BACKGROUND: Few studies have examined the impact of multidisciplinary tumor board (MDT) discussion on pancreatic cancer treatment and diagnosis. We aimed to determine whether these patients who are presented at MDT meetings have improved outcomes compared with those who are not presented.
METHODS: A retrospective chart review was performed of patients diagnosed with pancreatic cancer at our tertiary referral center between 2019 and 2022.
RESULTS: Of the 190 patients included in the study, 166 (87.4%) were presented at MDT. Patients presented were more likely to receive appropriate pretherapy

BACKGROUND: The coexistence of Pancreatic cancer (PC) with diabetes mellitus (DM) and metabolic disorders (MDs) may further exacerbate the disease burden; however, the long-term mortality trends remain unclear.
METHODS: We utilized the CDC WONDER database to extract mortality data for PC and its comorbidities with diabetes mellitus and metabolic disorders among the U.S. population aged 25 and older from 1999 to 2023. We calculated the Age-Adjusted Mortality Rate (AAMR) and employed Joinpoint regression analysis to assess the average annual percentage change (AAPC), stratifying the data by sex

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) carries high mortality despite neoadjuvant chemotherapy (NAC) and surgical resection, creating an urgent need for prognostic biomarkers to guide treatment. We aimed to determine whether circulating tumor DNA (ctDNA) detected by next-generation sequencing (NGS) predicts survival in patients with localized PDAC.
METHODS: Patients with localized PDAC were prospectively enrolled at an academic institution between October 2020 and November 2024 (NCT04616131). Patients underwent next-generation sequencing (NGS), with blood samples collected at dia

Pancreatic ductal adenocarcinomas (PDACs) are found to account for more than 5% of all cancer-related deaths worldwide. The 5-year survival rates are still low (<5% for in the metastatic setting). In patients with metastatic disease, combination chemotherapy regimens are currently standard of care. PDACs are K-RAS addicted (incidence >90%), however, RAS oncogenes have been regarded to be "undruggable." Several highly specific inhibitors have been developed with atebimetinib (MEK inhibitor), daraxonrasib (tri-complex pan-K-RAS inhibitor), zoldonrasib (tri-complex K-RAS G12D inhibitor), INCB1617

Pancreatic ductal adenocarcinoma (PDAC) is characterized by aggressive metastatic progression and poor clinical outcomes. During metastatic dissemination, detached tumor cells must overcome anoikis, a form of apoptosis induced by loss of extracellular matrix attachment. Although anoikis resistance is considered a critical prerequisite for metastasis, the adaptive mechanisms supporting detached survival in pancreatic cancer remain incompletely understood. In this study, anoikis-resistant (AR) pancreatic cancer cell models were established through repeated long-term suspension culture using Capa

BACKGROUND: Pancreatic cancer is one of the most fatal malignancies with the lowest 5-year survival worldwide. Radioresistance remains the major challenge for pancreatic cancer treatment. The aim of this study is to unravel the mechanism underlying PAFAH1B1-regulated radiosensitivity in pancreatic ductal adenocarcinoma (PDAC).
METHODS: Immunohistochemistry (IHC), western blot or qRT-PCR were employed to detect the expression of PAFAH1B1, PUM2 and other molecules. Two radiation-resistant (RR) PDAC cell lines and a xenograft mouse model were established. CCK-8, colony formation assays and flow

Oncogenic mutations in the K-RAS small GTPase occur in more than 90% of human PDACs and cause dysregulation of multiple metabolic pathways, including polyamine metabolism. The goal of this study was to determine whether blocking polyamine synthesis with the irreversible inhibitor α-difluoromethylornithine (DFMO) provides a valid approach to targeting metabolic dependency in mutant K-RAS pancreatic tumors, compared with direct suppression of the oncogenic K-RAS protein. The anti-tumor activity of DFMO was evaluated in vitro and in vivo in the MIA PaCa-2 pancreatic cell line (K-RASG12C) and its

Liver-organotropic metastasis occurs in cancers such as uveal melanoma (UM) and pancreatic ductal adenocarcinoma (PDAC) and is a leading cause of mortality at the terminal stage. Colonization is a rate-limiting and converging step in the metastatic cascade. The drivers underlying liver colonization have largely remained elusive. In this report, we hypothesized that pioneer transcription factors (TFs) increase chromatin accessibility and superenhancer (SE) remodeling, which subsequently promote colonization-associated cellular features [e.g., proliferation, survival, cancer stem cells (CSCs), m

Undifferentiated carcinoma (UC) is a rare, highly aggressive subtype of pancreatic cancer; UC with rhabdoid features (UCRF) is particularly associated with poor prognosis. This study aimed to compare clinicopathologic characteristics and outcomes of UC between groups of patients defined by presence versus absence of RF. We retrospectively reviewed patients diagnosed with UC by EUS-guided tissue acquisition from 2017 to 2024. Histologically, 11 patients were classified as anaplastic UC and two had UC with osteoclast-like giant cell. Six patients had UCRF. Only 2 patients underwent surgery, whil

OBJECTIVES: Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal solid tumours. Alterations in Hippo, NF-κB and interferon (IFN)-related signalling have each been implicated in its biology, but the extent to which these transcriptional programmes are coordinately dysregulated in tumour tissue remains incompletely defined. This study therefore set out to determine whether these programmes are coordinately upregulated in PDAC tissue, and whether individual transcripts discriminate tumour from non-tumour tissue reproducibly in an independent cohort.
METHODS: In this retrospective st

Pancreatic cancer is a highly aggressive malignancy of the digestive tract, often associated with poor prognosis, short survival, and diminished quality of life upon diagnosis. To improve clinical outcomes and extend patient survival, early intervention at the etiological level is essential to reduce the risk of onset and progression. Recent studies have highlighted the significant roles of both psychological and physical stress in the initiation and development of pancreatic cancer. This article systematically examines the link between stress and pancreatic carcinogenesis from the dual dimens

BACKGROUND: Hepatic cysts are fluid-filled liver lesions that are mostly asymptomatic, but fewer than 15% of patients present clinically significant symptoms. Reported prevalence ranges from 2.5% to 71%, yet prior studies have been limited by small populations and incomplete assessment of risk factors. This study aimed to determine the prevalence, incidence, and risk factors using data from a large health screening cohort in Korea.
METHODS: We performed a retrospective cohort analysis of 78,676 participants in the Health and Prevention Enhancement cohort who underwent abdominal ultrasound at

AIMS: Autoimmune pancreatitis (AIP) is a representative manifestation of IgG4-related disease characterized by lymphoplasmacytic inflammation and storiform fibrosis. Although epigenetic dysregulation has been implicated in autoimmune diseases, its contribution to the pathogenesis of AIP remains unclear.
METHODS AND RESULTS: Genome-wide DNA methylation profiling was performed using the Infinium MethylationEPIC array in tissues affected by AIP and obstructive pancreatitis (OP). Differentially methylated regions (DMRs) were identified and subjected to pathway enrichment analysis. Among the signi

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) is an aggressive malignancy characterized by late diagnosis, rapid progression, and resistance to conventional therapies. Mutations in the tumor suppressor gene TP53 are prevalent in this cancer and have been associated with gain-of-function activities that promote tumor survival. Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, represents a potential therapeutic vulnerability. However, the influence of mutant p53 on ferroptosis susceptibility remains poorly understood.
METHODS: We employed isogenic p

Gastroenteropancreatic neuroendocrine neoplasms form a heterogeneous and complex group of tumors with varying degrees of malignant potential. Their incidence has significantly increased in recent decades. Due to their generally indolent nature, they are often discovered incidentally or following extensive investigations for nonspecific symptoms, leading to diagnostic delays. Notable advances have been made in diagnostic and therapeutic options, including radionuclide therapy, endoscopic resection or radiofrequency ablation, somatostatin analogs, and targeted molecular therapies. A multidiscipl

BACKGROUND: People living with cystic fibrosis (CF) may suffer from chronic respiratory infections due to acquired environmental organisms, ultimately leading in increased morbidity and mortality. Consequently, parents of children living with CF frequently ask their CF healthcare professional about sources and potential routes of transmission of environmental organisms to their child. This allows parents to make an informed decision regarding risk assessment/risk management of outdoor/ nature-based activities. Mitigations can then be put in place to help minimise or absolutely avoid exposure o

Pancreaticoduodenectomy is a technically demanding procedure associated with substantial surgical risk. Conventional laparoscopic approaches have several limitations, including restricted visualization, which can make exposure of the superior mesenteric artery and identification of vessels and lymph nodes challenging. In patients with vascular adhesion or tumor invasion, surgical dissection is particularly difficult and may increase the risk of intraoperative vascular injury and hemorrhage. In addition, poor delineation of anatomical planes may result in incomplete tumor dissection, inadequate
