Engineering Marker-Free Lettuce Chloroplast Genome to Express Functional Glucagon-Like Peptide-1 Receptor Agonists Exenatide and Lixisenatide.

Diabetes Mellitus is an epidemic affecting > 500 million, claiming 6-7 million lives annually. Chemically synthesised Glucagon-like peptide-1 receptor agonists (GLP-1RAs) containing artificial amino acids reduce haemoglobin A1c and obesity but are not yet affordable and require invasive injections. High dosage requirement and gastrointestinal complications are among the current limitations of oral GLP-1RAs. Therefore, we expressed codon optimised Exenatide and Lixisenatide fused with Cholera-toxin B-subunit (CTB) in lettuce chloroplasts to facilitate their oral delivery, increase affordability

Analysis of the Short- and Long-Term Immune Response in BALB/c Mice Immunized with Total Naegleria fowleri Extract Co-Administered with Cholera Toxin.

BACKGROUND: Naegleria fowleri is a free-living amoeba that inhabits warm freshwater and causes primary amoebic meningoencephalitis (PAM), a rapidly fatal infection with >95% mortality. Due to the lack of early diagnosis and effective therapy, preventive vaccination represents a promising strategy. METHODS: This study evaluated short- and long-term immune protection in BALB/c mice (20 mice per group) immunized intranasally with total N. fowleri extract co-administered with cholera toxin (CT). Mice were challenged with a lethal dose of trophozoites either 24 h (short-term) or three months (long

Altered gut microbiota, SCFAs, and barrier integrity markers in Alzheimer's and Parkinson's disease patients.

Parkinson's disease (PD) and Alzheimer's disease(AD) have significant gut-brain axis interaction via microbial dysbiosis. In this study, Iranian patients with PD (n = 25), ad (n = 25), and neurological disorders (ND, n = 20) were compared with healthy controls (HC, n = 20) in terms of gut microbiota abundance, short-chain fatty acid levels, and gut inflammation markers (calprotectin and zonulin). Stool and blood samples were collected from all participants and analyzed using real-time PCR, high-performance liquid chromatography, and enzyme-linked immunosorbent assay. Significant reductions in

Phylogeny-guided multi-epitope vaccine design targeting conserved and divergent regions of Brucella OMP2, OMP25, and OMP31: an immunoinformatics study.

Brucella is a zoonotic intracellular bacterial pathogen. Currently, no human vaccines for brucellosis are approved, and only animal vaccines are available. Despite the widespread use of live attenuated vaccines, several drawbacks have prompted investigation into outer membrane proteins (OMPs) as potential vaccine candidates. This study employs immunoinformatics to design a multi-epitope vaccine targeting conserved regions of the OMP2a, OMP2b, OMP1, OMP25, and OMP31 antigens against four common Brucella species. Helper T-lymphocyte (HTL), Cytotoxic T-lymphocyte (CTL), and B-cell epitopes of OMP

Fusobacterium nucleatum Enhances Intestinal Adaptation of Vibrio cholerae via Interspecies Biofilm Formation.

Biofilm formation represents a key survival strategy employed by Vibrio cholerae to adapt to the complex intestinal environment of the host. While most previous studies on V. cholerae biofilms have focused on genetic regulation and monospecies cultures, its ability to form dual-species biofilms with other intestinal pathogens is still poorly understood. In this study, using samples from both cholera patients and healthy individuals, Fusobacterium nucleatum was identified as a bacterium capable of co-aggregating with V. cholerae. Untargeted metabolomic analysis revealed that F. nucleatum-derive

Forging ophthalmology in Puerto Rico: Dr Ramón Emeterio Betances (1827-1898).

Ramon Emeterio Betances (1827-1898) stands out as a pivotal figure in the fields of ophthalmology and public health in nineteenth-century Puerto Rico. Trained in France, he returned to the island in 1856, where he treated cholera patients and introduced vital public health interventions. Known as 'The Physician of the Poor', Betances dedicated himself to serving marginalised populations, including enslaved individuals. He co-founded a secret abolitionist society and championed Puerto Rican independence, actions that resulted in multiple periods of exile. Together with his contemporaries, Betan

Dissemination of public health information during the global cholera outbreak response: WHO's experience.

Since 2022, the seventh cholera pandemic has escalated globally, with reported cases rising from 223 370 in 2021 to 560 823 in 2024 across affected countries. Cholera-related deaths also increased significantly, reaching 6028 in 2024. These figures likely underestimate cholera's true burden due to surveillance and reporting challenges. In response, the WHO classified the resurgence as a Grade 3 emergency in January 2023, activating its Incident Management System for a global response. To provide updates on cholera trends, public health responses and challenges, the WHO published 33 Rapid Risk

Tunable triazole-based cholera toxin inhibitors: A QSAR-guided design and evaluation approach.

Cholera toxin B subunit (CTB) is a validated target for anticholera therapeutics, but current inhibitors often suffer from synthetic complexity and limited tunability. This study aimed to develop a compact, tunable triazole scaffold exhibiting low-micromolar potency combined with high synthetic tractability. We applied a unified, QSAR-driven, multiscale computational-experimental workflow integrating molecular docking, induced fit docking (IFD), molecular dynamics (MD), QM/MM calculations, and descriptor-based QSAR modeling to prioritize a focused library of 44 N-sulfonyl triazole inhibitors.

Drought and armed conflict as drivers of overcrowding in Sudan's refugee camps.

Forced migration is a result of variable changes, including human-made factors, environmental factors or both. Climate change has been a significant cause of increased forced migration in Sudan. The prolonged drought attributed to climate change affects Sudanese citizens, with 80% of the population relying on agriculture and pastoralism as their main source of income. Since April 2023, the conflict has displaced over 14.5 million people. Four million have sought safety in neighbouring countries. The refugee camps lack essential resources, and the displaced people face a health crisis, with out

Aeromonas in South Asia: genomic insights into an environmental pathogen and reservoir of antimicrobial resistance.

Aeromonads are an ecologically versatile group of bacteria that cause infections in aquatic animals and are recognised as emerging human pathogens. Despite this, our understanding of Aeromonas diversity, especially the relationship between clinical and environmental strains, remains limited. Here, we present a genomic analysis of the Aeromonas genus, comprising 1853 genomes, and a detailed comparison of clinical and environmental strains from South Asia, including 996 newly sequenced genomes from Bangladesh and India. Phylogenetic analyses revealed that Aeromonas is a highly diverse genus, wit

In-silico identification of anti-cholera phytochemicals from Indian medicinal plants.

Cholera is a severe diarrheal disease caused by ingestion of food or water contaminated with pathogenic Vibrio cholerae. Treatment for cholera includes rehydration therapy and antibiotics to avert death and reduce bacterial burden to prevent rapid transmission of the disease. In addition, in Indian subcontinent, there is historical evidence of using plants for treating cholera. This study was designed to investigate the cholera toxin-inhibitory properties of phytochemicals sourced from Indian medicinal plants. For this, three reported genotypes of cholera toxin subunit B (ctxB) associated with

Fractional nonlinear dynamics and forward bifurcation in a memory-based cholera model.

This study explores the use of fractional-order epidemic models to capture the memory-dependent and nonlinear behaviors inherent in cholera transmission. We present a fractional-order susceptible-infected-recovered-individuals adopting preventive measures-bacteria model that integrates preventive behavior and environmental feedback. By applying the Caputo derivative, we demonstrate the existence, uniqueness, and boundedness of the model's solutions and derive an analytical expression for the basic reproduction number R0. Our stability and bifurcation analyses show how memory influences the sys

TseVF-TsiVF, a novel bacteriolytic effector-immunity pair of Vibrio fluvialis VflT6SS2, provides a fitness advantage in microbial competition.

Vibrio fluvialis (V. fluvialis) is a halophilic Gram-negative bacterium regarded as an emerging pathogen that can cause severe cholera-like diarrhea and various extraintestinal infections. Type VI secretion systems (T6SSs) are nano-weapons used by bacteria to deliver toxic effectors into host cells or antagonize competitors. VflT6SS2 of V. fluvialis is involved in bacterial pathogenicity and competitive environmental survival. Previously, we have identified some regulatory factors of VflT6SS2 and a pesticin domain-containing effector, TssI2. Here we reported a novel effector-immunity pair of V

A Vibrio-susceptibility class of antimicrobial peptide Ajapocin via membranolytic pattern to combat "non-cholera" pathogens in vivo infection models.

Pathogenic "non-cholera" Vibrio species of Vibrio parahaemolyticus (V. parahaemolyticus) and Vibrio vulnificus (V. vulnificus) frequently pose a serious threat to aquaculture security and public health by causing infectious diseases. In this study, we reported the discovery of a marine-sourced antimicrobial peptide (AMP) called Ajapocin, which identified through a sequence optimization strategy. Ajapocin exhibited potent activity against V. parahaemolyticus and V. vulnificus pathogens, with minimum inhibitory concentrations (MICs) of 6-12 μM-comparable to the clinical agent Polymyxin B (PMB).

Antimicrobial resistance and genomic characteristics of avian Pasteurella multocida.

BACKGROUND: Pasteurella multocida, the cause of fowl cholera, poses a significant economic threat to poultry. The escalation of antimicrobial resistance (AMR) in this pathogen, driven by drug misuse, is a serious concern. Crucially, a comprehensive understanding of AMR patterns, molecular epidemiology, and virulence in Chinese avian strains is still lacking. This study aimed to elucidate the phenotypic and genomic characteristics of P. multocida strains isolated from avian in Zhejiang Province, China. RESULT: Twenty avian-origin P. multocida strains isolated in Zhejiang Province were subjecte

Co-administration of oral killed whole-cell recombinant cholera toxin B-subunit vaccine (WC-rCTB) and live Salmonella Typhi Ty21a vaccine: a prospective randomized open-label trial.

BACKGROUND: Cholera and typhoid fever are often co-endemic, making vaccine co-administration practical. However, due to lack of immunogenicity data, current guidelines advise against co-administration of the oral inactivated whole-cell recombinant cholera toxin B-subunit vaccine (WC-rCTB) and the oral live Salmonella Typhi Ty21a vaccine. METHODS: Healthy adults (18-65 years) were randomized 1:1:1 to receive WC-rCTB with Ty21a (group Ch + Ty), WC-rCTB alone (group Ch) or Ty21a alone (group Ty). Peripheral blood mononuclear cells (PBMCs) were isolated on Days 0, 5 and 7 from all, plus on Days 1

Event-based surveillance in the WHO African region: baseline implementation before the COVID-19 pandemic.

Event-based surveillance (EBS) is a critical component of epidemic intelligence, supporting the timely detection of unusual public health events. However, prior to the COVID-19 pandemic, the status of EBS implementation across the WHO African region was not well documented. This study establishes a regional baseline of EBS modalities in place before the pandemic to inform future surveillance system evaluation and strengthening. We conducted a cross-sectional survey across all 47 WHO Member States in the African region between December 2019 and February 2020. A structured questionnaire was diss
