Listening in Reverse: A Case for Retrograde Signaling During Auditory Inner Hair Cell Ribbon Synapse Maturation.

Cochlear inner hair cells (IHCs) conduct rapid, sustained, and temporally precise transmission of acoustic information onto spiral ganglion neurons (SGNs). For this purpose, IHCs utilize highly specialized sensory synapses. During development, these so-called 'ribbon' synapses and their postsynaptic counterparts undergo extensive structural and functional refinement-a process driven by pre-sensory spontaneous activity. The underlying molecular mechanisms remain incompletely understood to date. Interestingly, little is known about the involvement of retrograde feedback provided by postsynaptic

Time-dependent pore-network modeling of Ostwald ripening in porous media.

We present a time-dependent pore-network model that couples transient mass transfer in the aqueous phase, capillary pressure heterogeneity, and realistic pore-throat geometries to capture the dynamic evolution of gas clusters during Ostwald ripening in porous media. The model is applied to Bentheimer sandstone to study Ostwald ripening after imbibition to residual gas saturation. Both imbibition (shrinkage) and drainage (growth) events occur as the local capillary pressure in trapped gas clusters approaches equilibrium. The model tracks event statistics, capillary pressure equilibration, clust

Therapeutic extracellular vesicles isolated from mesenchymal stem cells: properties, isolation, characterization, and applications in optic neuropathies.

Extracellular vesicles (EVs) are promising cell-free therapeutic agents because of their low immunogenicity, excellent biocompatibility, and intrinsic capacity for intercellular communication. Of these, mesenchymal stem cell-derived EVs (MSC-EVs) have attracted considerable interest for their potential in treating optic neuropathies; however, their clinical translation remains limited by standardized manufacturing, quality evaluation, and mechanistic understanding. This review summarizes key advances in therapeutic EV development, including MSC culture, EV production, isolation, and characteri

Detection of the RNA-Editing Enzyme ADARB1 in Primary Auditory Neurons and Anti-ADARB1 Immunofluorescence as a Means for Automated Counting.

Spiral ganglion neurons (SGNs), the primary auditory afferents in the inner ear, are vulnerable to degeneration from neuropathies, concussion, and aging, among other causes. Accurate quantification of SGNs is critical for assessing degeneration and therapeutic interventions, but manual counting is arduous due to dense cellular crowding and unreliable nuclear markers. Because SGNs receive input from GluA2-containing AMPA receptors, the transcripts for which must be RNA-edited for calcium impermeability, we hypothesized that SGNs express high levels of the RNA-editing enzyme ADARB1. Here, we con

Synergistic Inhibition of Spinal TRPV1 Signaling by Combined Eye Acupuncture and Electroacupuncture Alleviates Inflammatory Pain in a Rat Model of Knee Osteoarthritis.

BACKGROUND: Chronic pain caused by knee osteoarthritis (KOA) remains inadequately managed by safe, non-pharmacological therapies. Although acupuncture is recommended for KOA pain by clinical guidelines, the efficacy of single-modality acupuncture remains modest, and the synergistic mechanisms of combining different acupuncture modalities are poorly understood. OBJECTIVE: To determine whether combining eye acupuncture (EAp) with electroacupuncture (EA) produces greater analgesia in a rat KOA model via regulation of TRPV1 signaling in the spinal dorsal horn (SDH) and dorsal root ganglia (DRG).

AKAP1 Protects Retinal Ganglion Cells and Preserves Vision by Improving Mitochondrial and Synaptic Health in Glaucomatous Neurodegeneration.

Glaucoma, a leading cause of irreversible blindness, is characterized by progressive loss of retinal ganglion cells (RGCs) and lacks effective neuroprotective therapies. Despite evidence implicating mitochondrial dysfunction in glaucoma, the mechanisms linking elevated intraocular pressure, synaptic dysfunction, mitochondrial failure, and RGC death remain incompletely understood. Here we identify the mitochondrial scaffold protein A-kinase anchoring protein 1 (AKAP1) as a critical regulator of RGC resilience in glaucoma. AKAP1 expression is reduced in glaucomatous retinas and correlates with d

Single-cell atlas of neuroma-induced DRG remodeling reveals intervention-specific glial and immune programs.

Peripheral nerve injury caused by cut, crush, or excessive stretch can lead to disordered regeneration and painful neuroma formation. Regrowing axons may terminate in a swollen, hypersensitive nerve stump, producing pain that can outweigh sensory or motor deficits. Despite the frequency of neuromas, treatment outcomes remain inconsistent, and the mechanisms that initiate and sustain neuroma-associated pain are poorly understood. A proximal nerve crush performed upstream of a neuroma or transection may reshape axonal growth and interrupt retrograde injury signaling, potentially promoting a rege

Psoas-lumbar vertebral index and short-term pain relief after pulsed radiofrequency treatment of the dorsal root ganglion in postherpetic neuralgia in Korea: a retrospective pilot study.

PURPOSE: The psoas-lumbar vertebral index (PLVI), a marker of central sarcopenia measured using computed tomography (CT) or magnetic resonance imaging (MRI), has been linked to the development of postherpetic neuralgia (PHN). Whether PLVI predicts short-term pain relief after pulsed radiofrequency (pRF) treatment of the dorsal root ganglion (DRG) in established PHN remains unknown. This pilot study examined the association between PLVI and the reduction in numeric rating scale (NRS) pain scores 1 month after pRF. METHODS: Patients with PHN who underwent DRG pRF and had CT or MRI scans availab

Piezoelectric actuator-evoked mechano-gated currents in trigeminal neurons innervating the mouse temporomandibular joint.

The most burdensome symptom of temporomandibular joint disorders (TMJDs) is mechanically evoked pain. To identify the trigeminal ganglion (TG) neuronal subtypes mediating mechanical sensitivity, we combined whole-cell patch-clamp electrophysiology with immunohistochemistry (IHC) to characterize mouse TMJ-innervating TG neurons retrogradely labeled from the TMJ. Electrophysiological analysis identified five distinct neuronal groups, only one of which responded to piezo actuator-induced mechanical stimulation. TMJ innervation was dominated by C- and A-nociceptors, comprising four of the five ide

PRDM16 is necessary for sensory neuronal development and differentiation in the trigeminal ganglion.

BACKGROUND: Cranial neural crest cells (cNCC) generate craniofacial cartilage, bone, and peripheral neurons and glia, and birth defects arise when the cartilage/neuronal/glial progenitors fail to differentiate. PRDM16 is a transcriptional regulator containing both zinc-finger and SET domains, implicated in craniofacial development and orofacial clefting, but its role in cranial sensory ganglion formation has not been defined. RESULTS: Here, we demonstrate that prdm16 is required for trigeminal ganglion (TG) assembly and sensory neurogenesis from cranial neural crest lineages. In zebrafish, pr

The PLLA/Gel@BBR 3D Nanoscaffold Protects Hair Cells From Neomycin Damage by Reducing Oxidative Stress.

Sensorineural hearing loss (SNHL) is the most prevalent form of hearing impairment observed in clinical practice. The primary pathological characteristic of this condition is the irreversible damage to the cochlear hair cells (HCs) and the associated spiral ganglion neurons (SGNs). Since HCs in the mammalian inner ear lack intrinsic regenerative capacity, damage to these cells often results in irreversible hearing loss. In recent years, two-dimensional (2D) culture systems have made substantial progress in supporting the regeneration of auditory cells. However, the regenerated HCs in these 2D

Caveolin-1 defines astrocyte mechanobiology in the glial lamina of the optic nerve head.

The optic nerve head is the principal site of retinal ganglion cell axonal injury in primary open angle glaucoma, where astrocyte mechanobiology and extracellular matrix remodeling are thought to play critical roles in disease progression. Caveolae, which are mechanosensitive plasma membrane invaginations formed by Caveolin and Cavin proteins, are key regulators of cellular responses to mechanical stress, yet their involvement in optic nerve head pathophysiology has not been defined. Here, we characterize the presence, regulation, and mechanobiological relevance of Caveolin and Cavin proteins

Mas-related G protein-coupled receptors contribute to compound 48/80-induced mast cell-independent activation of dorsal root ganglion neurons and itch in mice.

OBJECTIVE: Compound 48/80 (C48/80) is widely used as a mast cell degranulator, and its pruritogenic action has generally been attributed to mast cell-derived mediators. Whether C48/80 also drives itch through a mast cell-independent neuronal mechanism remains unresolved. This study aimed to determine whether C48/80 evokes itch through direct, mast cell-independent activation of dorsal root ganglion (DRG) sensory neurons and to identify the receptors involved. METHODS: C48/80-evoked scratching was assessed in mice after resiniferatoxin (RTX)-mediated ablation of capsaicin-sensitive afferents,

Modulation of human dorsal root ganglion neuron excitability by Nav1.7 inhibition.

Nav1.7 voltage-gated sodium channels are strongly expressed in human primary pain-sensing neurons (nociceptors), and selective Nav1.7 inhibitors have been developed as possible therapeutic agents for treating pain, so far with disappointing clinical results. In contrast, a selective Nav1.8 channel inhibitor (suzetrigine) has had successful clinical trials. Because nociceptors express both Nav1.7 and Nav1.8 channels, it is of interest to compare effects of Nav1.7 and Nav1.8 inhibitors on the excitability of human nociceptors. To compare with previous results with suzetrigine, we characterized t

Behavioral genomics identifies Creld1 as a cross-modal regulator of somatosensation.

Somatosensation is mediated by dorsal root ganglion (DRG) neurons, which convert physical and chemical stimuli into electrical signals. However, the full repertoire of molecular components underlying somatosensation remains incompletely identified. Here we developed an efficient postnatal CRISPR-Cas9 knockout platform for interrogating DRG-expressed genes (such as Piezo2, Trpv1 and Scn9a) in mice using AAV9-mediated single-guide RNA delivery. Combining this approach with somatosensory behavioral assays, we performed a targeted screen of 20 genes and identified Creld1 as a cross-model regulator

Comparison of palmaris longus and split flexor carpi radialis tendon grafts in two-stage flexor tendon reconstruction.

Two-stage flexor tendon reconstruction continues to be an important option for delayed or missed Zone II flexor tendon injuries when direct repair is no longer feasible. Although the palmaris longus is commonly used as a donor tendon, it may be absent, unavailable, or unsuitable in some patients. In this retrospective comparative study, we evaluated the feasibility of split flexor carpi radialis tendon grafts as an alternative donor source and described their outcomes in relation to palmaris longus grafts. Thirty patients treated between December 2022 and February 2025 were included; 14 receiv

Multichannel optical cochlear implants enable spectrally distinct auditory activity.

When hearing fails, cochlear implants (CIs) restore auditory perception. Yet, coding of spectral information remains a bottleneck because each electrode broadly activates the auditory nerve. As light can be more conveniently confined in space, optical (o)CIs present an alternative. Here, we combined expression of the potent channelrhodopsin ChReef in spiral ganglion neurons (SGNs) with oCIs comprising 5-10 green LEDs in gerbils. This combination enabled systematic comparison of encoding intensity and spectral information by individual oCI channels to acoustic and electrical stimulation within
