An anti-fouling multifunctional interface enables hierarchical validation and ultrasensitive electrochemical detection of Coxsackievirus A6 specific nucleic acids.

Coxsackievirus A6 (CVA6) has emerged as the dominant etiological agent of hand, foot and mouth disease worldwide. At present, neither specific antivirals nor effective vaccines are available, creating an urgent need for rapid, low-cost in vitro diagnostics that can interrupt transmission. This study developed an electrochemical biosensor based on a "screening-validation" strategy. It utilizes a MoS2/MWCNT/AgNPs three-dimensional highly conductive network as the substrate, tightly anchored with p(NIPAm-co-SBMA) microgel to form a "protective-conductive" synergistic interface. The capture probe

Dysfunction of the plasmacytoid dendritic cells in enterovirus 71-induced HFMD affecting children.

The role of Plasmacytoid Dendritic Cells (pDCs) in Enterovirus-71 (EV71)-induced Hand, Foot, and Mouth Disease (HFMD) remains unclear. In this study, we investigated pDCs in severe HFMD patients infected with EV71. Flow cytometry revealed reduced pDC percentage in severe patients compared to healthy controls. In vitro, as EV71 replicated in pDCs, live cell numbers and levels of Interferon-alpha (IFN-a) and Interleukin (IL)-6 decreased, while IL-10 and Tumour Necrosis Factor-alpha (TNF-a) increased, particularly in critically ill patients. Notably, pDCs derived from critically ill patients exhi

Long-term protection of an inactivated enterovirus type 71 vaccine against hand, foot, and mouth diseases in children: a modelling study.

OBJECTIVES: Despite the availability of enterovirus type 71 (EV71) vaccines, evidence on long-term protection remains limited. We aimed to establish a mathematical model for predicting the 20-year long-term protection of the EV71 vaccine. METHODS: We utilized data from a phase 3 trial in which infants and young children were randomized to receive either two doses of 400 U EV71 vaccine or placebo. Neutralizing antibody data (NTAb) were obtained from the immunogenicity subpopulation who were followed for 5 years. We compared candidate decay models using Akaike Information Criterion (AIC) and Ba

Pembrolizumab With Anlotinib in Advanced Oral Mucosal Melanoma: A Retrospective Trial.

INTRODUCTION: Oral mucosal melanoma (OMM) is a rare subtype of melanoma but exhibits highly invasive biological behavior. Programmed cell death protein 1 (PD-1) monotherapy showed lower response in OMM than other subtypes of melanoma. MATERIALS AND METHODS: We retrospectively analyzed the efficacy and safety of pembrolizumab with anlotinib in patients with advanced OMM between August 2018 and September 2024. The primary endpoint was objective response rate (ORR); the secondary endpoints included disease control rate (DCR), duration of response (DOR), progression-free survival (PFS), overall s

Rapid and Visual Detection of Coxsackievirus A6 Using Reverse Transcription Multiple Cross Displacement Amplification Combined with Nanoparticle-Based Biosensor Platform.

Coxsackievirus A6 (CVA6) is the most common causative pathogen of hand, foot, and mouth disease (HFMD) in children under 5 years of age and has caused multiple outbreaks in recent years. Currently, no effective vaccines or antiviral treatments are available. The present study introduces a detection assay, termed CVA6-RT-MCDA-LFB, which combines reverse transcription multiple cross displacement amplification (RT-MCDA) with a nanoparticle-based lateral flow biosensor (LFB). Ten specific primers targeting the VP1 gene region were designed for CVA6-RT-MCDA. The assay demonstrated an analytical sen

Structural tuning of bridging linkers on M13 phages for high-affinity detection of enterovirus A71.

Enterovirus A71 (EV-A71), the principal etiological agent of hand, foot, and mouth disease, poses a serious public health concern because infection can induce severe and potentially fatal neurological complications through direct invasion and inflammation of the central nervous system. Current detection methods lack rapid and sensitive analytical capabilities, particularly in the context of wastewater surveillance. This study reports a PEG4-biotin-functionalized M13 bacteriophage engineered as a molecular receptor for the colorimetric detection of EV-A71. VP1-specific phage ligands were identi

A randomised trial of a bioreactor-produced EV-A71 vaccine for endemic control in children.

Enterovirus (EV) A71 causes endemic outbreaks of hand-foot-mouth disease (HFMD) and herpangina in young children, occasionally leading to severe complications and fatal outcomes. We evaluated EnVAX-A71, a bioreactor-produced, aluminum-adjuvanted inactivated EV-A71 (B4/E59) vaccine, in a randomized, double-blind, placebo-controlled phase 3 trial in Taiwan and Vietnam. A total of 4011 children aged 2-71 months were randomized 3:2 to vaccine or placebo; the primary endpoint was laboratory-confirmed EV-A71-associated HFMD/herpangina, evaluated from 28 days after dose two over a follow-up of 1 to 2

Real-time dynamic prediction of HFMD transmission using SEIRQ-ARIMA hybrid model optimized by multi-stage ABC-GWO algorithm.

In Hand, Foot, and Mouth Disease (HFMD) control, conventional SEIR/SIR models rely on static parameters, which limits their adaptability to dynamic real‑world conditions. To address this, we propose a SEIRQ–ARIMA hybrid model that integrates a quarantine‑enhanced SEIR framework (SEIRQ) with an ARIMA time‑series model, whose dynamic parameters are optimized via a multi‑stage Artificial Bee Colony–Grey Wolf Optimization (ABC–GWO) algorithm. This study contributes three key novelties to the field: (i) an IoT-driven dynamic SEIRQ parameterization with explicit intervention tracking via time-varyin

Lineage replacement of coxsackievirus A10 in Vietnam associated with increased detection and altered disease severity grading of hand, foot, and mouth disease.

Coxsackievirus A10 (CV-A10) has become an important cause of hand, foot, and mouth disease (HFMD). Molecular surveillance in northern Vietnam during 2018–2020 revealed the introduction and rapid dominance of the genotype C lineage (CHN), replacing the previously endemic lineage (VNM). This shift was associated with a significant increase in detection and altered clinical severity, predominantly grade 2A (83.8%). Phylogenetic analysis revealed the close relatedness between the emerging Vietnamese strains and those circulating in China (2017–2024). These findings suggest lineage-dependent virule

Impact of COVID-19 on the transmission dynamics of HFMD associated enterovirus serotypes in Japan: A modelling study of surveillance data.

A notable decline in incidence of hand, foot, and mouth disease (HFMD) was observed globally since the beginning of the COVID-19 pandemic, indicating a change in the transmission dynamics of enteroviruses (EVs) causing HFMD. This study estimates the impact of COVID-19 non-pharmaceutical interventions (NPIs) on the transmission of EVs in Japan. Japan has a well-established sentinel surveillance system for enteroviruses and HFMD. A susceptible-infected-recovered stochastic transmission model was fitted to the time-series of weekly reported number of HFMD enterovirus A71 (EV-A71), coxsackievirus

Regional and facility-type variations in infectious diseases in childcare and early childhood education facilities in Japan during the COVID-19 pandemic: A nationwide cross-sectional questionnaire survey.

Young children are susceptible to infectious diseases due to their developing immune systems and close contact in group care settings. During the coronavirus disease 2019 (COVID-19) pandemic, infection prevention measures may have altered the epidemiology of common childhood infections, yet evidence on variations by facility type and region remains limited. In this study, the occurrence of COVID-19 and child-specific infectious diseases in childcare and early childhood education facilities in Japan was examined with particular focus on facility type and regional population density. Neutral sphingomyelinase 2 knockdown attenuates disease severity and modulates immune responses in enterovirus A71-infected mice.

Severe hand, foot, and mouth disease (HFMD) is a febrile illness that is caused by enterovirus A71 (EV-A71). We previously identified neutral sphingomyelinase 2 (NSM2) as a critical host factor that supports EV-A71 replication in vitro. To define its role in vivo, we employed a suckling mouse model of EV-A71 infection. Using a small interfering RNA (siRNA) targeting NSM2 (si-NSM2), we achieved efficient knockdown. Treatment with si-NSM2 significantly reduced EV-A71 viral protein levels and genomic RNA loads in the brain, lung, heart, and intestine of infected ICR suckling mice. We further eval

Acute Fulminant Cerebral Edema Caused by Coxsackievirus A6 Infection: A Case Report.

In recent years, Coxsackievirus A6 (CV-A6) has gradually replaced Enterovirus 71 (EV-71) and Coxsackievirus A16 (CV-A16) as the main pathogen causing hand foot mouth disease (HFMD) in China. This article reports a fatal case of HFMD caused by CV-A6, leading to fulminant cerebral edema and cardiopulmonary arrest. A 6-year-old boy was admitted with a chief complaint of "fever for 1 day, two episodes of seizures". On admission, the patient exhibited unresponsiveness, no spontaneous breathing, bilateral fixed and dilated pupils, complete muscle weakness, and loss of muscle tone. The brain computed

Validation of important genes and bioinformatics investigation of the relationship between ferroptosis and CVA16 infection.

Coxsackievirus A16 (CVA16) is a major pathogen linked to hand, foot, and mouth disease (HFMD), and effective therapies and vaccines are lacking. Ferroptosis, an iron-dependent form of cell death, may play a role in CVA16 infection and remains underexplored. We analyzed microarray datasets (GSE71673) from the Gene Expression Omnibus (GEO) and ferroptosis-related genes (FRGs) from FerrDb. A Venn diagram revealed 37 ferroptosis-related differentially expressed genes (FR-DEGs), and Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were conducted on these genes.
