Neurofibromatosis type 1 (NF1) is the most prevalent neurocutaneous syndrome, affecting approximately 1 in 3000 individuals. This genetic disorder involves multiple organ systems and exhibits marked variability in clinical presentation, with characteristic cutaneous, ophthalmological, neurological, cardiovascular, skeletal, and neurodevelopmental features. It may also be associated with multiple neoplasms. Timely and accurate diagnosis is essential to ensure optimal patient care. A lifelong, multidisciplinary management approach is recommended. The objective of this consensus is to summarize t

BACKGROUND/OBJECTIVES: Neurofibromatosis type 1 (NF1) is an autosomal dominant disorder caused by pathogenic variants in the NF1 gene, characterized by high phenotypic variability.
METHODS: We present clinical and molecular data from a Romanian cohort of 54 patients initially diagnosed clinically.
RESULTS: Phenotypic evaluation (n = 54) revealed a high prevalence of café-au-lait macules (100%), Lisch nodules (64.8%), axillary/inguinal freckling (61.1%), and cutaneous neurofibromas (42.6%). Due to financial constraints (genetic testing not covered by the national health system), molecular con

We report the case of an 11-year-old female who presented with a progressively enlarging cystic neck swelling. CT demonstrated a multiloculated cystic lesion involving the right cervical soft tissue planes, suggestive of a cystic lymphangioma. There was no significant family history or clinical evidence of neurofibromatosis type 1 (NF1) at presentation. Surgical excision was performed, and histopathological examination with S100 immunohistochemistry confirmed the diagnosis of plexiform neurofibroma. At the one-year follow-up, the patient developed multiple café-au-lait macules and Lisch nodule

Abetalipoproteinemia (ABL) affects fewer than 1 per 1 million people, whereas neurofibromatosis type 1 (NF1) occurs in approximately 1 in 3000 to 4000 individuals. We describe the clinical findings in a 38-year-old woman with both disorders. ABL was diagnosed in the neonatal period, followed by NF1 at age 7 years based on multiple café-au-lait spots, intertriginous freckling, and Lisch nodules. Genetic testing revealed a homozygous pathogenic MTTP frameshift variant [c.215del (p.Pro72Leufs*8)] and a heterozygous pathogenic NF1 splice variant [c.205-2A>C]. To our knowledge, this co-occurrence h

Neurofibromatosis type 1 (NF1) is a common autosomal dominant neurocutaneous syndrome that affects 1 in 2,500-3,000 births. The phenotypic hallmarks include Lisch nodules, optic nerve gliomas, café-au-lait spots, axillary freckling, and benign neurofibromas. NF1 patients carry an 8-13% lifetime risk of malignant peripheral nerve sheath tumors, the leading cause of NF1 mortality. Current genetic testing detects NF1 variants in 95-97% of cases. In this prospective cohort of 103 cases, we used targeted NF1 sequencing by next-generation sequencing, multiplex ligation-dependent probe amplification

Plexiform neurofibroma is a benign peripheral nerve sheath tumor considered pathognomonic for neurofibromatosis type 1. Isolated scrotal occurrence without systemic manifestations of neurofibromatosis type 1 is exceedingly rare. We report a 16-year-old male with a progressively enlarging right scrotal mass measuring 8 × 4 cm. There were no café-au-lait spots, axillary freckling, or family history of neurofibromatosis type 1. Magnetic resonance imaging showed the characteristic target sign. Complete surgical excision via a scrotal approach with testicular preservation was performed. Histopathol

BACKGROUND: Malignant Triton Tumor (MTT) is a rare, highly aggressive peripheral nerve sheath tumor characterized by rhabdomyoblastic differentiation, seldom occurring primarily in the thoracic cavity. This report presents a rare case of MTT initially manifesting as chest pain, and reviews relevant literature to summarize its clinical features and therapeutic strategies.
METHODS: A 23-year-old woman experienced chest pain for ten days and her condition worsened within 3 days. The patient had a known history of neurofibromatosis type 1 (NF1) and exhibited classic café-au-lait spots on physical

BACKGROUND: Neurofibromatosis type 1 (NF1) is a complex neurocutaneous disorder caused by NF1 variants that disrupt neurofibromin regulation of the Ras pathway, increasing tumor risk. Despite the global recognition, data from the Middle East remain scarce with no large cohorts reported. This study investigates large cohort of NF1 patients in Oman.
METHODS: A retrospective study was conducted to analyze the epidemiologic, clinical, and molecular data of NF1 patients followed at Sultan Qaboos University Hospital and Royal Hospital between 2010 and 2024.
RESULTS: A total of 211 NF1 patients wer

INTRODUCTION: Gastrointestinal stromal tumors (GISTs) are the most common mesenchymal tumors of the GI tract. GISTs in patients with neurofibromatosis type 1 (NF1) are relatively common, but periampullary GISTs associated with NF1 and idiopathic thrombocytopenic purpura (ITP) are exceedingly rare. This case report presents a unique and challenging instance of a periampullary GIST in a patient with NF1 and ITP, treated surgically with an uneventful recovery.
CASE PRESENTATION: A 40-year-old female with a known history of NF1 presented with epigastric pain, multiple café-au-lait spots, and cuta

BACKGROUND Neurofibromatosis type 1 (NF1) and Marfan syndrome (MFS) are genetically determined systemic disorders. Their simultaneous occurrence is exceptionally rare, with only a few cases reported in the literature. CASE REPORT A 13-year-old boy was admitted to a cardiology clinic due to mitral and tricuspid valve prolapse. He was tall, with severe scoliosis and distinctive dysmorphic features typical of MFS. More than 20 cafe-au-lait spots, characteristic of NF1, were present on his skin. The family history included NF1 in his twin brother, mother, maternal uncle, and maternal grandmother.

The Journal of Genetics, started by William Bateson in 1910, played a distinguished role in the early years of genetics. However, it stopped publishing in 1978. The Indian Academy of Sciences revived it in 1985, and has published it regularly since then. To commemorate this landmark, I highlight one of the 17 articles published that year. 'The isolation and genetic analysis of a Caenorhabditis elegans… X-chromosome balancer' by András Fodor and Péter Deak, of the Institute of Genetics, Biological Research Centre, Hungarian Academy of Sciences, Szeged, Hungary. Transcription factors from the family of Signal Transducers and Activators of Transcription (STAT) are activated by numerous cytokines. Two members of this family, STAT5A and STAT5B (collectively called STAT5), have gained prominence in that they are activated by a wide variety of cytokines such as interleukins, erythropoietin, growth hormone, and prolactin. Furthermore, constitutive STAT5 activation is observed in the majority of leukemias and many solid tumors. Inactivation studies in mice as well as human mutations have provided insight into many of STAT5's functions. Disruption of cytokine

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

NF1 is an autosomal dominant hereditary disease, with a prevalence of at least 1 in 4000-5000 population. The diagnosis criteria of NF1 included typical manifestations such as café-au-lait spots, frecking in the axilla or inguinal region, multiple neurofibromas, Lisch nodeules, and distinctive osseous lesions. Genetic testing shows NF1 mutation. It is essential for tumor surveillance in NF1 patients because their life expectancy is about 54 years due to malignancy. A case of NF-1 patient receive laparoscopic small bowel resection and finally diagnosed as adenocarcinoma and ganglioneuroma. Abou

To investigate the efficacy and safety of picosecond (PS) and nanosecond (NS) 1064-nm neodymium-doped yttrium aluminum garnet (Nd: YAG) laser in treating Café-au-lait macules (CALMs). We retrospectively analyzed the medical records of patients with CALMs, who were treated with PS or NS 1064-nm lasers from January 2020 to January 2022. The efficacy was determined based on the before and after pictures by two independent investigators. The incidence of adverse events and recurrence were explored after treatments. A total of 116 patients (83 in the PS group and 33 in the NS group) were recruited.
