tRNA-m1A modification safeguards fetal liver HSPCs from DNA damage by maintaining iron homeostasis.

Hematopoietic stem and progenitor cell (HSPC) development requires finely tuned gene expression programs, yet the role of transfer RNA (tRNA) modifications in this process remains largely unknown. In this study, we uncover the essential function of the tRNA methyltransferase, Trmt61a, in sustaining fetal liver HSPCs through N1-methyladenosine (m1A) deposition. Ribosome profiling revealed globally declined translation efficiency because of translational blockage upon Trmt61a loss in HSPCs, notably of the transferrin receptor (Tfrc). Mechanistically, m1A reduction caused ribosomal stalling at ar

Does immobilisation improve outcomes in proximal tibial stress fractures among military recruits?

INTRODUCTION: Proximal tibial stress fractures (PTSFs) are a significant cause of morbidity in military recruits, often requiring prolonged rehabilitation and leading to attrition from training. Despite their impact, little data exist to guide optimal treatment strategies, and most recommendations are extrapolated from diaphyseal tibial stress fractures. This study aims to describe fracture characteristics, treatment protocols and outcomes of Marine Corps recruits diagnosed with PTSFs. METHODS: A retrospective review was conducted of Marine recruits at the Marine Corps Recruit Depot San Diego

Suffering and the collapse of language: between the sayable and the impossible.

This article explores the relationship between human suffering and the limits of language, positing that suffering is not simply an experience of physical pain or emotional distress but a fracture in the capacity to articulate meaning. It clearly distinguishes between pain (a localised sensory experience that is potentially communicable) and suffering (an existential experience that involves a rupture of the symbolic order). It is argued that suffering appears when personal narrative is interrupted or becomes incoherent, provoking what is termed a 'collapse of language'-moments when experience

Mechanosensitive ion channels as novel targets in osteoporosis.

Osteoporosis is the most prevalent metabolic bone disease globally, leading to an increased risk of fractures. Recent advances in ion channel research have shed light on the importance of mechanosensitive ion channels as novel players in these pathophysiological processes. This perspective discusses the involvement of the mechanosensitive ion channels TREK-1, Piezo, and volume-regulated anion channels (VRACs) as potential novel pharmacological targets for the treatment of osteoporosis. TREK-1, a mechanosensitive K2P channel is important for maintaining the resting membrane potential in many ce

The Effect of Tricyclic Antidepressants on Fracture Healing: An Experimental Study.

INTRODUCTION: Disorders of mood and post-traumatic stress disorder (PTSD) are common after major trauma, and one of the treatments used is Tricyclic Antidepressants (TCA). These medications work by inhibiting the re-uptake of neurotransmitters like serotonin and noradrenaline. Serotonin is known to have measurable effects on bone tissue due to the presence of specific receptors on bone cells. However, there are conflicting reports about how serotonin signaling affects bone tissue and the process of fracture healing. This study aimed to evaluate the effect of TCAs on fracture healing. METHODS:

Synergistic enhancement of the mechanical properties of polyurethane ionogel by halometallate ionic liquid-induced phase separation and hydrogen bond cross-linking.

Ionogels, an emerging and rapidly developing type of soft material, are widely employed in flexible electronic devices. However, the solvation effect induced by ionic liquids (ILs) causes increased fracture strain and reduced fracture strength in polymer matrices, and balancing this trade-off remains challenging. In this work, halometallate ILs were dispersed in a poly(urethane-urea) (PUU-AD) network using a swelling strategy to prepare high-strength (37.65 MPa) and high-toughness (153.30 MJ m-3) PUU-AD ionogels (PUU-AD/IL), with 67.6% and 76.2% higher strength and toughness than the pure PUU-

Numerical Simulations of In-Plane and Transmural Tear Propagations in Aortic Dissection: Possible Mechanisms Behind Dissection Progression.

The early development of aortic dissections (AD) is manifested by tear propagation. The direction and extent of tear propagation are important for surgical strategy selection and outcomes, yet the mechanism underlying early tear propagation is largely unknown. Interface damage, leading to in-plane propagation, is modeled using the cohesive zone method. Bulk material damage, causing transmural propagation, is modeled using a strain-energy-based damage criterion. The influences of geometrical parameters are examined with a double-layer finite-element model of a three-dimensional (3D) idealized a

Vitamin C as an epigenetic gatekeeper of osteogenesis and skeletal health: a new perspective on an ancient molecule.

Vitamin C has been long recognized as an important nutrient for skeletal biology, historically attributed to its role in collagen synthesis and connective tissue integrity. Recent studies, however, reveal vitamin C as a critical epigenetic regulator of cellular differentiation. As a required cofactor for α-ketoglutarate-dependent dioxygenases, vitamin C controls the enzymatic activity of a broad array of histone and DNA demethylases, thereby modulating chromatin accessibility and driving cell-specific gene expression. This review provides a novel, integrated perspective that directly links vit

A Probabilistic Method to Model Progressive Metatarsal Displacement and Stiffness During Fatigue Testing.

To better understand the mechanisms of bone stress injuries (BSI) in metatarsals, we developed an algorithm that adapts finite element (FE) models of metatarsals to simulate fatigue displacements through progressive stiffness loss. Twenty-two human metatarsals were imaged using computed tomography (CT) and then cyclically loaded in uniaxial compression until failure. CT images were used to generate specimen-specific FE models, and a custom program was developed to iteratively simulate cyclic loading and progressive stiffness loss associated with microdamage accumulation. Probability was incorp

Subject-Specific Modeling of the Proximal Femur During Falls: Dual Importance of Impact Dynamics and Bone Morphology.

Clinical risk factors for hip fracture can influence both fall-induced loading and underlying femur morphology/strength; however, these effects are generally studied in isolation. We evaluated the potential effects of fall-type, biological sex, and trochanteric soft tissue thickness (TSTT) on femoral neck stresses and fracture risk index during lateral impacts. Thirty-two young adults completed voluntary falls, representative of falls in older adults. Peak impact force magnitude, direction, and point of application were extracted and applied to subject-specific beam models generated from dual-

Bond strength of denture teeth to denture bases in CAD/CAM-milled, printed, and conventional fabrication techniques.

OBJECTIVE: This in-vitro study examined the bond between denture teeth (milled, printed, and prefabricated) and milled, printed, and autopolymerized denture bases. METHOD AND MATERIALS: In total, 88 specimens (11 groups, n = 8 per group) of denture teeth and denture base resins were fabricated using systems of three different manufacturers (Vita Zahnfabrik, Ivoclar Vivadent, and Formlabs). The groups consisted of combinations of six different teeth, five different denture base materials, and four different adhesive bonding options. Teeth were either printed, milled, or prefabricated, while de

Achieving strength and toughness limits of anisotropic microstructured alumina ceramics through interface engineering.

Advanced ceramic composites face a critical challenge: achieving the combined strength and toughness as in natural materials like nacre. Bioinspired anisotropic microstructured ceramics (AMCs) address this by mimicking nacre's hierarchical architecture. However, the critical role of the ceramic-matrix interface is often overlooked, due to a fundamental conflict: the weak interfaces needed for toughening through crack deflection are naturally difficult to achieve with conventional sintering processes that prioritize densification. Here, we bridge this gap by establishing universal energy- and s

PCK2-Mediated PQBP1 Lactylation Promotes Asthmatic Inflammation through PRMT5 Inhibition.

Based on their immunodominant nature and ability to induce appropriate immune responses in the host, several antigens of Mycobacterium tuberculosis have shown promise of protection. However, most of the candidate vaccines developed by employing various strategies have afforded protection that is at best comparable with bacillus Calmette-Guérin (BCG) in animal models. Due to the inherent ability of BCG to prime cellular responses in the host, it has become an attractive vehicle for development of a vaccine against intracellular infections. In this study, we have cloned the genes of three immuno

Load Transfer in Tibial Intramedullary Nailing: Effects of Fracture Level, Screw Configuration and Nail-Canal Clearance.

Intramedullary nails are widely recognized for their high stabilization capacity and are considered the gold standard for tibial fracture treatment. However, hardware failure, especially of distal screws, is frequently reported, especially in cases involving distal fractures. Although various experimental studies explore screw configurations, few quantify the mechanical loads acting on individual screws at different fracture locations. This study numerically evaluates two fixation systems: one with four screws and another with five, including an additional distal screw (DS3). The results show

Mechanically robust polyurethane elastomers enabled by soft-segment-regulated hydrogen bonds and microphase separation for ultrasound imaging medical catheters.

Conventional strategies for enhancing the mechanical robustness of thermoplastic polyurethane elastomers (TPUs) rely on hard-segment engineering, such as introducing dynamic covalent/noncovalent bonds or optimizing chain extenders, yet overlook the critical role of soft segments in governing microphase separation. Here, we present a soft-segment-regulated design that leverages crystallizable polyols to synergize hierarchical hydrogen bonding, tunable microphase separation, and strain-induced crystallization (SIC), achieving excellent mechanical performance. Among them, PU-PTMEG exhibits except
