GPR15-guided CD8+ T regulatory cells control intestinal inflammation.

Inflammatory bowel disease (IBD) causes chronic suffering from gastrointestinal inflammation and dysfunction that can progress to colon cancer1,2. The prevalence of the disease is increasing, and there is an urgent need to better understand its pathogenic mechanisms to improve treatment. We show that GPR15-a G-protein-coupled receptor expressed in immune cells and described previously as an entry co-factor for human and simian immunodeficiency viruses3-is a marker and homing receptor for a subset of intramucosal GPR15-guided regulatory CD8+ T lymphocytes (CD8+ TIGR cells). Deleterious GPR15 ge

NIR-II-Trackable LYTACs Phyto-Nanotheranostics for Source-Microenvironment Dual-Track ROS Regulation in Acute Gouty Arthritis.

The acute gouty arthritis (GA) is critically driven by the NOX2-mediated reactive oxygen species (ROS) explosion in macrophages, while current clinical drugs lack precise regulatory capability. Dealing with the challenge, a novel nanomaterial named TPR-LYTACs has been developed in this study. The TPR-LYTACs construction involves the assembly of a nano-core (TPR) from tea polyphenols and acidified rutin. The antioxidant TPR core is conjugated via modular assembly with a NOX2-targeting antibody and maleylated bovine serum albumin for source inhibition, enabling dual-pathway ROS regulation. In th

Bridging dermatology and imaging: Interpreting nail ultrasound in pediatric psoriasis.

Nail involvement in pediatric psoriasis is a clinically significant yet often underrecognized feature associated with increased disease severity and risk of psoriatic arthritis. In this Letter to the Editor, I comment on the study by Tavares et al., which evaluates high-frequency ultrasound findings, including Power Doppler activity and structural alterations, in pediatric nail psoriasis. The study provides valuable insight into the potential role of imaging in detecting subclinical nail involvement through systematic correlation of clinical and sonographic findings. From a dermatologic perspe

Factors associated with elevated serum IgG4 in rheumatic diseases: A retrospective single-center observational study.

Elevated serum immunoglobulin G4 (IgG4) levels have been reported in various rheumatic diseases; however, the factors associated with IgG4 elevation remain incompletely understood. This study aimed to evaluate the frequency of elevated serum IgG4 in different rheumatic diseases and to explore its association with disease classification based on typical inflammatory characteristics and autoantibody status. This retrospective single-center study included 255 patients with rheumatic diseases who underwent serum IgG4 testing between January 2017 and December 2022. Patients were categorized for exp

Trigger finger and the risk of systemic inflammatory and autoimmune rheumatic diseases: real-world evidence.

BACKGROUND: Trigger finger (TF) is a common hand disorder traditionally viewed as a localized mechanical condition. Recent evidences suggests potential links between tenosynovial pathology and systemic inflammatory diseases, especially rheumatoid arthritis, though large-scale longitudinal data remain limited. METHODS: We conducted a retrospective cohort study using the TriNetX Global Collaborative Network. Adults diagnosed with TF between 2005 and 2018 were identified and compared with individuals undergoing routine health evaluations without TF. Patients with prior malignancy, systemic rheum

Psoriatic arthritis patients have increased morbidity already at the time of diagnosis: a case-control study.

To evaluate the prevalence of comorbidities preceding the diagnosis of psoriatic arthritis (PsA) compared with the general population. We identified all adult patients granted a special reimbursement (SR) for disease-modifying anti-rheumatic drugs for PsA between 2002 and 2014 from the Finnish Social Insurance Institution register (N = 6077). Three general population controls were matched to each case. Visits to specialized medical care (Classification of Disease 10th revision coded), and prescription drug purchases categorized by Anatomical Therapeutic Chemical codes were collected for cases

Engineering technology of implantable galvanic cells for silencing the bone-neural axis in gouty arthritis therapy.

Gouty arthritis faces multiple challenges, such as recalcitrance to cure and recurrent flares, owing to its intricate pathophysiological microenvironment. Recent studies have focused on urate reduction, pain relief, or anti-inflammation effects alone, which often fail to provide satisfactory results. To address these challenges, an implantable manganese platinum microscale galvanic cell (Mn@Pt micro-GC) is developed. This system utilizes the urate oxidase-like activity of platinum to degrade deposited monosodium urate crystals, while the Mn2+ and hydrogen gas work synergistically to effectivel

Multiplexed SERS Profiling of Gout Biomarkers in Sweat via Polymer-Mediated Ag Nanoflakes.

Personalized management of gout requires profiling multiple metabolites over time. Sweat offers a noninvasive matrix for such profiling. However, simultaneous detection of trace metabolic precursors amidst a complex chemical background remains a major analytical challenge. In this study, a flexible surface-enhanced Raman spectroscopy (SERS) substrate is developed for the multiplexed monitoring of a gout biomarker panel, including uric acid (UA), xanthine, hypoxanthine, and creatinine. The sensor is fabricated via in situ droplet-grown Ag nanostructures on microwell-patterned polydimethylsiloxa

AI-guided CRISPR screening reveals therapeutic targets in psoriasis.

Psoriasis affects over 125 million people globally. Biologics targeting the IL-17/IL-17RA axis are effective but require systemic administration, are costly, and are unsuitable for some patients. Developing topical small-molecule alternatives requires a better understanding of how IL-17 receptor A (IL17RA) is regulated in keratinocytes, the principal effector cells of psoriatic lesions. Here, we report a genome-wide CRISPR knockout screen for regulators of surface IL17RA in primary human epidermal keratinocytes. We prioritize hits using experimental enrichment together with VirtualCRISPR, a la

Pathogenesis of autoantibodies in inflammatory myopathies.

PURPOSE OF REVIEW: While autoantibodies targeting extracellular antigens, such as those in myasthenia gravis, are established mediators of disease, autoantibodies recognizing intracellular antigens in inflammatory myopathies have traditionally been considered nonpathogenic. This review examines evidence challenging that paradigm and highlights recent advances in understanding the origins and pathogenic functions of autoantibodies in myositis. RECENT FINDINGS: In several forms of myositis, immunoglobulin accumulates within muscle fibers in a distribution mirroring that of the cognate autoantig

Potential of a bacterial immunomodulatory enzyme to restore health in a Th1 cytokine cocktail induced skin psoriasis-like model.

Psoriasis is a chronic inflammatory disorder driven by immune dysregulation. A common characteristic is hyperproliferation and aberrant differentiation of keratinocytes. Th1-type cytokines, containing TNFα, IL-1α and IL-6, play a role in various cellular processes such as the activation of dendritic cells, antigen processing and presentation, cell adhesion and migration, as well as cytokine and chemokine production. They are believed to be key contributors to the pathogenesis of psoriasis. In this work, healthy human epidermis equivalents (HEEs) were developed using neonatal human epidermal ke

Association between systemic oxidative stress imbalance and inflammatory markers in gouty patients: a cross-sectional case-control study in Yemen.

BACKGROUND: Gout is increasingly recognized as a systemic metabolic and inflammatory disorder. The urate paradox-where uric acid shifts from a plasma antioxidant to an intracellular pro-oxidant-remains a clinical challenge in managing systemic complications. OBJECTIVE: This study evaluates the association between systemic oxidative stress imbalance and high-sensitivity C-reactive protein (hs-CRP) as a marker of inflammatory burden in gouty patients. METHODS: A case-control study was conducted on 200 participants (100 gouty patients and 100 healthy controls) in Yemen. Biomarkers including mal
