One dose of adjuvant 8 Gray postoperative radiotherapy for Merkel cell carcinoma excised with narrow margins: Low toxicity and comparable efficacy to conventional radiotherapy.

BACKGROUND: Merkel cell carcinoma (MCC) recurs in 17% to 30% of patients with stage I to II disease, with local recurrence rates between 2% and 27%. Per established guidelines, conventional postoperative radiotherapy (cPORT; ∼50 Gray across 25 fractions) should be considered for localized MCC when risk factors are present. cPORT can be morbid and logistically challenging. Single-fraction radiotherapy (SFRT) of 8 Gray has utility and low toxicity for palliation but remains underexplored postoperatively. OBJECTIVE: Evaluate postoperative SFRT for stage I to II MCC, compared to observation and c

Glutamine synthetase genes family mediates nitrogen metabolism and disease responses in Kentucky bluegrass.

BACKGROUND: Kentucky bluegrass (Poa pratensis L.), an important cool-season turfgrass species, requires adequate nitrogen (N) fertilization for normal growth and development. It is also susceptible to infection by pathogens. The glutamine synthetase (GS) gene family, known to play central roles in plant N metabolism and disease responses across higher plants, has functions that remain uncharacterized in Kentucky bluegrass. RESULTS: We identified and characterized four GS genes (PpGS1.1, PpGS1.2, PpGS1.3, and PpGS2) in Kentucky bluegrass. Phylogenetic analysis revealed a close relationship wit

miR171c and miR172a confer resistance to tomato mottle mosaic virus in tobacco by targeting SCL and AP2 transcription factors.

BACKGROUND: Tomato mottle mosaic virus (ToMMV) is an emerging tobamovirus that posing a significant threat to the yield and quality of solanaceous crops globally. To elucidate the regulatory role of small RNAs in the host response to ToMMV, we used Nicotiana tabacum var. Xanthi nc as a model system and performed small RNA sequencing across three distinct disease stages: mosaic symptom onset, linear leaf symptom development, and the recovery phase. By integrating qRT-PCR validation with functional assays, including miRNA overexpression, virus-induced gene silencing, and biological inoculation,

Predictors of routine HPV-based cervical cancer screening adoption in federally qualified health centers within a multi-state practice research network 2012-2022: retrospective observational study.

BACKGROUND: Cervical cancer screening guidelines in the United States have evolved with the introduction of human papillomavirus (HPV)-based modalities, including co-testing in 2012 and primary HPV testing in 2018, in addition to Pap-only screening. However, adoption of these modalities in federally qualified health centers (FQHCs) has been underexplored. We examined longitudinal patterns of HPV-based cervical cancer screening adoption (e.g., proportion screened with co- and primary HPV testing) across three FQHC systems in Washington and Idaho and explored factors associated with receipt of H

Virosome-masked ratiometric nanoprobes for in vivo dynamic imaging of influenza a virus infection.

In vivo fluorescent in situ visualization of viral infection dynamics is crucial for elucidating the mechanisms of viral pathogenesis. However, current approaches lack sufficient specificity and sensitivity for spatiotemporal monitoring of viral infection in vivo. Here, we proposed virosome-masked ratiometric nanoprobes (VMR-NPs) integrated with dual-stimuli-responsive near-infrared (NIR) Förster resonance energy transfer (FRET) reporters that specifically recognize influenza A virus (IAV) viral RNA (vRNA) and apurinic/apyrimidinic endonuclease 1 (APE1). Sustained Utilization of Video-Based Outpatient Psychiatry Visits Following the COVID-19 Public Health Emergency at an Academic Health Center.

BACKGROUND: Telepsychiatry expanded rapidly during the COVID-19 pandemic to support continuity of psychiatric care, although its long-term durability following the expiration of pandemic-era policies remains uncertain. This study evaluated long-term utilization of telepsychiatry and patient and clinician acceptability within outpatient practice. METHODS: A repeated cross-sectional electronic health record analysis of outpatient psychiatric visits from 2020 to 2025 compared in-person and virtual visit completion and no-show rates across five annual intervals. Telepsychiatry acceptability was e

Identification of microRNAs as biomarkers for triage of cervical intraepithelial neoplasia in cervical scrape samples from high-risk HPV-positive women.

BACKGROUND: Cervical cancer (CC) is the fourth leading cause of cancer death in women worldwide. High-risk human papillomavirus (HR-HPV) testing is highly sensitive but lacks specificity for identifying women at risk of progression. Reliable biomarkers are urgently needed to identify HR-HPV-positive women most likely to develop cervical cancer. Aberrant microRNA (miRNA) expression contributes to HPV-driven carcinogenesis and represents a promising diagnostic tool. METHODS: Cervical scrape samples from HR-HPV-positive women were histologically classified as low-grade (≤CIN1) or high-grade (CIN

Viral control of translation: rise of the ribosome.

Viruses have evolved a myriad of strategies to manipulate their host's protein synthesis machinery, studies of which uncovered many founding principles of translational control. Decades of research have established our now-extensive understanding of how these pathogens target the array of translation factors that recruit ribosomes and regulate scanning, start site selection, decoding, and termination. Meanwhile, the ribosome itself was largely regarded as a code-reading machine that lacks regulatory influence. Recent advances in cryo-electron microscopy and quantitative proteomics have upended

TRIM28 induces the PDCoV NS6 degradation through the ubiquitin proteasome pathway.

Porcine deltacoronavirus (PDCoV) causes a significant threat to the global swine industry. The PDCoV accessory protein NS6 has the capability to antagonize type I interferon (IFN-I) production and influence viral proliferation. In this study, we discovered that tripartite motif-containing 28 (TRIM28) interacts with NS6 via its RING and coiled-coil (CC) domains, facilitating K48-linked ubiquitination of NS6, thereby leading to the degradation of NS6. Furthermore, NS6 attenuates the expression of interferon-stimulated genes (ISGs). The inhibition of the interferon (IFN) response is further inten

Primer: Giant viruses of protists.

Giant viruses infecting protists have expanded our understanding of viral diversity. These members of the phylum Nucleocytoviricota possess genomes that can exceed 2.5 Mb and encode hundreds to thousands of proteins, including functions once thought unique to cellular organisms. They infect amoebae, algae, and other unicellular eukaryotes, forming large factories and interacting with virophages and mobile genetic elements. Their virions display diverse architectures, from icosahedral particles to amphora-shaped pandoraviruses and tail-bearing tupanviruses. Comparative genomics highlights evolu

A foundation-model approach to pediatric headache classification from resting-state fMRI.

Headache is the most common neurological disorder in children and substantially affects quality of life. We investigated whether resting-state functional MRI (rs-fMRI) can support pediatric headache classification using machine learning. We encoded rs-fMRI data using NeuroSTORM, a recent foundation model, and fine-tuned it to distinguish healthy controls from children with headache and subsequently classify headache subtypes. We then compared NeuroSTORM with a standard neuroscience approach that uses functional connectivity (FC) matrices derived from brain activity as predictors. Using 189 rs-

Ecophysiological mechanisms underlying tomato spotted wilt of tobacco under climate change.

Tomato spotted wilt is an important viral constraint on tobacco production, but the direction and magnitude of future climate effects remain uncertain. This systematized narrative review evaluates evidence linking climate drivers with cultivated tobacco physiology, tomato spotted wilt virus (TSWV), thrips-vector ecology, weed reservoirs, surveillance, and management. Direct multifactorial experiments that simultaneously manipulate climate, TSWV, tobacco, and vector species are scarce; therefore, the synthesis explicitly separates direct tobacco evidence from evidence derived from other TSWV ho

Immunohistochemical Analysis of INSM1, MCPyV, and PHOX2B Expression in Merkel Cell Carcinoma.

BACKGROUND: Merkel cell carcinoma (MCC) is a rare and aggressive cutaneous neuroendocrine malignancy with distinct etiological pathways, including Merkel cell polyomavirus (MCPyV) infection and ultraviolet (UV)-induced carcinogenesis. Owing to the lack of large case series, the etiology of MCC in Turkey remains unclear. INSM1 is a highly specific neuroendocrine marker that supports the diagnosis of MCC. PHOX2B, although known to be expressed in neuroblastoma, has been only rarely investigated in MCC. METHODS: We retrospectively evaluated 14 MCC cases diagnosed between 2008 and 2023 at Istanbu

Reconstituting human and SV40 replisomes with Okazaki fragment maturation reveals synchronized leading- and lagging-strand synthesis.

Eukaryotic lagging-strand synthesis requires iterative priming of Okazaki fragments (OFs) by Pol α, extension by Pol δ, and subsequent primer removal and ligation through Pol δ-mediated maturation of OFs (MOF). How the replisome coordinates these time-consuming lagging-strand processes while maintaining rapid and synchronized DNA synthesis remains unclear. Here, we reconstitute and compare the human and Simian Virus 40 (SV40) replisomes in the presence of MOF. In the human replisome, Pol α is recruited distributively to the CMG helicase for priming and transiently slows leading-strand synthesi

Preventing cancer Down Under: Australian screening guidelines for prevention of anal cancer in people living with HIV: the pathologists' viewpoint.

Anal squamous cell carcinoma (ASCC) is a human papillomavirus (HPV)-associated cancer with a rising incidence. People living with human immunodeficiency virus (PLHIV) are at the highest risk. Recent evidence has shown that treatment of the precursor lesion, high-grade squamous intraepithelial lesion (HSIL), can prevent cancer development. Consequently, several countries have published screening guidelines for anal HSIL as a strategy to decrease cancer incidence. In Australia, the Australasian Society for HIV, Viral Hepatitis and Sexual Health Medicine (ASHM) has recently released guidelines fo

RNA-encoded PGT121-LS anti-HIV antibody: Comprehensive preclinical characterization and translational pharmacokinetics.

Human immunodeficiency virus (HIV)-1 broadly neutralizing antibodies (bNAbs) have demonstrated clinical efficacy but face manufacturing challenges associated with recombinant protein production and purification. Here, we present a ribonucleic acid (RNA)-encoded bNAb (RibobNAb) platform that enables in vivo antibody production of the clinically validated bNAb PGT121 via lipid nanoparticle (LNP) delivery, supporting rapid evaluation of Fc variants (LS, del294, and LS-del294) in vitro and in vivo. Detection of Respiratory Syncytial Virus (RSV) in Black-Horned Capuchin Monkeys, Brazil.

We carried out a viral surveillance study on 30 black-horned capuchin monkeys (Sapajus nigritus) in Rio Doce State Park, a protected area in the Brazilian Atlantic Forest. Samples were tested for orthopoxviruses, flaviviruses, coronaviruses, influenza viruses, and a wide range of other respiratory viruses using molecular and serological methods. RT-qPCR detected the respiratory syncytial virus (RSV) genome in oral swabs from two asymptomatic juvenile females sampled in an area with high human interaction. This finding indicates a possible exposure of an population to RSV, a contagious human pa

Comparison of three routine methods for the identification of Kerstersia gyiorum from routine diagnostics and review of the literature.

INTRODUCTION: Since its first description in 2003, clinical case reports have repeatedly documented that the bacterial species Kerstersia gyiorum is a human pathogen. K. gyiorum has been described in the context of wound and middle ear infections, has been found in urine samples, and has even been recognized as a causative agent of septicemia. However, the pathogenic potential of K. gyiorum cannot be clearly determined due to the current lack of information about this species. To reveal the clinical significance of K. gyiorum, it is therefore essential to first create a clearly defined referen

mGem: Innate immune sensing of HIV-1 transcription in myeloid cells-a tale of two transcription start sites.

Suppressive antiretroviral therapy (ART) in people with HIV (PWH) arrests progression to AIDS and onward viral transmission. However, viral transcription from integrated HIV-1 proviruses persists on treatment and is linked to markers of chronic immune activation, contributing to the accelerated development of comorbidities in PWH. Of particular interest are persistent responses in long-lived tissue-resident myeloid cells, which direct tissue-level inflammatory processes through cytokine secretion and interactions with other immune cell populations. Here, we review recent findings on viral nucl
