Robot-Assisted Radical Prostatectomy in a Post-Enucleation Gland.

PURPOSE: Holmium laser enucleation of the prostate (HoLEP) is an established treatment for benign prostatic obstruction, and some patients subsequently prove to have prostate cancer (1-3). Enucleation leaves fibrosis that displaces the anatomical landmarks relevant to radical prostatectomy (4, 5). We present a robot-assisted radical prostatectomy (RARP) performed after HoLEP. MATERIALS AND METHODS: A 64-year-old man underwent HoLEP; biopsy subsequently showed adenocarcinoma of the right lobe, ISUP grade 3. PSA was 0.51 ng/mL; MRI demonstrated a 0.4 cm PI-RADS 4 lesion at the right mid-base. R

Sleep duration and prostate cancer-specific mortality in the Southern Community Cohort Study.

PURPOSE: Sleep patterns may affect metabolic and behavioral processes, which subsequently could be linked to cancer mortality. However, the relationship between sleep patterns and prostate cancer-specific mortality remains unclear. We evaluated the association of sleep and prostate cancer-specific mortality and investigated whether race/ethnicity influences the association. METHODS: The Southern Community Cohort Study (SCCS), a prospective cohort study, recruited participants from 12 Southeastern states between March 2002 and September 2009. The cohort included 34,166 males, predominantly Bla

[PSMA-PET/CT SCAN WAS USEFUL IN DIFFERENTIATING PRIMARY URETERAL CANCER FROM METASTATIC ONE IN A PROSTATE CANCER WITH LEFT HYDRONEPHROSIS: A CASE REPORT].

A 70 year-old male patient with a history of prostate adenocarcinoma developed left hydronephrosis 11 years after receiving heavy particle therapy. Computed tomography (CT) revealed possible left primary ureteral cancer. However, this lesion was positive on prostate-specific membrane antigen positron emission (PSMA-PET) /CT, suggesting metastasis from prostate cancer. After three months of androgen deprivation therapy (ADT), the lesion became negative on PSMA-PET/CT. Six months after initiation of ADT, the left hydronephrosis improved. These findings suggest that PSMA-PET/CT help differentiate

FOXG1 Enhances Paclitaxel Resistance in Prostate Cancer via NANOG-Mediated Cellular Stemness.

BackgroundPaclitaxel (PTX) is an effective chemotherapeutic agent for prostate cancer (PCa), yet resistance emerges with disease progression. Cancer-cell stemness is intimately linked to PTX resistance. This work clarifies whether FOXG1 modulates PTX resistance in PCa via cellular stemness.MethodsTCGA database was interrogated for FOXG1 expression in PCa. PTX-resistant PCa cell lines were generated by stepwise escalation of PTX concentration. qRT-PCR quantified FOXG1 and NANOG levels in PCa cells. CCK-8, sphere-formation assay, and flow cytometry evaluated cell viability, stemness, and apoptos

Dual-Targeting Nanoplatform Integrating Metabolic Glycan Labeling and Bioorthogonal Chemistry for Precise Delivery of Oxaliplatin to PSMA-Positive Prostate Cancer.

Platinum-based chemotherapy drugs are widely used in the treatment of various cancers due to their broad antitumor spectrum, but their clinical application is severely limited by significant systemic toxicity resulting from nonspecific cytotoxicity. Prostate-specific membrane antigen (PSMA), which is highly overexpressed in most prostate cancer (PCa) cells, presents a promising target for tumor-selective drug delivery. In this study, we designed and synthesized a novel dual-targeting nanoplatform that combines metabolic glycan labeling and bioorthogonal chemistry for the precise delivery of ox

Development and External Validation of a Machine Learning-Derived Risk Stratification Score for Metastatic Castration-Resistant Prostate Cancer.

PURPOSE: Metastatic castration-resistant prostate cancer (mCRPC) shows marked clinical heterogeneity and poor long-term survival, highlighting the need for simple and reliable risk stratification tools beyond the limited usability of existing complex prognostic models. In this study, we developed and validated PROGRESS (Prostate Cancer Global Risk Evaluation and Stratification Score), a simplified prognostic score derived through machine learning‑guided feature selection distilled into a simple laboratory-based tool. METHODS: Baseline data from 2,035 patients with mCRPC (training set) were em

MINDIN-Induced Attenuation of Primary Cilia Activates Parathyroid Hormone Receptor Type I (PTH1R) to Drive Osteomimicry and Bone Tropism in Prostate Cancer.

Advanced prostate cancer metastasizes primarily to bone, a process driven in part by the de novo expression of bone-associated factors (osteomimicry) that enables tumor cells to colonize and thrive in the skeletal environment. During the early stages of tumorigenesis in multiple cancer types, the primary cilium, a non-motile microtubule-based sensory organelle, is lost. However, the causes of primary cilium loss and the consequences of this process for tumor progression are still ill-defined. The aim of this study was to determine whether MINDIN, a protein secreted by prostate tumor cells, pro

From a Cochineal Dye to the Possibility of an Anticancer Agent: Free Radical-Mediated Activity of Carminic Acid and Its CoII Complex on Nucleobases, DNA, and PC3 Prostate Carcinoma Cells.

Anthracycline-based anticancer agents perform through different modes; the free radical pathway is just one of them. Although they are effective anticancer agents, for different reasons, there is a lookout for simpler analogs that mimic their actions. Carminic acid, having a close resemblance to anthracyclines, and its CoII-complex were investigated for activity related to the generation of semiquinone. The compounds were reduced at their respective reduction potentials, which were previously identified using cyclic voltammetry. Interactions between reduced species of the compounds and biologi

GSTP1 promoter hypermethylation in prostate cancer detection: current evidence, clinical positioning, and translational gaps.

GSTP1 promoter hypermethylation is one of the most consistently documented molecular alterations in prostate cancer, with high prevalence in malignant tissue and a well-established relationship with transcriptional silencing. However, despite more than two decades of biological and diagnostic investigation, GSTP1-based testing has not acquired a clearly defined stand-alone role in contemporary prostate cancer detection. This narrative review examines why extensive biological evidence has not translated into routine clinical use and where, if anywhere, GSTP1 testing might provide meaningful val

Impact of reduced scan extent on tumor detection in 68Ga-PSMA PET/CT for biochemical recurrence of prostate cancer.

BACKGROUND: PSMA PET/CT plays an important role in biochemically recurrent prostate cancer (BCR-PCa). In general, PET/CT is performed as a limited or true whole-body examination. The majority of PET/CT scanners operate by measuring several bed positions or continuous motion with a limited axial field of view (FOV). Shortening the scan area thus results in a time saving, with unknown information loss regarding the detection of tumor manifestations. METHODOLOGY: This retrospective single-center study included 200 patients with BCR-PCa after initial curative therapy who underwent a 68Ga-PSMA-PET

Clinical impact of PSMA PET/CT on the management of biochemical recurrence in prostate cancer.

BACKGROUND: Ga-68 prostate-specific membrane antigen (PSMA) positron emission tomography/computed tomography (PET/CT) improves lesion detection in patients with biochemical recurrence (BCR) of prostate cancer. However, real-world evidence linking imaging-driven management changes to early biochemical outcomes remains limited, particularly in Korean populations. This study aimed to evaluate the impact of Ga-68 PSMA PET/CT on clinical management and its association with early biochemical response. METHODS: This study included retrospectively registered 75 patients who underwent Ga-68 PSMA PET/C

High-throughput droplet-based enzymatic DNA methylome profiling enables single-cell resolution of epigenetic heterogeneity.

DNA methylation is a fundamental epigenetic modification that regulates gene expression, cellular identity, and disease-associated regulatory programs. However, high-throughput single-cell DNA methylation technologies have been limited by DNA degradation, low CpG recovery, and the poor scalability of bisulfite-based methods. Here, we present scEM-seq, a droplet-based single-cell enzymatic methylome sequencing method that integrates barcode ligation within a single emulsification step, followed by postemulsion cytosine-to-uracil conversion. This design substantially reduces DNA loss and enables

Comparison of short-term oncological and functional outcomes between the da Vinci and toumai platforms in robotic radical prostatectomy.

Robot-assisted radical prostatectomy (RARP) is a standard treatment for localized prostate cancer (PCa). As new, lower-cost robotic platforms reach Latin America, comparative evidence remains scarce. We compared complications, and short-term oncological and functional outcomes between the da Vinci and the new Toumai platforms in RARP. Single-center, prospective, non-randomized study of patients older than 40 years with localized PCa undergoing RARP between June 2025 and July 2026. Transformer-Based Deep Learning Integrating Multi-Regional MRI and Clinical Features for Predicting Biochemical Recurrence After Radical Prostatectomy: A Multicenter Study.

BACKGROUND: Existing deep learning models for predicting biochemical recurrence (BCR) after radical prostatectomy often ignore the biological context beyond the tumor. This study aims to develop and validate a transformer-based model combining multi-regional magnetic resonance imaging (MRI) features and clinical data to improve BCR prediction after radical prostatectomy in prostate cancer (PCa). METHODS: We enrolled 232 Pca patients from three medical centers between 2013 and 2020.
