Overview of Chikungunya Virus Epidemiology, Biology, and Pathogenesis.

Chikungunya virus (CHIKV), an arthropod-borne alphavirus within the Togaviridae family, is transmitted primarily by Aedes aegypti and Aedes albopictus. The virus causes an acute febrile illness characterized by severe, often bilateral polyarthralgia, with potential progression to chronic musculoskeletal pain and rare systemic complications involving cardiovascular and neurological systems. CHIKV exhibits a spherical, enveloped virion (~70 nm) with T = 4 icosahedral symmetry, incorporating E1/E2 glycoprotein heterodimers that mediate receptor binding and membrane fusion. Its positive-sense RNA

RT-RPA-Assisted CRISPR/Cas12a-Based Isothermal Detection of Chikungunya Virus.

Chikungunya virus (CHIKV) is transmitted through the bite of Aedes mosquitoes, specifically A. aegypti and A. albopictus. CHIKV belongs to the alphavirus with a positive-sense ssRNA genome of 11-12 kb size. The virus has been reported from various geographical regions across the globe. Chikungunya fever is an acute febrile illness, which, if left untreated, may develop into chronic arthralgia that may persist for several months or acute encephalitis syndrome. Therefore, early diagnosis of CHIKV is crucial to initiate prompt supportive treatment. Laboratory diagnosis of CHIKV typically relies o

Screening of Antiviral Agents Against CHIKV Using Reporter Virus.

Chikungunya virus (CHIKV) causes a disease characterized by chronic musculoskeletal inflammation for which specific antivirals are not yet available. Currently, a supportive therapy to alleviate fever and pain is used, but it does not limit viral replication or the persistence of chronic arthritis symptoms. Thus, the identification and development of new active molecules against CHIKV is urgently needed. Here, we present a cell-based methodology that enables the implementation of a rapid and cost-effective strategy for high- and medium-throughput screening (HTS) of compounds, including repurpo

Chikungunya Virus Infection in Paraffin-Embedded Tissue: Analysis of Histological Alterations and Viral Detection by Immunohistochemistry.

Histopathological analysis of tissues infected with Chikungunya virus (CHIKV), including biopsy and autopsy specimens, can provide valuable insights into the pathogenesis of atypical and fatal cases. By examining tissue architecture and cellular alterations under the microscope, it is possible to identify patterns of injury, inflammation, and cellular degeneration. These morphological findings provide evidence of how the pathogen interacts with host cells and tissues. Moreover, immunohistochemistry may be performed in tissue sections for the detection of viral antigens. Importantly, this techn

Integrative Transcriptomic Network Modeling Coupled with Patient-Derived Validation for Circulating lncRNA Biomarker Discovery in NAFLD: A Comprehensive Workflow.

.: Nonalcoholic fatty liver disease (NAFLD) is a significant global health concern, impacting roughly 25% of people and leading to chronic liver conditions. It involves excess fat accumulation in the liver without significant alcohol intake and can develop into nonalcoholic steatohepatitis (NASH), fibrosis, or cirrhosis. While liver biopsy remains the gold standard for diagnosis, its invasive nature and associated risks restrict its routine use. Noninvasive biomarkers, such as serum ALT, AST, and various composite scores, are available; however, their clinical usefulness is often limited by va

In Silico Single-Cell Frame work for Modeling Intestinal Stem and Transit-Amplifying Progenitor Cells Dynamics.

Single-cell RNA sequencing (scRNA-seq) has revolutionized the ability to resolve cellular heterogeneity within complex tissues, enabling the identification of discrete cell states. Here, we present an in silico analytical pipeline designed to characterize intestinal stem cells (ISC), transit-amplifying (TA) progenitors, and BEST4⁺ enterocyte precursors from human scRNA-seq datasets, with a focus on inflammatory contexts such as inflammatory bowel disease (IBD). The pipeline integrates dataset acquisition, quality control, normalization, dimensionality reduction, unsupervised clustering, and ce

A Modelling Approach to Resolve Gene Temporal Dynamics in Intestinal Inflammation.

The intestinal epithelium is maintained by stem cells at the crypt base, however, acute or chronic inflammation can severely disrupt stem cell function and tissue homeostasis. To characterize mucosal inflammation and tissue damage, we evaluated time-course gene expression changes using a dextran sulfate sodium (DSS)-induced mouse model of colitis. By applying normalization, Z-score transformation, and spline curve modeling, we trace the temporal expression dynamics of key intestinal genes. This computational approach offer insights into the molecular responses during inflammation and may help

Mechanisms underlying rheumatic heart disease immune-mediated pathogenesis: T-cell responses, cytokine networks, and immune dysregulation as drivers of valvular injury.

Acute rheumatic fever is an immune-mediated sequela of Streptococcus pyogenes (group A Streptococcus) infection, presenting with fever, arthritis, chorea, rash, and, crucially, carditis. Without effective secondary prophylaxis and care, acute rheumatic fever can progress to chronic rheumatic heart disease, a leading yet under-recognised cause of cardiovascular morbidity and death that disproportionately affects children and young adults aged 5-24 years in low-resource settings. Robust evidence implicates the role of antigenic mimicry-particularly between streptococcal M protein and cardiac or

Labeling and Detection of Peptidoglycan in Chlamydia and Chlamydia-Related Organisms Utilizing D-Alanine Analogs.

The incorporation and visualization of modified D-amino acids into the peptidoglycan (PG) of actively replicating microbes have made a tremendous impact on the field of bacterial physiology. Here, we describe the optimization of this technique for labeling PG in obligate, intracellular Chlamydia species as well as Chlamydia-like organisms. Fluorescent D-amino acids (FDAAs) are D-alanine analogs that readily incorporate into the stem peptides of living bacteria and can be visualized rapidly with a fluorescence microscope without the need for laborious postfixation procedures. By comparison, D-a

Cost-effectiveness of the hepatitis B birth-dose vaccine in South Africa: a mathematical modelling analysis.

BACKGROUND: In 2022, South Africa had an estimated hepatitis B prevalence of 4·7-6·0%. In an effort to eliminate viral hepatitis as a public health threat, a targeted (ie, selective) hepatitis B birth-dose vaccination (HepB-BD) policy was introduced in 2023. Under this policy, only infants born to mothers with confirmed hepatitis B positivity are eligible for HepB-BD, which is given in addition to peripartum antiviral prophylaxis (PAP) for mothers from the second or third trimester of pregnancy. This policy differs from the WHO recommendation that all newborns should receive a HepB-BD vaccine

Quantification of Human T Cell Responses to Chlamydia trachomatis: Intracellular Cytokine Staining (ICS) and Activation-Induced Marker (AIM) Assays.

High-resolution characterization of antigen-specific T cell responses is essential for understanding cellular immunity to Chlamydia trachomatis infection in humans. This chapter describes two complementary flow cytometry-based approaches for measuring human antigen-specific T cell responses: intracellular cytokine staining (ICS) and activation-induced marker (AIM) staining. Both assays enable single-cell-level analysis following antigen stimulation but differ in their readouts. ICS quantifies functional cytokine production in response to short- or long-term stimulation, whereas the AIM assay i

Modulating effects of bee venom (Apis mellifera intermissa) against ethylene glycol-induced oxidative nephrotoxicity in male mice.

PURPOSE: Bee venom (BV) is a well-studied nephroprotective agent, however, its efficacy against ethylene glycol (EG)-induced nephrotoxicity has not been previously investigated. Therefore, this study aimed to assess the protective effect of BV in mitigating EG-mediated alterations in renal function markers, oxidative stress, and histopathological injury in male mice. METHODS: Renal toxicity was induced in adult male mice by daily oral gavage of 20% EG (2 mL/kg body weight). BV was administered intraperitoneally as a co-treatment over the same exposure period. Evalauted ndpoints included physi

Multimodality imaging for predicting regional wall motion recovery after percutaneous coronary intervention for chronic total occlusion.

BACKGROUND: Predicting regional left ventricular (LV) functional recovery is essential for optimizing patient selection for percutaneous coronary intervention (PCI) in chronic total occlusion (CTO), yet direct comparisons of imaging modalities remain limited. We aimed to compare the predictive performance of dobutamine stress echocardiography (DSE), cardiac magnetic resonance (CMR) imaging, and single-photon emission computed tomography (SPECT), alone and in combination, for regional LV wall motion recovery after PCI for CTO. METHODS AND RESULTS: We prospectively examined 66 patients with CTO

Assessment of Lurasidone Hydrochloride and Its Impurities for Neuronal and Cardiac Toxicities Using Integrated Toxicological Approaches.

Lurasidone hydrochloride (LH) has over 98 other impurities produced during its manufacturing and storage process. LH had two crucial impurities that were reported cytotoxic and genotoxic in nature: 4-chloro-1,2-benzisothiazole (Impurity 1) and 3-(1-piperazinyl)-1,2-benzisothiazole (Impurity 2). When the impurity threshold is exceeded due to chronic administration, evaluation of the toxicity of critical organs become crucial. A variety of endpoints were evaluated to assess the neurotoxicity of LH along with both impurities. The parameters related to neurotoxicity includes assessment of neurobeh

Bayesian Real-Time Decision Support for Strategy Optimization in Chronic Total Occlusion Percutaneous Coronary Intervention.

BACKGROUND: Complex percutaneous coronary intervention (PCI) for chronic total occlusion (CTO) is often prolonged, underscoring the need for timely strategy selection. Although several preprocedural scoring systems exist, they offer limited guidance regarding intraprocedural dynamics. This study aimed to develop a framework that updates success probabilities to support optimal strategy transitions during the procedure. METHODS AND RESULTS: Two endpoints were defined: (1) successful crossing using antegrade wire escalation (AWE); and (2) time-constrained success using either AWE (<15 min) or a
