The protective role of Nesfatin-1 against TNF-α-induced ferroptosis in chondrocytes: Implications for osteoarthritis therapy.

Osteoarthritis (OA) progression is significantly driven by chondrocyte ferroptosis, a form of iron-dependent cell death triggered by oxidative stress and lipid peroxidation. This study investigated the protective effect of Nesfatin-1 against TNF-α-induced ferroptosis in mouse primary chondrocytes and explored the underlying mechanism. A ferroptosis model was established using TNF-α (10 ng/mL). Cell viability, cytotoxicity, and key ferroptosis markers were assessed. We found that endogenous NUCB2/nesfatin-1 is expressed in chondrocytes and downregulated by TNF-α. Nesfatin-1 treatment significan

Mitochondrial Metabolism in Neutrophils: Emerging Roles in Inflammatory Skin Diseases.

Neutrophils, the most abundant of the circulating leucocytes, play crucial roles in antimicrobial defence, tissue remodelling, and immune regulation. Traditionally regarded as predominantly glycolytic, relying on aerobic glycolysis (the Warburg effect) for rapid ATP generation, neutrophils are now recognised to possess broader metabolic flexibility. Emerging evidence has revealed their capacity for oxidative phosphorylation (OXPHOS), fatty acid oxidation (FAO), and glutaminolysis. As a result, mitochondrial metabolism is dynamically reprogrammed during differentiation and activation. These met

Melatonin Improves Diabetic-Accelerated Experimental Osteoarthritis by Inhibiting NLRP3-Mediated Chondrocyte Pyroptosis via PKG/Nrf2.

Recent evidence suggests a potential link between diabetes and the worsening and severity of osteoarthritis (OA) symptoms. Melatonin (MLT) has a strong ability to scavenge reactive oxygen species and lipid peroxidation, and plays a vital role in the treatment of OA. However, the role and mechanism of MLT in diabetic OA are still unclear. This study aimed to confirm that diabetes aggravates OA cartilage degeneration and to explore the specific mechanism by which MLT inhibits cell pyroptosis and alleviates OA. In this study, a model of OA combined with type 2 diabetes was established, and it was

Low-dose deflazacort reduces postoperative pain and inflammation following primary total knee arthroplasty: A randomized controlled trial.

INTRODUCTION: Postoperative pain, inflammation, and sleep disturbance remain major challenges following total knee arthroplasty (TKA). While perioperative corticosteroids have demonstrated benefits, the role of extended low-dose oral corticosteroid therapy during postoperative recovery is not well defined. Therefore, this study aimed to evaluate the efficacy and safety of low-dose oral deflazacort in reducing postoperative pain and improving sleep quality following primary TKA. The hypothesis was that perioperative low-dose deflazacort would reduce postoperative pain and improve functional rec

Inflammatory stimulus enhances synaptic material uptake by adult APP microglia in a microfluidic neuron-microglia co-culture model.

Microglia play a critical role in synapse remodeling and neuroinflammation, both of which are dysregulated in Alzheimer’s disease (AD). However, most in vitro models rely on neonatal or immortalized microglia, limiting their relevance to adult pathophysiological context. Here, we present a compartmentalized microfluidic co-culture platform that enables spatially controlled interactions between primary cortical neurons and adult microglia from wild-type (WT) and APP-transgenic mice. This system allows precise functional analysis of microglia–synapse interactions under defined inflammatory condi

Chondroitin sulfate alleviates osteoarthritis by upregulating HSPA8 to inhibit chondrocyte ferroptosis.

Osteoarthritis (OA) is a prevalent degenerative joint disease with no curative treatment currently available. Recent evidence suggests that chondrocyte ferroptosis contributes to OA progression. Chondroitin sulfate (CS), widely used in OA management, exhibits anti-inflammatory and antioxidant properties, yet its role in modulating ferroptosis remains unclear. In this study, we investigated whether CS alleviates OA by inhibiting chondrocyte ferroptosis and explored the underlying mechanisms. Using an in vitro ferroptosis model induced by RSL3 in rat chondrocytes, we found that CS significantly

Dental plaque metabolites induce polymorphonuclear leukocyte death accompanying peptidyl arginine deiminase 4 and citrullinated protein release: Potential contribution to rheumatoid arthritis pathogenesis.

OBJECTIVES: Neutrophils form the front line of defense in the periodontal pocket. They release the web-like DNA structures, known as neutrophil extracellular traps (NETs). NETosis, a form of programmed cell death, is characterized by the release of NETs, peptidyl arginine deiminase (PAD), and citrullinated proteins. Plaque bacteria produce a variety of metabolites that can interact with neutrophils and potentially modulate its function. In this study, how these bacterial metabolites influence NETosis-like cell death in polymorphonuclear leukocytes (PMNL) was investigated. METHODS: SYTOX Green

Reappraising heme oxygenase-1 as a ferroptosis modulator in atherosclerosis: a mechanism-focused review.

Atherosclerosis is the primary pathological basis of cardiovascular diseases, with macrophage dysfunction, lipid accumulation, and oxidative stress driving plaque formation and progression. Ferroptosis, an iron-dependent form of regulated cell death characterized by lipid peroxidation, has recently emerged as a pivotal mechanism influencing atherosclerosis. Heme oxygenase-1 (HO-1), a key regulator of heme catabolism and iron homeostasis, exerts dual roles in this process: moderate HO-1 activity confers cytoprotection through antioxidant effects, whereas excessive HO-1 expression promotes intra

Self-Assembled EGCG Nanoparticles Achieved Long-Term ROS Scavenging to Delay Osteoarthritis Progression.

Oxidative stress disrupts the synthesis-degradation balance of the extracellular matrix in osteoarthritic (OA) cartilage, resulting in the loss of type II collagen (COLII). Here, we developed self-assembled nanoparticles (PE@NPs) driven by hydrophobic interaction, π-π stacking interactions and hydrogen bonding, forming an epigallocatechin-3-gallate (EGCG) core and a polyethylene glycol (PEG) shell. Compared with free EGCG, which possesses potent but short-lived antioxidant activity, PE@NPs improved molecular stability, extending reactive oxygen species scavenging activity to 24 h. Furthermore,

Neutrophil extracellular traps-inhibited nanoplatform achieves comprehensive treatment of rheumatoid arthritis at different stages.

Rheumatoid arthritis (RA) is an autoimmune disease characterized by cartilage and bone erosion. The development process of RA could be divided into early, full blown, and late stages. The pathogenesis of those stages is still far from fully understood, which poses a tremendous challenge for developing nano-preparations for efficiently treating RA across all phases. Activated neutrophils continuously infiltrate inflamed joints and exacerbate joint cartilage and bone damage by releasing neutrophil extracellular traps (NETs). Therefore, constructing a drug delivery system based on inhibiting NETs

Disrupting the Senescence-Associated Secretory Phenotype-M1Macrophage Feedback Loop in Synovitis Using Dual Nano-Switches To Restore Joint Homeostasis.

Osteoarthritis (OA), a leading cause of disability worldwide, impacts over 300 million people through progressive joint degeneration marked by chronic pain and functional impairment. A key driver of osteoarthritis progression is synovitis, characterized by inflamed synovial tissue harboring senescent fibroblasts and pro-inflammatory macrophages. These senescent cells secrete senescence-associated secretory phenotype (SASP) components, includining cytokines and proteases, which drive macrophage polarization toward a pro-inflammatory M1 state. Simultaneously, M1 macrophages release reactive oxyg
