Lineage replacement of coxsackievirus A10 in Vietnam associated with increased detection and altered disease severity grading of hand, foot, and mouth disease.

Coxsackievirus A10 (CV-A10) has become an important cause of hand, foot, and mouth disease (HFMD). Molecular surveillance in northern Vietnam during 2018–2020 revealed the introduction and rapid dominance of the genotype C lineage (CHN), replacing the previously endemic lineage (VNM). This shift was associated with a significant increase in detection and altered clinical severity, predominantly grade 2A (83.8%). Phylogenetic analysis revealed the close relatedness between the emerging Vietnamese strains and those circulating in China (2017–2024). These findings suggest lineage-dependent virule

Impact of COVID-19 on the transmission dynamics of HFMD associated enterovirus serotypes in Japan: A modelling study of surveillance data.

A notable decline in incidence of hand, foot, and mouth disease (HFMD) was observed globally since the beginning of the COVID-19 pandemic, indicating a change in the transmission dynamics of enteroviruses (EVs) causing HFMD. This study estimates the impact of COVID-19 non-pharmaceutical interventions (NPIs) on the transmission of EVs in Japan. Japan has a well-established sentinel surveillance system for enteroviruses and HFMD. A susceptible-infected-recovered stochastic transmission model was fitted to the time-series of weekly reported number of HFMD enterovirus A71 (EV-A71), coxsackievirus

Regional and facility-type variations in infectious diseases in childcare and early childhood education facilities in Japan during the COVID-19 pandemic: A nationwide cross-sectional questionnaire survey.

Young children are susceptible to infectious diseases due to their developing immune systems and close contact in group care settings. During the coronavirus disease 2019 (COVID-19) pandemic, infection prevention measures may have altered the epidemiology of common childhood infections, yet evidence on variations by facility type and region remains limited. In this study, the occurrence of COVID-19 and child-specific infectious diseases in childcare and early childhood education facilities in Japan was examined with particular focus on facility type and regional population density. A nationwid

Neutral sphingomyelinase 2 knockdown attenuates disease severity and modulates immune responses in enterovirus A71-infected mice.

Severe hand, foot, and mouth disease (HFMD) is a febrile illness that is caused by enterovirus A71 (EV-A71). We previously identified neutral sphingomyelinase 2 (NSM2) as a critical host factor that supports EV-A71 replication in vitro. To define its role in vivo, we employed a suckling mouse model of EV-A71 infection. Using a small interfering RNA (siRNA) targeting NSM2 (si-NSM2), we achieved efficient knockdown. Treatment with si-NSM2 significantly reduced EV-A71 viral protein levels and genomic RNA loads in the brain, lung, heart, and intestine of infected ICR suckling mice. We further eval

Acute Fulminant Cerebral Edema Caused by Coxsackievirus A6 Infection: A Case Report.

In recent years, Coxsackievirus A6 (CV-A6) has gradually replaced Enterovirus 71 (EV-71) and Coxsackievirus A16 (CV-A16) as the main pathogen causing hand foot mouth disease (HFMD) in China. This article reports a fatal case of HFMD caused by CV-A6, leading to fulminant cerebral edema and cardiopulmonary arrest. A 6-year-old boy was admitted with a chief complaint of "fever for 1 day, two episodes of seizures". On admission, the patient exhibited unresponsiveness, no spontaneous breathing, bilateral fixed and dilated pupils, complete muscle weakness, and loss of muscle tone. The brain computed

Validation of important genes and bioinformatics investigation of the relationship between ferroptosis and CVA16 infection.

Coxsackievirus A16 (CVA16) is a major pathogen linked to hand, foot, and mouth disease (HFMD), and effective therapies and vaccines are lacking. Ferroptosis, an iron-dependent form of cell death, may play a role in CVA16 infection and remains underexplored. We analyzed microarray datasets (GSE71673) from the Gene Expression Omnibus (GEO) and ferroptosis-related genes (FRGs) from FerrDb. A Venn diagram revealed 37 ferroptosis-related differentially expressed genes (FR-DEGs), and Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were conducted on these genes. Standardized neutralizing antibody assay for Coxsackievirus A10 in clinical trial: A framework for multivalent hand, foot, and mouth disease vaccine evaluation.

Enteroviruses (EVs) are the major etiological agents of hand, foot, and mouth disease (HFMD), which could lead to severe complications and even fatalities. Owing to the alternating or co-circulation of multiple EV serotypes and the lack of cross-protection, global institutions are pursuing the development of multivalent HFMD vaccines. Notably, the advancement of innovative multivalent HFMD vaccines into the clinical stage highlights the urgent need for a standardized platform to assess immunogenicity. In accordance with international regulatory guidelines, a standardized neutralizing antibody

CBX4 facilitates EV71 replication by SUMOylation and stabilizing 3D polymerase.

Enterovirus 71 (EV71) is a primary etiological agent of hand-foot-mouth disease (HFMD) in children under 5 years of age and can cause severe neurological disorders even death. Therefore, elucidating the infection mechanism and pathogenicity of EV71 is essential for developing more effective and targeted therapies to prevent and control EV71-associated diseases. Here, we initially reported that the SUMO E3 ligase CBX4 is important for EV71 replication. Furthermore, we found that CBX4 interacts with the EV71 3D polymerase, and overexpression of CBX4 significantly extends the half-life of 3D, whe
