Characteristics and Key Genetic Pathway Analysis of Cr(VI)-Resistant Bacillus subtilis Isolated from Contaminated Soil in Response to Cr(VI).

With increasing industrialization, hexavalent chromium (Cr(VI)) is used in various metal smelting and other industries, which, in turn, causes hexavalent chromium pollution. This study aimed to investigate the characteristics of isolated Bacillus subtilis (B. subtilis) from high-Cr(VI) soils and to evaluate its safety. Genomic and transcriptomic analyses were performed to explore its Cr(VI) response mechanisms, and a mouse model (24 mice) was established to evaluate the safety of the bacterium at different concentrations. Key genetic findings showed that Cr(VI) exposure significantly up-regula

Perioperative management of pediatric patients with inborn errors of metabolism during liver transplantation.

Inborn errors of metabolism (IEM) are rare disorders, most are liver-based with liver transplantation (LT) emerging as an effective cure in the pediatric population. LT has been shown to offer a cure or deter disease progression and provide symptomatic improvement in patients with IEM. Each metabolic disorder is unique, with the missing enzyme or transporter protein causing substrate deficiency or toxic byproduct production. Knowledge about the distribution of deficient enzymes, the percentage of enzymes replaced by LT, and the extent of extrahepatic involvement helps anticipate and manage com

Towards EU regulatory hazard assessment of metabolic endocrine disrupters: Integrating new biomarkers into OECD test guidelines.

Exposure to 'metabolic disrupting chemicals' (MDCs) are increasingly implicated in obesity, diabetes and/or fatty liver disease; indeed, these metabolic changes may play a role in the global metabolic disorders' epidemic. To better assess and manage the health risks of MDCs, improved hazard identification is needed. This review describes how current in vivo OECD Test Guidelines (TGs) can better capture MDC effects. The biological and clinical evidence to support the inclusion of promising human relevant biomarkers, blood parameters, endpoints and relevant tissues for MDCs for potential inclusi

Timing matters: circadian rhythm disruption in alcohol-associated peripheral organ pathophysiology.

Circadian rhythms influence the pathophysiological effects of alcohol. This review examines how alcohol disrupts circadian regulation in peripheral organs, particularly the liver, intestine, cardiovascular system, and skeletal muscle, and how circadian disruption exacerbates metabolic dysfunction and organ injury. Evidence from preclinical and human studies indicates that both genetic and environmental circadian disruption increases alcohol-induced gut permeability, systemic inflammation, and liver disease. This review also presents how other lifestyle and environmental factors such as diet, s

Proof of concept for an age- and inflammation-adjusted model for the establishment of pediatric serum copper reference intervals.

BACKGROUND & AIMS: Copper is a trace element essential for enzymatic reactions, but excessive accumulation can cause toxicity. Accurate interpretation of serum copper concentrations is crucial for diagnosing conditions such as Wilson's disease, liver dysfunction, and nutritional deficiencies. Current reference intervals often ignore the effect of inflammation and are typically established using discrete age groups rather than modelling age as a continuous variable. This study aimed to establish continuous, age-adjusted reference intervals for serum copper and to develop a method for correcting

Beta-caryophyllene restores liver-brain axis integrity in thioacetamide-induced hepatic encephalopathy: Behavioral and molecular insights.

Hepatic encephalopathy (HE) is a severe neuropsychiatric complication of liver dysfunction, driven by hyperammonemia, oxidative stress, neuroinflammation, apoptosis, and endoplasmic reticulum (ER) stress, which disrupt the hepato-encephalic axis and impair cognition and motor functions. Despite its clinical burden, effective therapies that target this multi-organ pathology remain limited. β-Caryophyllene (BCP), an antioxidant and anti-inflammatory dietary sesquiterpene, has not been evaluated for its ability to modulate liver-brain crosstalk in HE. This study investigated the hepatoprotective

Glyphosate exposure and metabolic syndrome: A scoping review of epidemiological and mechanistic evidence.

BACKGROUND: Glyphosate, the active ingredient in many widely used herbicides, is a pervasive environmental contaminant due to extensive agricultural and residential use. Initially deemed safe, emerging evidence suggests chronic glyphosate exposure may impair metabolic health. This scoping review maps the evidence linking glyphosate exposure with metabolic syndrome (MetS), type 2 diabetes (T2DM), and related outcomes, integrating findings from human, animal, and cellular studies. METHODS: We reviewed studies published from 2004 in PubMed and Google Scholar that examined glyphosate exposure in

Detoxification of T-2 toxin in human chondrocytes due to its hydroxylation by carboxylesterases and arylacetamide deacetylase in the liver.

T-2 toxin, a trichothecene mycotoxin produced by Fusarium species, is a global cereal contaminant and an important environmental risk factor for cartilage damage and Kashin-Beck disease (KBD). Although hydrolysis is a major metabolic pathway of T-2 toxin, the human enzymes responsible and their toxicological relevance remain poorly understood. In this study, human liver microsomes, recombinant hydrolases, and selective inhibitors were used to identify the key enzymes mediating T-2 hydrolysis, and LC-MS/MS was applied to quantify T-2 and its major metabolites. Human chondrocytes were further us

Caffeine clearance in severe intoxication: a comparative case series of hemodialysis, hemoadsorption, and continuous kidney replacement therapy.

INTRODUCTION: Extracorporeal treatments are effective in managing severe caffeine intoxication. We evaluated caffeine clearance in three patients treated with hemodialysis, hemoadsorption, or continuous kidney replacement therapy. CASE REPORTS: A 49-year-old woman with alcoholic liver cirrhosis underwent hemodialysis for serum caffeine concentration of 141 mg/L; clearance was 68.6 mL/min, reducing concentration below toxicity at 63.2 h post-admission. A 19-year-old woman with a polypharmacy overdose received hemoadsorption for an initial concentration of 194 mg/L; clearance was 50.1 mL/min, w

Integrated Network Toxicology and Transcriptomics Reveal Molecular Mechanisms of Cadmium-Exposed Liver Injury in Swine.

Cadmium (Cd) is an environmental toxicant that poses significant risks to food safety and public health through its bioaccumulation in the food chain. The liver is a primary target for chronic Cd toxicity, yet the system-level mechanisms, particularly in physiologically relevant swine models, remain incompletely understood. This study employed an integrated multi-omics approach to elucidate the mechanisms of Cd-exposed hepatotoxicity in weaned piglets. We combined histopathological examination, transmission electron microscopy, and transcriptome sequencing. Our results revealed severe hepatic

Non-invasive identification of brain signatures of acute liver injury.

Background: In many disorders, metabolic and inflammatory derangements that originate in peripheral organs have a deleterious impact on the brain. Brain functional impairment, defined as hepatic encephalopathy, is one of the main diagnostic criteria for acute liver failure (ALF), a severe complication of acute liver injury (ALI). While brain inflammation (neuroinflammation) and metabolic alterations significantly contribute to hepatic encephalopathy, their non-invasive evaluation remains challenging. Methods: To address this limitation, we utilized dual radiotracer [18F]-fluoro-2-deoxy-2-D-glu

A Computational Framework to Evaluate Interactions of BPA and Its Analogs with Human Liver X Receptor-Beta for Health Risk Assessment.

Bisphenols are widely used in industrial applications to produce plastics and other consumer products. Among them, bisphenol A (BPA) is the most extensively studied due to its well-documented endocrine-disrupting effects and its association with various health conditions, including metabolic disorders and liver disease. Due to its known toxicity, BPA use has been restricted in many countries, leading to the emergence of several structural analogs. Recent studies have shown that BPA can interfere with normal liver metabolism by interacting with Liver X Receptor-beta (LXRβ). Although some BPA an

Long-term outcomes of atezolizumab-bevacizumab in unresectable hepatocellular carcinoma: A real-world study.

BACKGROUND AND AIMS: Unlike other immune-based combinations for hepatocellular carcinoma (HCC), long-term data for atezolizumab-bevacizumab (AB) are lacking, as the IMbrave150 trial closed after a median follow-up of 15.6 months. Consequently, reports of long-term outcomes for AB rely mainly on real-world evidence. We evaluated long-term effectiveness, safety, and clinically relevant on-treatment events in a large prospective real-world cohort of patients treated with AB. APPROACH AND RESULTS: We analyzed 538 patients prospectively enrolled in the Italian ARTE database. Outcomes included over

INTRA-ARTERIAL CHEMOTHERAPY IN ADVANCED PANCREATIC CANCER: A LITERATURE REVIEW.

Pancreatic cancer is an extremely aggressive malignancy with a 5-year survival rate among all stages of about 7%. Treatment options for unresectable forms are limited to fluoropyrimidine- and gemcitabine-containing regimens, and starting from the 3rd line, international guidelines do not provide clear recommendations because of the poor clinical condition of patients and the low efficacy of the consecutive lines. The arterial infusion is a promising method of treatment for locally advanced or liver-only metastatic disease, providing a higher local concentration with the same or lower systemic
