Structural basis for broad orthopoxvirus neutralization by two antibodies targeting distinct L1R epitopes.

The 2022 mpox outbreak highlights the persistent threat posed by the mpox virus (MPXV) and related orthopoxviruses (OPXVs). Yet, approved antibody-based therapeutics for OPXVs are lacking. Here, we present two broadly neutralizing monoclonal antibodies (mAbs), G4 and B9, that target the MPXV M1R protein, homologous to L1R in other OPXVs. Both mAbs demonstrate cross-reactive binding and robust neutralization against multiple pathogenic OPXVs. Notably, G4 exhibits exceptional potency, neutralizing mature virions of VACV with an IC50 of 37.93 pM, whereas B9 has an IC50 of 41.13 nM. Cryo-electron

Structure-based multi-pocket virtual screening identifies FDA-approved candidate compounds targeting MPXV thymidylate kinase.

BACKGROUND: Mpox, caused by monkeypox virus (MPXV), has emerged as a growing global health concern, yet effective targeted antiviral therapies remain limited. Thymidylate kinase (TMK), an essential enzyme required for viral DNA replication, represents a potential therapeutic target. METHODS: We established a structure-based drug repurposing workflow integrating multi-pocket virtual screening and molecular dynamics (MD) simulations. A curated library of FDA-approved compounds was screened across five predicted TMK binding pockets, followed by consensus ranking based on cross-pocket recurrence

Occupational Mpox in personal protective equipment-compliant registered nurse reinforces importance of CDC 21-day self-monitoring after patient care.

A rare case of suspected occupational mpox infection occurred in a registered nurse who developed laboratory-confirmed disease despite strict adherence to full personal protective equipment and isolation precautions while caring for a suspected mpox patient. Retrospective review, staff interviews, and public health collaboration identified no clear breach in infection prevention practices, suggesting possible doffing-related or unrecognized exposure. This case highlights persistent transmission risk to healthcare workers and reinforces the importance of meticulous personal protective equipment

Mpox Vaccine Acceptance Among Health Care Workers in Central Asia: The Role of COVID-19 Experience.

BACKGROUND: The COVID-19 pandemic may have shaped health care workers' (HCWs') perceptions of emerging infectious threats and willingness to adopt new vaccines. This study examined whether COVID-19-related experiences and psychological factors were associated with Mpox vaccine acceptance among HCWs in Central Asia. METHODS: In this cross-sectional study, 3,054 HCWs from Kazakhstan, Kyrgyzstan, and Uzbekistan completed an online survey between July and November 2022. Mpox vaccine acceptance was assessed using a single 5-point Likert-scale item and analyzed as a continuous outcome. Measures inc

Development and evaluation of a two-tube multiplex real-time PCR assay for differential detection of Monkeypox virus clades Ia, Ib, IIa, and IIb.

Monkeypox virus (MPXV) has caused two Public Health Emergencies of International Concern since 2022, with co-circulation of distinct clades (Ia, Ib, IIa, IIb) that differ in virulence and transmission patterns. Rapid discrimination of these clades is critical for outbreak control and clinical management. Here, we developed and validated a two-tube multiplex real-time quantitative PCR (qPCR) assay targeting the H3L gene, designed to first distinguish clade I from clade II, followed by subtyping into clades Ia, Ib, IIa, and IIb in separate reactions. The assay demonstrated high sensitivity, with

A retrospective comparative diagnostic evaluation of three commercial qPCR kits for the diagnosis of mpox virus on an enriched sample set.

Mpox virus (MPXV) is an enveloped double-stranded DNA virus belonging to the genus Orthopoxvirus within the family Poxviridae. It causes a zoonotic disease transmitted to humans through direct contact with infected animals or humans, as well as through exposure to contaminated materials. The aim of this study was to conduct a retrospective, comparative diagnostic evaluation of three commercial real-time PCR (qPCR) kits for the diagnosis of MPXV on an enriched, artificially balanced sample set that included 50% MPXV-positive samples. World Health Organization (WHO) reference molecular protocol

Long-term exposure to PM2.5-bound metals and female breast cancer incidence in New Mexico, USA.

PURPOSE: Anxiety and depressive symptoms are highly prevalent among individuals with type 2 diabetes mellitus (T2DM), yet they remain underdiagnosed despite their substantial impact on health outcomes. In addition to impairing treatment adherence, these conditions may contribute to the development of chronic complications, reduce quality of life, and, in severe cases, increase mortality. Therefore, this review aims to estimate the pooled prevalence of anxiety symptoms and depressive symptoms among adults with T2DM and to explore potential sources of between-study heterogeneity, including asses

Fine mapping of QTL and identification of candidate genes responsible for tuberous stem formation in Kohlrabi (Brassica oleracea var. gongylodes L.).

Monkeypox is a contagious viral disease that spreads between animals and people. The UK government guidance described the first case of 'Monkey Pox' in 1958, when it was found only in monkeys used for research purposes. Fortunately, for a third world fast developing country like India, monkeypox does not spread easily in the population but spread by close physical contact between people, and there is limited information available about the impact on pregnancy. The virus can enter the body through broken skin, the respiratory tract, or mucous membranes (the moist inner lining of cavities and so

Modeling the impact of vaccine dose prioritization strategies during the 2022 mpox outbreak.

Early in the 2022 mpox outbreak, the US recommendation was to administer two doses of the JYNNEOS vaccine 4 weeks apart. However, because of limited vaccine supply, New York City (NYC) prioritized single dose vaccination. We estimated mpox cases averted by this strategy compared to strategies that prioritized 2-dose vaccination for a smaller portion of the population. We fit a network transmission model to incident mpox cases in NYC. Model output consisted of predicted cases over time when vaccine doses were administered with the "first-dose priority" strategy, compared with counterfactual sim

A Mini Review on Monkey-Pox: Outbreak, Challenges, and Management Strategies.

Monkeypox (Mpox) has become a significant global health concern, particularly since 2022. It has spread rapidly to numerous locations, and we urgently need to learn more about it. This overview discusses what Mpox is, how it spreads, its effects on people, and the medications that can be used to assist. Between January 2022 and March 2025, approximately 129,523 people were infected with Mpox in 120 countries. This demonstrates that HPV has progressed from being passed down from animals to being transmitted between people, including through intimate touch, as in some sexual interactions. Scient
