Assessment of Lurasidone Hydrochloride and Its Impurities for Neuronal and Cardiac Toxicities Using Integrated Toxicological Approaches.

Lurasidone hydrochloride (LH) has over 98 other impurities produced during its manufacturing and storage process. LH had two crucial impurities that were reported cytotoxic and genotoxic in nature: 4-chloro-1,2-benzisothiazole (Impurity 1) and 3-(1-piperazinyl)-1,2-benzisothiazole (Impurity 2). When the impurity threshold is exceeded due to chronic administration, evaluation of the toxicity of critical organs become crucial. A variety of endpoints were evaluated to assess the neurotoxicity of LH along with both impurities. The parameters related to neurotoxicity includes assessment of neurobeh

Bayesian Real-Time Decision Support for Strategy Optimization in Chronic Total Occlusion Percutaneous Coronary Intervention.

BACKGROUND: Complex percutaneous coronary intervention (PCI) for chronic total occlusion (CTO) is often prolonged, underscoring the need for timely strategy selection. Although several preprocedural scoring systems exist, they offer limited guidance regarding intraprocedural dynamics. This study aimed to develop a framework that updates success probabilities to support optimal strategy transitions during the procedure. METHODS AND RESULTS: Two endpoints were defined: (1) successful crossing using antegrade wire escalation (AWE); and (2) time-constrained success using either AWE (<15 min) or a

Opportunities and Challenges for Promoting Sustainable Diets within "Food is Medicine" Programs in the United States: A Qualitative Research Study.

BACKGROUND: The United States population is experiencing concurrent, interrelated challenges of diet-related chronic diseases, food and nutrition insecurity, and climate change. Food is Medicine (FIM) has been described as a range of food-based programs that aim to prevent, manage, and treat diet-related chronic diseases in partnership with health care systems. Research is lacking in how these programs could promote plant-rich dietary patterns and other sustainability practices to support both human and planetary health. OBJECTIVES: This study explored how FIM experts in the United States def

Conventional transcutaneous electrical nerve stimulation shows no short-term safety signal in painful cervical dystonia: a pilot prospective study.

BACKGROUND AND PURPOSE: Cervical dystonia (CD) is frequently associated with chronic cervical pain refractory to botulinum neurotoxin (BoNT) therapy. Transcutaneous electrical nerve stimulation (TENS), a non-invasive neuromodulation technique with established analgesic properties, has not been formally evaluated in CD. This pilot study assessed the safety and tolerability of conventional TENS in painful CD, with exploratory evaluation of pain outcomes. METHODS: This prospective, observational, single-arm study (NCT04949594) included patients with painful CD treated or not with BoNT. The prima

Computational identification of candidate biomarkers and compounds associated with skeletal muscle insulin sensitivity improvement mediated by physical activity: an integrative multi-omics and computational chemistry study.

BACKGROUND: Insulin resistance (IR) underlies many chronic diseases. While physical activity improves skeletal muscle insulin sensitivity (IS), the underlying molecular mechanisms and targeted drug strategies remain elusive. This hypothesis-generating study integrates bioinformatics and computational chemistry using public datasets; all findings are computational predictions requiring experimental validation. METHODS: Using a skeletal muscle dataset (GSE24215, n = 10) of bed rest-induced IR and exercise-reversed IR, candidate genes were identified via WGCNA and differential expression analysi

Ecological risk of substituted p-phenylenediamines and quinones in Chinese freshwaters using QSAR-ICE modeling.

The high ecotoxicity of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPD-Q) has directed global attention to substituted p-phenylenediamines (PPDs) and their quinone derivatives (PPD-Qs). To date, however, comprehensive evaluations of their toxicity and associated ecological risks in aquatic ecosystems remain scarce. This study utilized an integrated approach combining quantitative structure-activity relationship and interspecies correlation estimation (QSAR-ICE) model predictions, species sensitivity distribution analysis, and ecological risk assessment to evaluate the potenti

NaV1.7, 1.8 and 1.9 mutations linked to painful channelopathies have no pain phenotype in UK Biobank.

The voltage-gated sodium channels NaV1.7, NaV1.8 and NaV1.9, encoded by the SCN9A, SCN10A and SCN11A genes, respectively, are integral to nociceptor excitability and pain sensation. Multiple gain-of-function mutations in these ion channels have been reported to cause autosomal dominant painful channelopathies, including primary erythromelalgia, paroxysmal extreme pain disorder and familial episodic pain. Conversely, autosomal recessive loss-of-function mutations in NaV1.7 have been reported to cause painless channelopathies. The prevalence and impact on carriers of these voltage-gated sodium c

[Digital transformation of anxiety and depression in adolescents: Phenomenology, neurobiology, clinical challenges].

Although numerous studies have examined digital activity's impact on mental health, how the digital environment transforms the phenomenology of mental disorders remains insufficiently understood. This narrative review synthesizes evidence on how the digital environment influences the clinical manifestations of anxiety and depression in adolescents. Digital phenomena such as social comparison, fear of missed opportunities, problematic use of social networks and internet games, and doomscrolling not only accompany affective symptoms but also become integral behavioral and cognitive components, s

Single-Item Self-Rated Health Question for Assessing General Health: A Population-Based Study Among Kurdish Population.

BACKGROUND: Single-item self-rated health (SRH) is widely used as a brief measure of perceived health because of its simplicity and feasibility in population-based research. However, its agreement with a reference measure and the extent to which its performance varies across sociodemographic and health-related subgroups remain important considerations. This study evaluated the performance of single-item SRH against the 28-item General Health Questionnaire (GHQ-28) reference criterion among adults participating in the Dehgolan Prospective Cohort Study (DehPCS) in Iran. METHODS: This cross-sect
