MTRX), has tremendous applications in the treatment of various tumors, autoimmune disorders, and inflammatory diseases, but hepatotoxicity limits its clinical use. This study aimed to investigate the role of AGM against MTRX-induced hepatotoxicity and its possible underlying mechanistic pathways. Mice were pre-treated with AGM (14 mg/kg, orally) and administered a single injection of MTRX (20 mg/kg, i.p.) on Day 6. MTRX-induced hepatic injury was evidenced by marked increa

Antioxidants play a crucial role in the treatment of liver pathologies, given that oxidative stress is involved in the pathogenesis of many hepatic diseases. This study aimed to investigate the pharmacodynamic and pharmacokinetic properties of the antioxidant 2,6-diisobornyl-4-methylphenol (IBP) in a rat model of carbon tetrachloride (CCl4)-induced liver injury. The introduction of IBP resulted in a 1.4- to 2-fold decrease in serum hepatic enzyme levels, a 25% and 18% rise in glutathione peroxidase and superoxide dismutase concentrations, respectively, and a 14% reduction in hepatic catalase a

Liver fibrosis arises from a sustained imbalance between oxidative stress, cell death, and matrix deposition. Using the carbon tetrachloride (CCl4) mouse model, we show that the dietary flavonoid, fisetin orchestrates a broad hepatoprotective program that blunts this cascade. Male C57BL/6 mice received CCl4 for six weeks, without/with daily fisetin at 50 or 100 mg/kg. Co-treatment with fisetin preserved body weight and lowered the liver index, and markedly attenuated biochemical injury, reducing serum ALT, AST, ALP, GGT, and TBIL while restoring total protein and albumin levels. Fibrogenic bur

This study investigated the protective effects of dietary supplementation with 2% black cumin (Nigella sativa) oil (BCO) against malathion-induced toxicity in Nile tilapia (Oreochromis niloticus). Over a 72-day feeding trial, fish were divided into four groups: control, 2% BCO, malathion (LC₅₀/20), and malathion + 2% BCO. The results demonstrated that sublethal malathion exposure significantly impaired growth performance including final weight, weight gain, and specific growth rate and worsening the feed conversion ratio. Hematological analysis revealed a severe anemic response, marked by sign

TINOSPORA: sagittata is a widely used medicinal herb whose safety has raised increasing concerns in recent years. Fibleucin (FBL), a furanoditerpenoid isolated from T sagittata, has not been systematically evaluated for its safety and toxicological mechanisms despite its presence as a major bioactive constituent in this medicinal plant. To address this toxicological knowledge gap, we comprehensively investigated the hepatotoxicity and/or metabolic activation of FBL in vivo (mice) and in vitro (mouse and human liver microsomal incubations as well as primary mouse hepatocytes).
