Calcium-sensing receptors promoted Homer1 expression and osteogenic differentiation in bone marrow mesenchymal stem cells.

Homer1 interacts with calcium-sensing receptors (CaSRs) in osteoblasts (OBs), with both CaSR and Homer1 playing essential roles in AKT phosphorylation. This study investigated the impact of CaSR on Homer1 expression during the differentiation of rat bone marrow mesenchymal stem cells (BMSCs) at morphological, imaging, and molecular levels, both in vivo and in vitro. A post-oophorectomy osteoporosis model was established in Sprague-Dawley rats, validated through micro-computed tomography, hematoxylin-eosin staining, and biomechanical testing to assess in vivo changes in CaSR expression. BMSCs w

Relationship between blood eosinophilia and disease severity in chronic rhinosinusitis with nasal polyps.

AIMS: New chemotherapeutic agents prolong survival of patients with pancreatic ductal adenocarcinoma (PDAC). Although their incidence is rising, patients with end-stage renal disease (ESRD) requiring hemodialysis (HD) are not included in the phase III trials evaluating the effects of these chemotherapies. Many experts recommend applying chemotherapy after HD using a reduced dose. Alternatively, the concept of prior dosing allows for the application of dialyzable chemotherapeutic drugs using a normal dose, with an HD followed shortly after to mimic normal renal function. In this work, we provid

Bone loss after oophorectomy among high-risk women: an NRG oncology/gynecologic oncology group study.

OBJECTIVE: Women undergoing premenopausal oophorectomy for a variety of reasons, including to reduce ovarian or breast cancer risk were evaluated for accelerated bone loss. METHODS: The Gynecologic Oncology Group (GOG)-0215 randomized phase-II trial of zoledronic acid was initiated to determine if postoophorectomy bisphosphonate therapy could prevent this bone loss. The study was closed after slow accrual prevented evaluation of the primary study endpoint. We analyzed changes in bone mineral density (BMD) among the 80 women randomized to the observation arm of this study, as measured 3, 9, an

Bone resorption versus estrogen loss following oophorectomy and menopause.

OBJECTIVES: To investigate if menopause and oophorectomy may represent different risk factors for bone resorption/loss. METHODS: The urinary levels of pyridinoline (Pyr) and deoxypyridinoline (D-pyr), the serum levels of type I carboxy-terminal pyridinoline cross-linked telopeptide (ICTP), and lumbar bone mineral density (BMD), were compared in 80 Japanese women after menopause or oophorectomy. These women were divided into four groups of 20 women each as follows: early postmenopausal stage (early physiologic menopause < 3 years before study entry); late postmenopausal stage (physiologic meno

Changes in serum levels of type I collagen-related proteins after surgically induced menopause and correlations with bone loss in the lumbar spine.

The purpose of this prospective study was to characterize the changes in serum levels of two proteins produced during the synthesis and degradation of type I collagen, i.e., the carboxyterminal propeptide of type I procollagen (PICP) and the pyridinoline cross-linked carboxyterminal telopeptide of type I collagen (ICTP), respectively, after oophorectomy, and to assess the degree of correlation between changes in the serum values of these proteins and changes in bone mineral density (BMD) of the lumbar spine. Serum levels of PICP, ICTP and bone gla protein (BGP) were determined in 18 women befo

Cytokines and osteoporosis.

Bone remodeling is a complex process that involves cells from various lineages, the calcified extracellular bone matrix and a multitude of regulation factors that act at both the tissular and cellular levels. Resorption and formation of bone are regulated at various levels during the series of events that lead from stem cell proliferation to differentiation of mature cells. These events are controlled by calciotropic hormones, whose indirect cellular effects are mediated by the production of local factors. A host of cytokines and growth factors influence the proliferation and differentiation o

Prevention of post-oophorectomy bone loss with Tibolone.

The results of a 1-year placebo-controlled study in 25 women who had hysterectomy and bilateral oophorectomy receiving a daily oral dose of 2.5 mg of Tibolone (Org OD 14) are presented. Tibolone is a steroid compound with mild oestrogenic in additional to progestagenic effects. Post-oophorectomy bone loss has been reported to accelerate, at least during the first year after surgery. The efficacy of Tibolone to prevent this accelerated bone loss is questionable. All patients were scheduled to participate in the study before oophorectomy. Patients had a detailed pre-operative examination, includ

Parathyroidectomy does not prevent bone loss in the oophorectomized rat.

The evidence for a role of parathyroid hormone in the bone loss after the menopause remains controversial. This study examines the effect of parathyroidectomy on femoral trabecular bone volume, thickness, and spacing and biochemical markers of bone turnover in the oophorectomized rat. Female Sprague-Dawley rats 3 months old were double sham operated (sham), oophorectomized (OPX), parathyroidectomized (PTX), or oophorectomized and parathyroidectomized (O/P) under halothane anesthesia. At 9 weeks postoperation, femoral trabecular bone volume (BV/TV) was lower in OPX and O/P rats compared with sh

Differences in the time course of the effects of oophorectomy in women on parameters of bone metabolism and interleukin-1 levels in the circulation.

Interleukin-1 is a cytokine with bone-resorbing activity which is claimed to play a role in the pathogenesis of osteoporosis and of post-oophorectomy bone loss. We measured plasma levels of interleukin-1 (IL-1) in 16 women who underwent hysterectomy and bilateral oophorectomy and in 12 women with hysterectomy alone before surgery and 7, 15, 30, 45 and 60 days after surgery. Only oophorectomized patients showed a significant increase of circulating interleukin-1 levels. This increase was significant only 30 days after surgery, whereas a rise at 7 days (P < 0.001) was observed for serum calcium

Susceptibility effects of bone trabeculae. Quantification in vivo using an asymmetric spin-echo technique.

RATIONALE AND OBJECTIVES: The authors assessed the capability of magnetic resonance imaging in measuring bone mineral density in vivo. METHODS: Using the in-phase asymmetric spin-echo technique, the susceptibility effect (T2*susceptibility) was measured in lumbar vertebrae in vivo. Calcium density was measured with single energy quantitative computed tomography scan. The values of the susceptibility effect were correlated with calcium density. RESULTS: A statistically significant positive correlation was observed between signal loss due to susceptibility-induced inhomogeneity and the calcium

[Prevention of post-ovariectomy osteoporosis in the rat. Comparative study of synthetic salmon calcitonin and eel calcitonin aminosuberic analogue].

The authors evaluate the efficacy of salmon calcitonin and aminosuberic analogue of eel calcitonin in the prevention of post-oophorectomy osteoporosis in rats. Both drugs, administered at the same dosage, are equally effective in preventing oophorectomy bone loss in rats. Besides, plasma biochemical evaluations demonstrate that calcitonins reduce bone turnover in treated rats, compared with control oophorectomized ones.

Effects of salmon calcitonin suppositories in the prevention of bone loss in oophorectomized women.

To evaluate the efficacy of salmon calcitonin (sCT) suppositories in inhibiting postmenopausal bone demineralization and the tolerability of this formulation, we treated ten women oophorectomized in reproductive age with intrarectal sCT at the dose of 200 IU/day for 12 months postoperatively. Modifications of calcium and phosphate metabolism were quantified by measurements of vertebral and peripheral bone mineral content, serum alkaline phosphatase, and hydroxyprolinuria at baseline and at 6 and 12 months of treatment. Tolerability was assessed by recording the appearance of local or systemic

Fuller Albright. His concept of postmenopausal osteoporosis and what came of it.

Fifty years ago Albright contributed the following to understanding osteoporosis: (1) He recognized it as a deficiency of formation, not of mineralization of bone matrix; (2) he observed that 40 of 42 patients with osteoporosis before age 65 were women past menopause or young women postoophorectomy; (3) he concluded that estrogen stimulates osteoblasts (a conclusion later challenged); (4) he demonstrated by metabolic balance studies that estrogen causes a positive calcium balance in postmenopausal osteoporosis; (5) he introduced periodic progesterone to prevent or treat endometrial hyperplasia

Measurement of bone mineral density in amenorrheic women with dual photon absorptiometry.

Bone mineral density (BMD) was measured in 17 patients with gonadal dysgenesis, 21 patients with premature ovarian failure, 14 patients with hyperprolactinemia, 23 patients postoophorectomy and 41 normal healthy women. Each group was divided into two subgroups, on the basis of whether or not they were receiving estrogen replacement therapy. Linear regression analysis showed a significant loss of BMD at a rate of 0.006 g/cm2 per year in the lumbar spine of normal healthy women. The distribution of individual values in each study group was near or within normal confidence limits, except for the

Serum bone gla protein and the vitamin D endocrine system in the oophorectomized rat.

Because of the importance of estrogen in osteoporosis, the effects of decreased estrogen production using sensitive measurements of bone mineral metabolism were studied in oophorectomized rats. Serum levels of ionized calcium, bone gla protein (BGP), vitamin D metabolites (25-hydroxyvitamin D and 1,25-dihydroxyvitamin D), and estradiol were measured before and serially for 6 weeks after oophorectomy in the rat. In addition, static and dynamic indices of bone histomorphometry were determined after double tetracycline labeling. Fifty Sprague-Dawley female rats (approximately 250 g) were studied.

Postoophorectomy bone loss is associated with reduced bone Gla protein serum levels: a possible effect of osteoblastic insufficiency.

To further investigate the relationship between oophorectomy (OF) and mineral bone loss, 15 women who underwent total hysterectomy and bilateral oophorectomy were studied for 12 months after surgery. Mineralometric and metabolic data were obtained before and after 3, 6, and 12 months. The women lost bone mineral content (measured by single photon absorptiometry) at the same rate they lost cortical and trabecular bone, suggesting that bone loss after OF is a generalized phenomenon. Our data also show that an increase in bone resorption takes place only in the first period after OF; the persiste

Prevention of spinal osteoporosis in oophorectomised women.

100 women who had taken part in a prospective controlled trial of oestrogen therapy for prevention of post-oophorectomy bone loss were reviewed after a median follow-up period of nine years. A significant reduction in height occurred among the placebo-treated group, but not in the group treated with mestranol (mean 23 x 3 micrograms/day). The placebo-treated group had a higher spine score, lower central vertebral height, and larger wedge-angle than the oestrogen group. Within each group none of these spinal morphometric changes correlated with changes in mineral content of metacarpal or radial
