COVID-19, caused by the SARS-CoV-2 virus, manifests with varying degrees of severity, affecting individuals worldwide. The spectrum of symptoms ranges from mild to severe, with respiratory failure being a leading cause of death. Immunological factors, particularly excessive cytokine production such as IL-18 and IL-1β, significantly contribute to disease severity. In this context, the NLRP3 inflammasome, a key component of the innate immune system, has influenced COVID-19 outcomes. This study investigated the association between single-nucleotide variants (SNVs) in the NLRP3 inflammasome and CO

Intensive rearing of farmed fish may risk disease spillover into free-living populations. This study concerns the blood-feeding gill monogenean of salmonids, Discocotyle sagitatta, on the Isle of Man, UK. Heavy infections in 2 fish farms have led to severe disease with periodic mass mortality. Infection levels in rainbow trout (Oncorhynchus mykiss, overall n = 556) increased with age (i.e. years exposed): by year 3, prevalence was 100%, mean intensity c. 100 (maximum 1150) worms/host. Output from farms of many millions of parasite eggs/day has the potential for transmission to downstream popul

INTRODUCTION: Pythium insidiosum keratitis (PIK) is a rapidly progressive, aggressive corneal infection that closely mimics fungal keratitis but fails to respond to conventional antifungal therapy. Misdiagnosis and delayed intervention frequently result in poor outcomes, including high rates of therapeutic keratoplasty and irreversible vision loss. Timely and accurate identification is therefore vital, yet conventional microbiological methods are slow, and histopathology is invasive and often inconclusive. Recent advances in imaging, molecular diagnostics, and biosensor technology are revoluti

The diagnosis of active toxoplasmosis in fetus, newborns, and immunocompromised patients relies on molecular detection of the parasite DNA in various samples according to the clinical setting. The quality of molecular diagnosis is essential to guide specific treatment. Today, real-time PCR (qPCR) based on the REP-529 target is the reference method, but given the emergence of droplet digital PCR (ddPCR) in other diagnostic fields, it is essential to evaluate its clinical performance for the diagnosis of toxoplasmosis. In this study, we evaluated the Naica Crystal ddPCR system (Stilla Technologi

INTRODUCTION: Malaria remains a major health challenge, with increasing resistance to frontline chemotherapy. Recent cheminformatics studies have revealed that potent antiplasmodials occupy a distinct antimalarial chemical space (AMCS), defined by specific property cutoffs.
AREAS COVERED: We show that only ~ 6-8% of compounds in the representative commercial libraries are AMCS-compliant, emphasizing the need for pre-filtering to improve their suitability for phenotypic screening. Most clinically used antimalarials and several clinically used oral drugs repurposed for antimalarial activity com

INTRODUCTION: Anisakiasis is a zoonotic parasitic disease transmitted through consumption of fish, with increasing global cases. While thorough cooking kills Anisakis larvae, thermo-stable allergens can still cause allergic reactions. Traditional Mediterranean seafood methods such as marinating, salting, and brining, which lack heat treatment, pose a high risk of anisakiasis.
MATERIALS AND METHODS: This study investigated the antilarval effects of nanoemulsions made from thyme, garlic, and grape seed essential oils combined with brines on Anisakis L3 larvae in vitro.
RESULTS: Larvae were col

Malaria remains a critical global health challenge, with increasing resistance to frontline therapies necessitating novel drug targets. The proteasome has emerged as a promising target for antimalarial drug discovery. This study describes efforts to optimize a series of species-selective reversible inhibitors targeting the Plasmodium falciparum 20S proteasome. Starting from the carboxypiperidine scaffold identified through a high-throughput viability screen, we conducted iterative structure-activity relationship studies, leading to the development of highly potent and selective inhibitors with

Background/Objectives: Malaria remains the most critical parasitic disease globally, responsible for over 600.000 deaths annually. In sub-Saharan Africa, co-infections of Plasmodium falciparum with other pathogens, particularly Staphylococcus aureus, are common in children with severe malaria. Therefore, the design of new compounds targeting both pathogens appears to be an urgent priority. Methods: A small series of hybrid compounds was designed and synthesized by linking the pharmacophore of the antimalarial drug chloroquine with the phenothiazine core. These compounds were tested in vitro ag

Diketopiperazines (DKPs) are biologically important cyclic dipeptides widespread in nature, associated primarily with microorganisms. This is the case for the 2,5-DKP derivative cyclo(L-Leu-L-Pro) (cLP), also known as gancidin W or PPDHMP, identified from a variety of bacteria and fungi, and occasionally found in food products. The present review retraces the discovery of cLP, its identification in living species, its chemical syntheses, and its biochemical properties. In bacteria, cLP is often associated with other DKPs to serve as a defense element against other microorganisms and/or as a re

Candida parapsilosis has emerged as a prominent cause of nosocomial candidemia, particularly among critically ill patients. The increasing prevalence of fluconazole-resistant C. parapsilosis (FR-Cp) poses major therapeutic challenges, especially in resource-limited settings. We conducted a retrospective study of 144 patients with C. parapsilosis candidemia admitted to two post-surgical ICUs at a Brazilian tertiary cardiothoracic hospital between 2016 and August 2024. Demographic, clinical, microbiological, and therapeutic data were analyzed. Predictors of 30-day mortality were identified throu

The cereal cyst nematode, Heterodera avena, is responsible for substantial economic losses in the global production of wheat, barley, and other cereal crops. Extracellular enzymes, particularly those from the glycoside hydrolase 18 (GH18) family, such as chitinases secreted by Trichoderma spp., play a crucial role in nematode control. However, the genome-wide analysis of Trichoderma longibrachiatum T6 (T6) GH18 family genes in controlling of H. avenae remains unexplored. Through phylogenetic analysis and bioinformatics tools, we identified and conducted a detailed analysis of 18 GH18 genes dis

Leishmaniasis is a parasitic disease caused by Leishmania spp., transmitted by sandflies, and endemic in 98 countries. Leishmania infantum, the main agent of visceral leishmaniasis in Europe, commonly infects both humans and animals, with dogs as the principal domestic reservoir. Clinical manifestations in dogs depend on the host immune response. A robust Th1 response facilitates macrophage activation and parasite control, while persistently elevated TNF-α and IL-6 can lead to chronic inflammation and tissue damage. Current treatments reduce parasite load but rarely achieve complete cure and a

BACKGROUND: Migration phenomena from low- to high-income countries have been constantly increasing in the past years, and migrants living with HIV (MLHIVs) currently represent a non-negligible proportion of the population living with HIV in the European setting. When taken into care, MLHIVs should be screened for tropical diseases capable of asymptomatically persisting for years and leading to severe and/or chronic complications, especially in immunocompromised populations.
METHODS: We conducted a retrospective study aimed at analyzing the screening ratios and seroprevalences of strongyloidia
