The immunological effects of lipopolysaccharides (LPSs) from gut microbiota remain poorly explored, overshadowed by the longstanding view of LPS as a prototypical pro-inflammatory molecule. Herein, we report the first comprehensive chemical and immunological characterization of LPS from Segatella copri DSM 18205, a prominent member of the human oral and intestinal microbiota. This LPS features a unique chemical architecture, including a mannose- and glucose-rich oligosaccharide (OS) and a highly heterogeneous, hypo-acylated lipid A domain, as elucidated by advanced mass spectrometry (MS) and n

Extended-spectrum β-lactamase-producing Escherichia coli (ESBL-Ec) and Klebsiella pneumoniae (ESBL-Kp) are among the leading causes of hospital-acquired infections globally. Fluoroquinolone-resistant ESBL-Ec/Kp infections have limited therapeutic options. This study investigated the prevalence and antibiotic resistance patterns of ESBL-Ec/Kp isolated from hospitalized patients, hospital environments, and wastewaters in Yaoundé, Cameroon. It further characterized ESBL genes and clonal relatedness in all ESBL-Ec/Kp isolates and assessed plasmid-mediated quinolone resistance (PMQR) genes specific

Anorexia nervosa (AN) is a psychiatric disease of unknown etiology. Recently, caseinolytic protease B (ClpB), produced by Enterobacteriaceae, was identified as an antigen mimetic of α-melanocyte-stimulating hormone (α-MSH), an anorexigenic neuropeptide. Here, we studied the role of ClpB in the development of activity-based anorexia (ABA), a rodent model of AN. In two series of ClpB immunization experiments, we show that when ClpB was lowered by antibodies, mice were protected against ABA, showing lean mass preservation, an increase in food intake, and reduced feeding anticipatory activity. OBJECTIVES: The rapid dissemination of carbapenem-resistant Enterobacteriaceae (CRE) poses a severe global health threat due to limited treatment options. Polymyxin is currently considered a last-resort therapy for human infections caused by CRE. The increasing clinical use of polymyxin has resulted in alarming resistance rates. This study investigated the clinical characteristics, molecular epidemiological characteristics, and resistance mechanisms of polymyxin-resistant CRE (PR-CRE) in Chongqing, China.
METHODS: Antimicrobial susceptibility and resistance genes were analyzed by PCR-based am

Although mechanical tension from luminal distension is a primary regulator of gut motility, we reveal a parallel chemosensory pathway wherein long-chain unsaturated free fatty acids (LUFFAs) from dietary or enterobacterial sources directly modulate gastrointestinal motor function. Using ex vivo and in vivo contractility assays in human and murine intestinal tissues, we found that LUFFAs, particularly Omega-3 fatty acids, suppressed spontaneous contractions and delayed intestinal transit in a double bond-dependent manner. Mechanistically, selective activation of free fatty acid receptor 1/4 (FF

Sodium butyrate (NaB), a major intestinal metabolite, has been suggested to protect against osteoporosis (OP). This study aimed to elucidate the mechanism by which NaB regulates ferroptosis in OP. An ovariectomy-induced mouse OP model was established, and treated with NaB or the ferroptosis inhibitor Fer-1. Bone mineral density, bone microstructure, bone formation and resorption, and ferroptosis markers were assessed. In vitro, mouse bone marrow mesenchymal stem cells (BMSCs) were treated with NaB and the ferroptosis inducer Erastin to evaluate osteogenic differentiation and ferroptosis. Phosp

Plasmids carrying the mobilized colistin-resistance gene mcr-1 are prevalent among multidrug-resistant Gram-negative pathogens, yet their broad impact on bacterial physiology and virulence remains unclear. Here, we demonstrate that acquisition of an mcr-1 plasmid concurrently increases antimicrobial resistance and pathogenicity in Escherichia coli. On the same plasmid, the XRE-family transcriptional regulator EcaR cooperates with MCR-1 to activate the wec operon, driving biosynthesis of two surface polysaccharides: enterobacterial common antigen (ECA) and a high-molecular-weight O-chain. Acute oak decline (AOD) is a rapidly progressing disease affecting various oak species (Quercus spp.). Recent studies have shown that AOD is associated with a consortium of Gram-negative, facultatively anaerobic bacteria (e.g., in Enterobacterales) in the United Kingdom and continental Europe. However, there is limited information on the bacterial contributions and key genera associated with oak diseases and broadleaf forest ecosystems in Nordic countries. The primary objective of this brief study was to collect the first data on the bark microbiomes of symptomatic, declining sessile oaks (Q.

α-melanocyte-stimulating hormone (α-MSH) is an anorexigenic peptide. In this exploratory study, a targeted proteomic approach was used to detect α-MSH-like bacterial proteins in the gut microbiota of patients with anorexia nervosa (AN) and healthy controls. Enterobacteriaceae chaperone DnaK was identified as an α-MSH-like protein common to both study groups, while caseinolytic protease B (ClpB) was present in patients with AN. These data further link anorexigenic ClpB to the AN pathophysiology.

In this study, we isolated 214 Vibrio cholerae strains from aquatic (shrimp, crab, grass carp, and crucian carp) and their cultured environment in Shanghai, China. The virulence, serotype, and antimicrobial susceptibility were tested, and polymerase chain reaction (PCR) was used to detect antimicrobial resistance genes. Enterobacterial repetitive intergenic consensus polymerase chain reaction (ERIC-PCR) was employed for cluster analysis of the isolated strains. The results showed that V. cholerae was found in 47.9% (114/238) of aquatic samples, with the highest detection rate in shrimp (81.1%)

Bacteriophage therapy is a promising approach for targeting antibiotic-resistant bacteria and modulating gut microbiota in metabolic diseases such as obesity. This study evaluated the impact of a two-phage cocktail on an ex vivo colonic simulation model of gut microbiota derived from obese individuals, both in its normalized state and after enrichment with Enterobacter cloacae, an obesity-related bacteria. Microbiological analyses confirmed that the phage cocktail remained active throughout the colonic regions over three digestion cycles and effectively reduced enterobacterial populations in t
