Establishment and translational evaluation of animal models for skin wound healing: a systematic review.

BACKGROUND: Skin wounds, encompassing acute injuries and chronic refractory ulcers, impose substantial physical and economic burdens globally. While animal models are indispensable for dissecting wound healing pathophysiology and testing therapeutic interventions, the discordance between preclinical findings and clinical outcomes remains a critical challenge. METHODS: To provide a standardized reference for model selection, we conducted a systematic review in accordance with PRISMA guidelines. We comprehensively searched PubMed, Web of Science, and Scopus for studies published between January

Clinical characteristics and risk factors for calcinosis cutis in systemic sclerosis: insights from the EUSTAR database.

OBJECTIVES: Calcinosis Cutis (CaC) represents one of the most frequent and disabling non-lethal manifestations in SSc. Our objectives were to evaluate (1) associations of CaC with SSc clinical characteristics and (2) identify risk factors for CaC development. METHODS: EUSTAR database-registered SSc patients with available information on their CaC status were included. We compared baseline patient characteristics (with vs without CaC at baseline) and investigated predictors of CaC development at 5 and 10 years (in those without baseline CaC) with logistic regression analyses. RESULTS: A total

Subungual melanoma of the fifth digit: A case report of a rare cutaneous malignancy documented in Syria.

RATIONALE: Subungual melanoma is the rarest subtype of cutaneous melanoma, accounting for approximately 1.9% of cases. Involvement of the fifth digit is exceptionally uncommon, often contributing to delayed diagnosis and poor outcomes. PATIENT CONCERNS: We report a case involving a 45-year-old male patient who presented with longitudinal melanonychia, a positive Hutchinson's sign, and ulceration of the nail plate on the right fifth finger. DIAGNOSES: Histopathological analysis confirmed subungual melanoma with a Breslow thickness of 4.3 mm. INTERVENTIONS: The patient underwent surgical ampu

From blood to bowel: correlating serological markers with mucosal healing and inflammation in patients with inflammatory bowel disease.

BACKGROUND: Inflammatory bowel disease (IBD) is a systemic disease characterized by chronic inflammation of the gastrointestinal tract. Serological testing has the advantages of wide sources and easy detection. OBJECTIVE: To explore the correlation between the serological markers and endoscopic mucosal healing and histological healing. We also explored the predictive value of individual and combined detection on the disease activity of ulcerative colitis (UC) and Crohn's disease (CD) and provide reference for clinical non-invasive evaluation. METHODS: We retrospectively analyzed 267 IBD pati

Plasma metabolite octadecadienedioate: A novel protective factor against diabetes-related Charcot foot.

Charcot foot (CF) is a severe, progressive neuro-osteoarthropathy predominantly affecting diabetic patients with peripheral neuropathy. Characterized by progressive bone and joint destruction, CF can lead to debilitating foot deformities, ulceration, infection, and ultimately lower extremity amputation. Despite its significant clinical impact, the global prevalence remains uncertain, with an estimated 1.6 million people affected worldwide. Early diagnosis is challenging due to the insidious onset and limited specificity of current clinical, imaging, and laboratory assessments. The role of circ

Exploring the mechanism underlying negative pressure wound therapy in promoting the healing of diabetic foot ulcers via integrated multi-omics and bioinformatics approaches.

BACKGROUND: Diabetic foot ulcers (DFU) are one of the most serious complications of diabetes, closely associated with high amputation rates, mortality, and healthcare burdens, and the clinical treatment research is still limited. Negative Pressure Wound Therapy (NPWT) is a treatment modality for DFU, but its specific mechanisms promoting wound healing remain largely unknown. METHODS: The bulk RNA-seq and scRNA-seq data for DFU used in this study were obtained from the GEO database. First, this study used differential analysis and multiple machine learning algorithms to identify key genes acti

Remote periosteal distraction promotes ischemic diabetic foot wound repair associated with VEGF/VEGFR2-related angiogenic responses.

Bone transport-induced remote microinjury may promote diabetic foot ulcer healing but requires osteotomy and may cause fracture or osteomyelitis. This study evaluated whether osteotomy-free remote periosteal distraction (RPD) could promote repair in a rabbit ischemic diabetic foot wound model. Seventy-two diabetic male rabbits were randomized into RPD, Sham, and Control groups, and ischemic foot wounds were created. In the RPD group, a custom-made plate was implanted beneath the tibial periosteum and connected to an external distractor. The periosteum was distracted outward for 7 days and retu

Fatal intoxication caused by conventional-dose methotrexate in an elderly patient with end-stage renal disease: A cautionary case report.

RATIONALE: Methotrexate (MTX) is a first-line agent for rheumatoid arthritis, and impaired renal excretion drastically elevates the risk of fatal drug accumulation. End-stage renal disease (ESRD) patients face an extreme toxicity risk even under conventional low-dose MTX, yet related fatal cases remain underreported. This case highlights critical clinical warnings for rheumatologists and emergency physicians. PATIENT CONCERNS: A 71-year-old male with a 5-year history of rheumatoid arthritis and 1-year untreated chronic kidney disease stage 5 received weekly 10 mg subcutaneous MTX for 3 consec

Incidence of diabetes-related foot disease: results from 2 prospective cohort studies and meta-analysis.

We aimed to estimate the incidence for overall and individual components of diabetes-related foot disease (DFD), and explore potential heterogeneity across studies. We included patients with diabetes who were free of DFD from UK Biobank and Chongqing Diabetes Registry (CDR). We defined DFD according to the International Working Group on the Diabetic Foot 2023 criteria. We further performed meta-analyses by integrating results from the two cohorts and 64 cohorts identified from a systematic review of literature, and quantifying the extent of variation in reported incidence rates. The incidence

Transient receptor potential vanilloid channels in inflammatory bowel diseases: linking inflammation, angiogenesis, and oxidative stress.

Inflammatory bowel disease (IBD), including Crohn's disease (CD) and ulcerative colitis (UC), is characterized by chronic relapsing intestinal inflammation driven by complex interactions among immune dysregulation, epithelial barrier disruption, vascular dysfunction, oxidative stress, and gut microbiota imbalance. Despite advances in biologic and small-molecule therapies, a substantial proportion of patients remain refractory to treatment or experience significant adverse effects, underscoring the need for novel mechanistic insights and therapeutic targets. Emerging evidence suggests that the

Unmasking Behçet's disease in the European population: a case series report.

Behçet's disease (BD) is rare in European populations. Because clinical and immunological characteristics may differ between Caucasians and Middle Eastern patients, we analyzed BD patients from the Polish population. Data from eight patients with BD treated at a rheumatology clinic were analyzed and collected, including demographic data, clinical features, laboratory test results, comorbidities, and treatment options. The study included eight patients (mean age 38.2 ± 13.7 years; 75% female). None of the patients had a family history of the disease. The mean delay in diagnosis was 5.28 ± 4.6 y

Integrated mechanical and metabolic limb salvage for advanced diabetic Charcot hindfoot collapse with non-traumatic calcaneal fracture: an 8-year follow-up case report.

BACKGROUND: Advanced Charcot neuro-osteoarthropathy involving the hindfoot is associated with severe deformity, instability, ulceration, infection, and major lower-limb amputation. Non-traumatic calcaneal fracture associated with diabetic Charcot hindfoot collapse is uncommon and difficult to treat because of neuropathy, impaired bone quality, tendon imbalance, soft tissue compromise, and a high risk of fixation failure. Reports describing long-term functional limb preservation in patients with advanced Charcot hindfoot collapse associated with severe metabolic bone impairment remain limited.

Influence of socioeconomic development on incidence and disease phenotype in inflammatory bowel disease: a population-based incident study across 16 regions.

INTRODUCTION: The global rise in inflammatory bowel disease (IBD) parallels socio-economic development, but its influence on disease epidemiology and phenotype remains unclear. We aimed to characterise IBD incidence, clinical features, and associations with socio-economic indices across newly industrialised countries. METHODS: This prospective population-based study spanned 16 regions in Africa, Asia, Latin America, and the Middle East from October 2021 for one year. Poisson regression estimated incidence rate ratios (IRR) for Human Development Index (HDI), Socio-demographic Index (SDI), subn

A case of rheumatoid neutrophilic dermatosis complicated by neutrophilic tubulointerstitial nephritis.

As an extra-articular manifestation of rheumatoid arthritis (RA), neutrophilic dermatosis may occur. Here, we report a rare case of RA complicated by both neutrophilic dermatosis and neutrophilic tubulointerstitial nephritis. In addition to polyarthritis, the patient exhibited purpura and erythema with haemorrhagic crusts and ulceration accompanied by marked neutrophilic infiltration, extending from the fingers to the forearms and from both lower legs to the toes, as well as interstitial lung disease, leading to a diagnosis of RA. After the initiation of tocilizumab, the arthritis improved. Ho
