Single-cell and spatial transcriptomic profiling of cardiac fibroblasts following myocardial infarction.

Cardiac fibroblasts (CFs) are key mediators of heart repair following myocardial infarction (MI). A specific CF subpopulation, termed Reparative Cardiac Fibroblasts (RCFs), has been shown to orchestrate scar formation and prevent ventricular rupture after MI. However, the timing of RCF appearance and the molecular events underlying this transition remain largely undefined. Here, we present a multi-modal dataset capturing the transcriptional dynamics of CFs during the early phase post-MI. Our integrative dataset combines bulk RNA sequencing, RNAscope in situ hybridization, and spatial transcrip

Integrated motion-corrected extracellular volume fraction mapping reveals subtle extracellular remodeling in remote myocardium following chronic myocardial infarction.

BACKGROUND: Emerging evidence indicates that diffuse fibrosis in remote myocardium contributes to post-infarction ventricular remodeling, yet conventional late gadolinium enhancement (LGE) lacks sensitivity for extracellular matrix (ECM) quantification. This study establishes a semi-automated, motion-corrected extracellular volume fraction (ECV) mapping protocol to noninvasively characterize ECM expansion in non-infarcted myocardium after chronic myocardial infarction (CMI), addressing critical gaps in early detection of subclinical remodeling. MATERIALS: This prospective dual-center study enr

Neuroprotective potential of phloroglucinol in focal cerebral ischemia in rats: a mechanistic study.

Ischemic stroke remains a leading cause of mortality and disability worldwide, with limited therapeutic options that effectively target its complex pathophysiology. This study evaluates the neuroprotective potential of phloroglucinol (PH), a polyphenolic compound with known antioxidant and anti-inflammatory properties, in a rat model of focal cerebral ischemia induced by middle cerebral artery occlusion (MCAO). Animals were randomly assigned to different groups. Phloroglucinol was administered intraperitoneally at doses of 15 and 30 mg/kg, beginning immediately after a 1-hour MCAO injury and c

Site-Specific Blockade of LPA-TRPV1 Interaction at K710 Residue Influences on Myocardial Ischemia/Reperfusion Injury in Ex Vivo Mouse Hearts.

BACKGROUND: Myocardial ischemia/reperfusion (I/R) injury commonly occurs in patients undergoing cardiac or noncardiac surgeries, increasing perioperative mortality risk. Although numerous endogenous mediators released during I/R contribute to myocardial damage, their mechanisms require further elucidation. We investigated whether lysophosphatidic acid (LPA), a bioactive phospholipid, mediates myocardial I/R injury by interacting with cardiac transient receptor potential vanilloid 1 (TRPV1). METHODS: A TRPV1 K710N knock-in mouse model was generated by CRISPR/Cas9, introducing a point mutation

Preventive and therapeutic effects of ginsenosides on myocardial ischemia-reperfusion injury in animal models: a systematic review and meta-analysis.

BACKGROUND: Myocardial ischemia-reperfusion injury (MIRI) markedly impairs cardiac functional recovery and represents a major determinant of adverse outcomes in patients with ischemic heart disease. Ginsenosides, the principal bioactive constituents of ginseng, exert significant cardioprotection against MIRI. This review systematically summarizes and analyzes in vivo (animal) studies to clarify the efficacy and underlying mechanisms of ginsenosides in MIRI. METHODS: The PubMed, EMbase, Web of Science, Cochrane Library, CNKI, WanFang, and Cqvip databases were systematically searched from incep

Grhl3 Downregulation Facilitates ECM Adaptation for Fibroblast to iCM Commitment.

BACKGROUND: Direct cardiac reprogramming offers a promising therapeutic strategy for heart regeneration by converting endogenous fibroblasts to functional induced cardiomyocytes (iCMs) that integrate into the myocardium to restore heart structure and function. While ECM (extracellular matrix) plays critical roles in cardiac disease and repair, the dynamic changes and transcriptional regulation underlying ECM remodeling during reprogramming remain poorly understood. METHODS: We investigated ECM dynamics during iCM reprogramming using integrated transcriptomic, proteomic, and epigenetic analyse

Drp1-mediated mitochondrial fission protects macrophages from mtDNA/ZBP1-mediated inflammation and inhibits post-infarct cardiac remodelling.

AIMS: Ischaemic heart disease is a leading cause of death worldwide, and heart failure after myocardial infarction (MI) is a growing issue in an ageing society. Macrophages play a central role in left ventricular (LV) remodelling after MI. Mitochondria consistently change their morphology, including fission and fusion; however, the role of these morphological changes, particularly in macrophages, remains unknown. This study investigated the role of dynamin-related protein 1 (Drp1), a key mediator of mitochondrial fission, in macrophages and its involvement in the mechanisms of left ventricular

Single-nucleus analysis reveals Dengzhan Shengmai capsule prevents cardiac fibrosis after myocardial infarction in rat by targeting Ltbp2+ fibroblast subset.

BACKGROUND: Cardiac fibrosis is a distinguishing hallmark of ischemic heart diseases with limited therapeutic options due to the incomplete comprehension of cellular heterogeneity. Our previous research demonstrated that Dengzhan Shengmai capsule (DZSMC) could significantly attenuate myocardial fibrosis, yet its cell-type-specific targets remain elusive. OBJECTIVES: This work aimed to investigate the mechanisms of DZSMC on cardiac fibrosis in post-myocardial infarction (MI) rats at single-cell resolution. METHODS: MI rats were produced by ligating the left anterior descending artery. DZSMC (

Qishen Yiqi Dropping Pills ameliorate acute myocardial infarction by modulating PKM2/STAT3 pathway-mediated metabolic reprogramming in macrophages.

BACKGROUND AND PURPOSE: Acute myocardial infarction (AMI) is the leading cause of cardiovascular mortality worldwide. Timely reperfusion therapy is crucial to save the ischemic myocardium and prevent infarct expansion. Inflammation, a central pathological mechanism in AMI, exacerbates myocardial tissue damage by triggering immune activation in a cascading manner. Macrophage metabolic reprogramming plays a crucial role in regulating inflammatory phenotypes in this process. Qishen Yiqi Dropping Pills (QSYQ), a traditional Chinese medicine compound, greatly improves the cardiac inflammatory micro

Mitochondrial Intercellular Transfer via Platelets After Physical Training Exerts Neuro-Glial Protection Against Cerebral Ischemia.

While thrombolytic therapy can be effective for stroke, many patients are unable to benefit due to time restrictions. In an aging society, sarcopenia, a condition marked by reduced muscle volume, often worsens recovery after stroke. Our study explored how mitochondria, which are abundant in muscle, could aid in stroke recovery through exercise-induced migration. Using mouse models of chronic hypoperfusion and ischemia, alongside in vitro studies with rat primary cells under oxygen-glucose deprivation and CoCl2 exposure, we found that treadmill exercise protected against white matter injury, my

Maresin 1 ameliorates myocardial ischaemia‒reperfusion injury by promoting tissue resident macrophage efferocytosis.

AIMS: Myocardial ischaemia‒reperfusion (I/R) injury triggers a robust inflammatory storm cascade that critically compromises reperfusion efficacy following acute myocardial infarction. Enhanced efferocytosis by cardiac resident macrophages (RMs) has therapeutic potential for inflammation resolution. The unsaturated long-chain fatty acid Maresin1 (MaR1) exhibits potent anti-inflammatory properties that is devoid of immunosuppressive effects. However, its therapeutic potential in myocardial I/R injury and regulatory mechanisms in cardiac RMs remains unexplored. METHODS AND RESULTS: A clinical c

Zinc finger BED-type containing 6 (ZBED6) ameliorates cardiac fibrosis by inhibiting Piezo1 transcription and YAP nuclear translocation.

Cardiac fibroblasts progressively replace deceased cardiomyocytes during the development of myocardial fibrosis, an irreversible pathological repair process that ultimately leads to cardiac dysfunction and heart failure. Cardiac injury was evaluated by echocardiography and Masson staining in myocardial  infarction (MI) mice with zinc finger BED-type containing 6 (ZBED6) knockdown or overexpression. Furthermore, chromatin immunoprecipitation (ChIP) assays, electrophoretic mobility shift assays (EMSAs), and luciferase reporter assays were used to explore the target of ZBED6. ZBED6 expression was

Apoptotic bodies derived from human umbilical cord mesenchymal stem cells improve recovery from myocardial infarction in swine.

Apoptotic bodies (ABs) are a type of extracellular vesicles (EVs) that could contribute to the paracrine effect of stem cells. However, their potential in treating cardiovascular diseases is largely unexplored. This study investigated the therapeutic effects of ABs derived from human umbilical cord mesenchymal stem cells (MSCs) on cardiac recovery in a porcine model of myocardial infarction (MI). In vitro, ABs reduced apoptosis and cytotoxicity in cardiomyocytes under oxygen and glucose deprivation (OGD) conditions and enhanced the capacity of migration and tube formation in endothelial cells.

Cardioprotective effects of notoginsenoside R1 on myocardial remodeling and cardiac function in restraint stress-induced MI in rats.

Myocardial infarction (MI) is one of the leading causes of death globally, and stress-aggravated cardiac injury exacerbates patient prognosis. The study aimed to clarify the role of notoginsenoside R1 (NR) in the myocardial infarction-induced differential gene 2 (CRIF1)/PINK1-Parkin complex by using a restrain stress-exacerbated MI rat model. Male Sprague Dawley rats were subjected to coronary artery ligation followed by treatment with NR (2.5, 5, or 10 mg/kg, p.o.) for 10 days post-surgery. NR significantly enhanced cardiac function (elevated LVEF (left ventricular ejection fraction), LVFS (l

A Brief Description of the Cellular Mechanisms Involved in Cardiac Chemical Hypoxia.

Hypoxia is involved in severe cardiac conditions such as heart failure and myocardial infarction. Therefore, understanding the molecular mechanisms underlying hypoxia is crucial for developing effective therapeutic strategies. Specifically, one in vitro hypoxia model involves reducing oxygen concentration, thereby emulating many features observed in cardiac ischemia in rodents and patients. One way to overcome the infrastructure challenges in basic research laboratories (e.g., anaerobic chamber) is to use chemically-induced hypoxia models. Cobalt chloride (CoCl2) treatment provides an inexpens

BACKGROUND: Myocardial ischemia-reperfusion injury(MIRI) is a significant contributor to poor prognosis after myocardial infarction. Ferroptosis, a novel form of regulated cell death, is involved in MIRI. However, the role of ferroptosis in the ischemia phase and reperfusion phase remains unclear, and key regulatory mechanisms still need further exploration. METHODS AND RESULTS: AC16 cardiomyocyte oxygen-glucose deprivation/reoxygenation (OGD/R) model and mice MIRI model were established to investigate the role of ferroptosis in ischemia phase and reperfusion phase, our study found that ferro

Dual-biomimetic Nanodecoys reprogram cardiac macrophages by suppressing STING signaling for heart repair.

Myocardial infarction (MI) initiates sterile inflammation through the release of cytosolic DNA from necrotic cardiomyocytes, which aberrantly activates the cGAS-STING pathway in infiltrating macrophages and drives their polarization toward a pro-inflammatory M1 phenotype. Although the immunosuppressive oligodeoxynucleotide A151 can antagonize cGAS activation, its therapeutic utility is limited by enzymatic instability and inefficient cellular delivery. Here, we report a dual-biomimetic nanodecoy (A151@APPL) that integrates platelet membrane vesicles for infarct-specific targeting with arginine
