Au clusterzymes encapsulated in nanogel activated by alternating magnetic field to promote white fat browning and combat obesity.

Obesity, a major global public-health challenge, is a well-established risk factor for numerous metabolic disorders. In this study, we developed a novel nanosystem based on Au clusterzymes (AuCZy) encapsulated in chitosan (CS) and functionalized with an adipose homing peptide (AHP), referred to as AHP-AuCZy@CS nanogel. Specifically, an alternating magnetic field (AMF) was applied to modulate its enzyme-mimicking activity. It was found that AMF stimulation significantly enhanced SOD-like and CAT-like activities, endowing AHP-AuCZy@CS nanogels with superior reactive oxygen species (ROS) scavengi

Is a variant of uncertain significance always 'insignificant'? A systematic review on PRF1 A91V in Hemophagocytic Lymphohistocytosis and comparative analysis with Still's disease.

BACKGROUND: Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory disorder that may arise secondary to rheumatic diseases such as Still’s disease, where macrophage activation syndrome (MAS) represents its clinical counterpart. The pathogenic significance of the PRF1 A91V variant remains uncertain, although functional data suggest partial perforin dysfunction and a possible contribution to late-onset or atypical HLH. This study aimed to clarify the clinical implications of the PRF1 A91V variant through a systematic review of published HLH and MAS cases and a comparati

Inflammation-targeted nanoaggregates encapsulated triptolide and hyperoside for M2 macrophage repolarization and oxidative stress reversal in rheumatoid arthritis therapy.

M1 macrophages are pivotal in rheumatoid arthritis (RA) pathogenesis, accompanied by releasing pro-inflammatory cytokines and reactive oxygen species (ROS). As a traditional anti-RA herbal ingredient, triptolide (TP) exerts potent anti-inflammatory effects via M1 macrophage modulation, but is clinically limited by poor solubility, ‌elevating ROS and systemic toxicity. Herein, we synthesized a β-cyclodextrin-grafted poly-L-glutamic acid copolymer, forming nanoaggregates (NPS) loaded with TP and hyperoside (HYP, a potent antioxidant) via host-guest interactions for RA treatment. The NPS exhibit

Ubiquitin-Specific Peptidase 16 Alleviates Chondrocyte Damage in Osteoarthritis Through Deubiquitinating and Stabilizing POU Class 2 Homeobox 1.

Osteoarthritis (OA), marked by articular cartilage degeneration, severely impairs joint function. Ubiquitin-specific peptidase 16 (USP16) is crucial for regulating cellular protein stability and signaling pathways. This study seeks to elucidate USP16's impact and mechanism on chondrocyte injury in OA. The expression of USP16 in OA was analyzed using the Gene Expression Omnibus (GEO) database. An in vitro model using IL-1β to induce OA-like conditions. Real-time quantitative PCR (RT-qPCR) and western blot were used to detect gene expression in tissues and cells. The cellular oxidative stress le

Mitochondrial-targeted therapy for osteoarthritis: Challenges and opportunities from basic research to clinical translation.

Osteoarthritis is a high-burden degenerative joint disease. Existing therapies only alleviate symptoms but fail to halt disease progression. Studies have identified mitochondrial dysfunction as a core driver of cartilage degeneration in OA. Key mechanisms include mitochondrial reactive oxygen species bursts that activate inflammatory and cell death pathways; imbalances in mitochondrial dynamics leading to fragmentation; autophagy defects causing damage accumulation; and reduced biogenesis coupled with hyperglycolysis, which exacerbates the energy crisis. Collectively, these processes accelerat

Unlocking an Efficient SOD-to-CAT Catalytic Relay in a MOF Nanozyme via Dopamine-Driven Interfacial Nanoreconstruction for Osteoarthritis Therapy.

Osteoarthritis (OA) is driven by a vicious cycle of inflammation and reactive oxygen species (ROS). While cobalt-based metal-organic framework (MOF) nanozymes are potent catalase (CAT) mimics, their therapeutic efficacy is crippled by an inherently weak superoxide dismutase (SOD)-like activity that prevents full-chain ROS scavenging. Here, we resolve this imbalance through an innovative "pre-embedding/activation" strategy. This approach involves pre-embedding Zn2+ into a cobalt-based framework to create a stable yet activatable precursor. The subsequent dopamine (DA)-driven "activation" then r

An in situ-forming hydrogel with a sol-spray system promotes diabetic wound healing via synergistic anti-inflammatory and antioxidant effects.

Chronic diabetic wounds remain a substantial clinical challenge, largely attributable to sustained inflammation, excessive oxidative stress, and deficient tissue regeneration. Herein, an in situ-forming hydrogel fabricated using a dual-component sol-spray system (AGBS-IG) is designed to effectively modulate the inflammatory-oxidative microenvironment and accelerate diabetic wound healing. The injectable sol component, composed of sodium alginate, polyvinyl alcohol, and itaconic acid, enables conformal filling of irregular wound beds and provides intrinsic anti-inflammatory activity, while the

PET-microplastics trigger endothelial glycocalyx loss via ER stress and ROS unleashing IL-1β-driven SMC switching and early aortic structural impairment.

Polyethylene terephthalate microplastics (PET-MPs), a major microplastics component identified in human vasculature, pose emerging environmental health risks. This study systemically profiled MPs in human aortic tissues and investigated the mechanisms underlying PET-MPs-induced aortic injury in vivo and in vitro. Chronic oral exposure of Sprague-Dawley rats to PET-MPs resulted in endothelial glycocalyx loss and structural impairment of aortic elastic fibers. Transcriptomic and proteomic analyses elucidated that PET-MPs triggered endoplasmic reticulum stress and reactive oxygen species generati

SIRT1 regulates dermal fibroblast senescence via impaired deacetylase function and mitochondrial dysfunction during skin aging induced by chronic oral cadmium exposure.

INTRODUCTION: Skin aging is a complex, multifactorial biological process that can be significantly accelerated by environmental toxicants such as cadmium (Cd), a highly toxic and ubiquitous heavy metal. Although the broad cytotoxic impacts of Cd have been extensively reported, a comprehensive understanding of the precise molecular pathways underlying Cd-induced skin senescence is still lacking. In this study, we investigated the protective role of Sirtuin 1 (SIRT1), a highly conserved nicotinamide adenine dinucleotide (NAD+)-dependent deacetylase that functions as a master regulator of mitocho

Serum IL-2 levels are associated with disease activity and related to dyslipidaemia and the immunological profile in systemic lupus erythematosus.

INTRODUCTION: Systemic lupus erythematosus (SLE) is an autoimmune disorder characterised by multisystem involvement, frequently including cardiovascular manifestations. Interleukin 2 (IL-2), a key cytokine in immune regulation, plays a pivotal role in maintaining tolerance by promoting regulatory T cell function. The relationship between serum IL-2 levels and specific SLE features remains incompletely defined. The aim of our study was to investigate the associations between serum IL-2 concentrations and disease activity, inflammatory markers, autoantibody profiles as well as cardiovascular and

HACCP-compatible ear acupuncture using biodegradable starch-based needles mitigates transport-induced physiological and oxidative stress in calves.

BACKGROUND AND AIM: Transportation is an unavoidable management practice in cattle production and is a major source of physiological, endocrine, and oxidative stress, leading to impaired welfare, immunity, and productivity. Drug-free and food-safety-compatible interventions to alleviate transport stress remain limited. Acupuncture has shown stress-reducing effects in livestock; however, conventional metal needles are incompatible with Hazard Analysis and Critical Control Point (HACCP) systems. Recently developed biodegradable starch-based Circular transdermal needles (CTNs) offer a novel HACCP

Reactive oxygen species in health and disease.

The traditional view of reactive oxygen species (ROS) as uniform toxicants has been superseded by the recognition of a fundamental radical/non-radical dichotomy. As radical and non-radical ROS differ in spatial and kinetic behaviors that dictate cellular impacts, understanding this dichotomy is essential for the design of ROS-targeting therapies. However, the roles of specific ROS types under physiological and pathological conditions remain inadequately defined, hindering precise clinical translation. By organizing ROS sources, neutralizing systems, reaction kinetics, biological effects, and t
