Moscatilin protects concanavalin A-induced autoimmune liver disease in mice.

BACKGROUND: Moscatilin has been shown to effectively suppress inflammation in macrophages in mice treated with concanavalin A (ConA). However, further investigation is needed to fully elucidate the specific underlying mechanisms. METHODS: A murine model of autoimmune liver disease was induced by administering ConA. Inflammatory markers were quantified using enzyme-linked immunosorbent assay (ELISA), while M1 and M2 macrophage biomarkers were analyzed by western blotting. Apoptosis was assessed via TdT-mediated dUTP Nick-End Labeling (TUNEL) staining, and immunohistochemical staining was used

Novel near-infrared probe for monitoring lipid peroxidation-mediated viscosity change in ferroptotic hepatocytes.

BACKGROUND/AIMS: Ferroptosis, recently emerged as a new cell death modality characterized by iron-dependent peroxidation of lipids, has been explored in various diseases. However, detection of ferroptosis, particularly in chronic liver disease models, is hampered by the lack of universal ferroptosis markers and limited number of fluorescence sensors for in vivo ferroptosis. METHODS: In this study, we developed TTM-4 as a highly sensitive near-infrared (NIR) fluorescent probe to detect ferroptosis. RESULTS: TTM-4 exhibited turn-on fluorescence upon viscosity change, enabling visualization of

Comparative Renal Safety of Besifovir Dipivoxil Maleate and Tenofovir Disoproxil Fumarate in Chronic Hepatitis B Patients: Insights From a Nationwide Cohort Study.

BACKGROUND: Besifovir dipivoxil maleate (BSV) is a novel antiviral agent approved in Korea for chronic hepatitis B (CHB) treatment. While previous comparative studies between BSV and tenofovir disoproxil fumarate (TDF) have suggested lower renal toxicity in BSV, these findings were limited by small sample sizes. This study aimed to comprehensively assess the incidence of chronic kidney disease (CKD) in CHB patients treated with BSV versus TDF using a nationwide cohort. METHODS: In this retrospective cohort study, we analyzed treatment-naïve CHB patients who initiated BSV or TDF therapy betwee

Immune-related adverse events in hepatocellular carcinoma: organ-specific patterns and management approaches.

Hepatocellular carcinoma (HCC) is a major cause of cancer-related mortality worldwide. The recent introduction of immune checkpoint inhibitors (ICIs) has transformed the therapeutic landscape for advanced HCC. Combination regimens such as atezolizumab plus bevacizumab, durvalumab plus tremelimumab, and nivolumab plus ipilimumab have demonstrated significant survival improvements over conventional tyrosine kinase inhibitors and have become the new standard of care. However, ICIs can trigger immune-related adverse events (irAEs) through overactivation of the immune system, affecting multiple org

Transarterial Radioembolization (TARE) Complicated by a Mycotic Aneurysm and Bleeding: A Case Presentation and Literature Review.

Hepatocellular carcinoma (HCC) is a primary cancer of the liver, typically developing in the setting of chronic liver disease, especially in patients with cirrhosis related to chronic hepatitis B or C virus infections, nonalcoholic steatohepatitis (NASH), or chronic alcohol abuse. HCC is the third leading cause of cancer-related deaths in the world and one of the fastest-growing causes of cancer death in the United States. The reason for this poor prognosis of most HCC patients is the advanced stage at diagnosis. HCC patients are frequently asymptomatic at early stages, and screening even for

A man in his 60s with immunodeficiency and hepatitis.

BACKGROUND: Major causes of elevated liver enzymes are toxic, infectious, neoplastic and autoimmune disease. In immunocompromised patients, the spectrum of infectious pathogens causing hepatitis is broader and the immune response is blunted, making diagnosis more challenging. CASE PRESENTATION: A man in his sixties had received CAR-T therapy for multiple myeloma. INTERPRETATION: The increasing number of immunocompromised patients means that more patients are at risk of developing chronic hepatitis E. Serologic tests may be unreliable for this patient population, and nucleic acid testing for

Immunomodulatory and anti-fibrotic effects of Toxocara canis infection in a murine model of thioacetamide-induced chronic hepatic fibrosis.

Chronic hepatic fibrosis is a serious result of chronic extracellular matrix (ECM) accumulation due to repeated liver injury from varied causes such as viruses, alcohol and chemicals. Toxocaral hepatitis, however, is brought about by inflammation resulting from migrating larvae of Toxocara canis (T. canis). Researchers have not yet explored the interaction between thioacetamide (TAA) induced and hepatic toxocariasis. In this experiment, the effect of these larvae as a potential safe vector that carries glycoconjugate antigens for treatment of liver fibrosis investigated in a model of chronic T

Antiviral therapy for chronic hepatitis delta: new insights from clinical trials and real-life studies.

Chronic hepatitis D (CHD) is the most severe form of viral hepatitis, carrying a greater risk of developing cirrhosis and its complications. For decades, pegylated interferon alpha (PegIFN-α) has represented the only therapeutic option, with limited virological response rates and poor tolerability. In 2020, the European Medicines Agency approved bulevirtide (BLV) at 2 mg/day, an entry inhibitor of hepatitis B virus (HBV)/hepatitis delta virus (HDV), which proved to be safe and effective as a monotherapy for up to 144 weeks in clinical trials and real-life studies, including patients with cirrh

Long-term inhibition of Hepatitis B virus gene expression by a primary microrna expressing ancestral adeno-associated viral vector.

Current treatments for chronic infection with the hepatitis B virus (HBV) rarely cure carriers from the disease. Previously reported use of serotype 8 adeno-associated viral (AAV8) vectors to deliver expression cassettes encoding anti-HBV artificial primary microRNAs (apri-miRs) has shown promise in preclinical studies. A recently designed synthetic ancestral AAV (Anc80L65) with high liver transduction efficiency is a promising new addition to the anti-HBV vector toolbox. This study engineered Anc80L65 to express HBx-targeting apri-miRs. Single dose administration of the vectors to cultured ce

Hepatocytes and Hepatic Stellate Cells Carry Different Levels of DNA Damage due to Their Sensitivity to Oxidative Stress in Chronic Hepatitis B.

Hepatocellular carcinoma (HCC) and liver cirrhosis (LC) occur in spite of current antiviral therapies in patients with chronic hepatitis (CHB). It is not yet known why HCC and LC are related to hepatocytes and hepatic stellate cells (HSCs), respectively, in the same inflammation circumstances. The expression of the phosphorylated form of histone H2AX (γ-H2AX), a biomarker of DNA damage, was detected in hepatocytes and interstitial cells within the liver tissues of 69 patients with CHB using immunohistochemical assay and immunofluorescence colocalisation technique. Hydrogen peroxide (H2O2) was

Vaccine Development T-cell (MHC-I) Epitopes Identification Against the Indian HCV Genotype: An Approach Based on Immunoinformatic.

Hepatitis C virus (HCV) infects approximately 58 million individuals worldwide, often progressing to chronic liver disease, cirrhosis, and hepatocellular carcinoma. The viral envelope glycoproteins E1 and E2 are critical for HCV entry and serve as primary targets for neutralizing antibodies. Recent advancements in cryo-electron tomography have provided high-resolution structures (3.5 Å) of the E1E2 heterodimer, revealing interactions between the E1 and E2 ectodomains, as well as neutralizing antibody complexes (e.g., AR4A, AT1209, IGH505). This structural information facilitates the design of

Assessing the virological response to direct-acting antiviral therapies in the HBV cure programme.

Hepatitis B virus (HBV) is a global health problem with over 250 million people affected worldwide. Nucleos(t)ide analogues remain the standard of care and suppress production of progeny virions; however, they have limited effect on the viral transcriptome and long-term treatment is associated with off-target toxicities. Promising results are emerging from clinical trials and several drug classes have been evaluated, including capsid assembly modulators and RNA interfering agents. Whilst peripheral biomarkers are used to monitor responses and define treatment endpoints, they fail to reflect th

[Fibrotic diseases in the gastrointestinal tract : Liver fibrosis and more].

Chronic liver damage, such as metabolic dysfunction-associated steatotic liver disease (MASLD), viral hepatitis B or C, cholestatic hepatitis (PBC, PSC), toxic damage (alcohol) or genetic alterations (hemochromatosis, Wilson's disease, etc.) usually cause a chronic inflammatory response in liver cells or bile duct epithelial cells. In the long term this chronic inflammatory response can lead to scarring of the liver, a condition known as fibrosis. The development of liver fibrosis is largely independent of the causative agent, although the pattern of initial fibrosis (periportal, pericentral o

Alcoholic cirrhosis-associated immune dysfunction: What does it imply for us?

Alcoholic cirrhosis is a leading cause of chronic advanced liver disease. With the gradual eradication of viral hepatitis and the rising levels of alcohol consumption, the incidence of alcoholic cirrhosis is expected to increase steadily. Alcohol is primarily metabolized in the gastrointestinal tract, producing toxic metabolites that enter the portal vein circulation and are subsequently transported to the liver. Excessive alcohol intake activates the microsomal ethanol oxidation system and disrupts the intestinal microbiota-driven microenvironment dictated by intestinal microbiota, and increa

Geographic diversity of human liver cancers mirrors global social inequalities.

Liver cancers show high interindividual and intratumor heterogeneity. Among them, hepatocellular carcinoma (HCCs) represents approximately 90% of liver cancers, followed by intrahepatic cholangiocarcinoma (iCCA; ~10 to 15%), childhood hepatoblastoma, angiosarcoma and hemangioendothelioma (< 1%). More than 80% of HCCs arise in a backdrop of chronic inflammatory liver diseases of diverse etiologies. These underlying liver diseases are major determinants of geographic diversity of HCCs. Across the world, substantial differences in the prevalence of chronic viral hepatitides, alcohol misuse, Metab

Antioxidant and hepatoprotective effect of Jatropha dioica against the valproic acid-induced damage in an in vivo model.

BACKGROUND: Understanding liver diseases is important worldwide due to their prevalence. Apart from liver disease arising from hepatitis C viral infection, most chronic liver diseases currently have no cure. Several therapeutic alternatives, including some natural products, have been proposed to treat liver diseases. The natural product Jatropha dioica has been reported to possess antioxidant activity and, by extension, could have hepatoprotective activity. Accordingly, our aim was to test the hypothesis that an extract of J. dioica is protective against liver damage induced by valproic acid (
