Effects and mechanisms of iron overload on the proliferation and differentiation of preosteoblastic cells via a 3D microsphere culture system.

BACKGROUND: Iron overload-related osteoporosis has garnered significant attention, yet its pathological mechanisms remain unclear. Traditional two-dimensional (2D) culture systems often fail to recapitulate the extracellular matrix (ECM) microenvironment, leading to discrepancies between in vitro and in vivo findings. METHODS: We developed a three-dimensional (3D) culture system using methacrylated gelatin (GelMA) microspheres to culture preosteoblastic cells, simulate the bone microenvironment under iron overload conditions, and systematically examine changes in cellular morphology, viabilit

Sialin-STAT3 axis regulates bone homeostasis in mice.

Mitochondrial regulation in mesenchymal stem cells (MSCs) serves as a critical determinant of bone formation and skeletal homeostasis. While dietary nitrate and its transporter Sialin are implicated in systemic homeostasis, their specific roles in MSCs' function remain unclear. Here, we demonstrate that Sialin deficiency impairs MSCs' function and disrupts bone homeostasis. Gain- and loss-of-function studies reveal that Sialin localizes to the mitochondrial membrane and promotes osteogenic differentiation by maintaining mitochondrial bioenergetic integrity. Mechanistically, Sialin recruits pST

Age-related GSS promoter methylation in BMSCs drives osteoporosis and the reversal by targeted GSH delivery.

Age-related osteoporosis arises from bone tissue with inadequate metabolic support for osteogenesis. We identified that DNA methylation-mediated suppression of glutathione synthetase (GSS) represents an upstream lesion limiting endogenous glutathione (GSH) synthesis and supply in aged bone, thereby constraining osteoblast differentiation. In turn, impaired GSH synthesis exacerbates oxidative stress levels and diminishes osteogenic capacity, and this metabolic bottleneck is independent of substrate availability: cysteine supplementation neither restored GSH synthesis flux in aged bone nor rescu

Initiation and Completion of Hormone Therapy in Older Women With Early-Stage Breast Cancer.

PURPOSE: Clinical trials suggest that adjuvant radiation therapy (RT) may be omitted in women aged ≥65 years with early-stage, hormone-receptor-positive breast cancer, provided completion of 5 years of hormone therapy. However, at the time of RT consult, it is often unknown whether the patient will start hormone therapy or will start but not complete 5 years of therapy, either of which, if known in advance, would alter RT recommendations. We studied a cohort of patients who would have been eligible for RT omission to examine factors associated with declining or discontinuing hormone therapy.

Dynamic monitoring of bone homeostasis by dual fluorescent imaging of osteoblast and osteoclast activity.

Bone homeostasis is maintained through a dynamic balance between osteoblasts (OBs)-mediated bone formation and osteoclasts (OCs)-driven bone resorption. While numerous probes have been developed to detect either alkaline phosphatase (ALP) or extracellular protons (H⁺), which serve as key biochemical markers for OBs and OCs activity, respectively, no existing single molecular imaging platform enables the simultaneous, real-time monitoring of both bone formation and resorption in situ. In this study, we developed a dual-responsive near-infrared (NIR) fluorescence (FL) imaging platform for real-t

Subchondral Insufficiency Fracture of the Humeral Head: A Rare Case in a Young Person.

Subchondral insufficiency fracture (SIF) of the humeral head is rare compared to SIF of the femoral head, and most of the reported cases have been of elderly women with osteoporosis. We report a rare case of SIF of the shoulder in a man aged 32 years. Following a consultation with a local physician, the patient was referred to our hospital with worsening right shoulder pain and suspected humeral head necrosis based on magnetic resonance imaging (MRI) findings. The patient had no history of steroid or excessive alcohol consumption. The patient had a limited range of motion in the shoulder joint

Local Delivery of Genistein in Peri-Implant Defects Enhances Osseointegration in Ovariectomized Rats.

BACKGROUND: Estrogen deficiency, a key factor in osteoporosis, may influence peri-implant healing, which requires cautious and individualized planning for dental implant placement. Functionalizing biomaterials with bioactive molecules, such as soy-derived isoflavone, could enhance their osteoconductive potential. OBJECTIVE: Investigate the performance of genistein in the functionalization of bioactive glass and deproteinized bovine bone by sonochemistry on peri-implant bone repair in ovariectomized rats. METHODS: In total, 50 female rats were divided into five groups (n = 10): blood clot (CL

Trends of clinical vertebral and hip fractures following local government involvement in fracture prevention from 2015 to 2021 in Kure, Japan.

UNLABELLED: Community-wide Fracture Liaison Services and educational outreach to local physicians on osteoporosis can effectively reduce the fragility fracture burden at the population level in a super-aged society. PURPOSE: Effective prevention of osteoporosis and fractures requires not only implementing a Fracture Liaison Service (FLS) but also enhancing osteoporosis awareness among local physicians, residents, and the municipality. We examined the impact of multidisciplinary, municipality-supported FLS and osteoporosis awareness activities on the incidence of clinical vertebral and hip fra

In situ electrospinning at the operating table to immobilise mesenchymal stem cells on the bony wall for the regeneration of osteoporotic bone defects.

Stem cell seeding is an important approach for the treatment of osteoporotic bone defects. However, existing seeding methods rely on carriers such as scaffolds to retain cells within the defect, making it difficult for cells to adhere directly to the complex surface of the bony wall. In this study, mesenchymal stem cells (MSCs) were delivered into in vivo bone defects with a handheld electrospinning device as an improved cell seeding strategy. First, flow cytometry and related assays confirmed that electrospinning had minimal effects on MSCs viability. MSCs were then immobilised by electrospin

Advancements in ex vivo bone biomechanics: multimodal technologies and their integration with artificial intelligence.

Bone biomechanics is essential for understanding how bone structure, composition, and mechanical function interact in healthy and diseased states. Conventional approaches such as mechanical testing, micro computed tomography, and histology provide valuable information but cannot fully capture the multiscale heterogeneity that governs bone strength and fragility. This review summarizes recent progress in advanced ex vivo bone biomechanical techniques that address these limitations, including synchrotron-based phase contrast imaging, digital volume correlation, three dimensional bioprinting, aco

NMN prevents obesity-induced osteoporosis by promoting mitophagy to restore type H vessels.

Obesity-induced osteoporosis is a growing global health challenge, necessitating more therapeutic strategies. Emerging evidence links the reduction of bone-specific type H vessels to osteoporosis due to their crucial role in angiogenesis-osteogenesis coupling. Nicotinamide Mononucleotide (NMN) is known to improve metabolic diseases, but its role in regulating type H vessels in obesity-induced osteoporosis remains unclear. An obesity-induced osteoporosis mouse model was established using a high-fat diet (HFD). In vitro, primary bone marrow mesenchymal stem cells (BMSCs) and human umbilical vein

ANGPTL8 accelerates bone loss in diabetic mice by promoting osteoclastic differentiation and inhibiting osteoblastic differentiation through AMPK pathway-mediated metabolic reprogramming.

Disruptions in energy metabolism within skeletal cells play a critical role in the pathogenesis of diabetic osteoporosis (DOP), primarily by affecting bone’s metabolic balance. The exact causes and regulatory mechanisms of these energy metabolism disorders in DOP remain unclear. Angiopoietin-like protein 8 (ANGPTL8), a protein involved in energy metabolism, is elevated in diabetics. This study aimed to investigate the role of ANGPTL8 in DOP development. In this study, a murine model of diabetic osteoporosis was developed, and an increase in plasma levels of ANGPTL8 was observed. The knockout o

Pedicled myofascial temporalis flap for closure of large maxillary defects after medication-related osteonecrosis of the jaws. A case series.

BACKGROUND: Medication-related osteonecrosis of the jaws (MRONJ) located in the maxilla may lead to challenging oro-antral and oro-nasal defects too extensive to be predictably closed with local soft tissue flaps. The use of a pedicled myofascial temporalis flap (PMTF) is, however, well-established for closing large maxillary defects following ablative craniomaxillofacial surgery. OBJECTIVES: The purpose of the present study was to evaluate the use of PMTF for maxillary defect closure in patients with stage 3 MRONJ. METHODS: A retrospective cohort study was conducted based on data from the C

A bibliometric analysis of research on bone marrow edema syndrome, transient osteoporosis of the hip, regional migratory osteoporosis, and related terms.

INTRODUCTION: Bone marrow edema syndrome (BMES), transient osteoporosis of the hip (TOH), and regional migratory osteoporosis are overlapping terms used inconsistently to describe a rare condition marked by transient joint pain, bone marrow edema, and localized demineralization. The lack of standardized terminology and diagnostic criteria has hindered research coherence. METHODS: In this bibliometric analysis, we aimed to characterize the historical and structural development of literature in this field and identify influential contributions, key themes, and collaborative patterns among autho
