Treatment of patients referred with Peyronie's disease.

INTRODUCTION: Peyronie's disease (PD) is a benign penile disorder characterised by fibrotic plaques, leading to curvature and potential sexual dysfunction. It often coexists with diabetes, erectile dysfunction and cardiovascular disease. In Denmark, patients are referred by general practitioners (GPs) to public or private hospitals under a treatment guarantee ensuring care within 30 days. METHODS: We conducted a retrospective study of PD patients referred by GPs to Aarhus University Hospital in 2019. Data were retrieved from the Central Denmark Region Business Intelligence Portal. Patient cha

Diagnostic and treatment delays in penile cancer: a call to improve awareness and referral.

INTRODUCTION: In men with penile cancer (PeCa), timely referral is crucial for optimizing clinical outcomes. We investigated the association between time from initial presentation to first surgical treatment and its effect on patient outcomes. MATERIALS AND METHODS: Data from a single center prospectively maintained database of patients with PeCa who underwent primary surgery between 2010 and 2024 were retrospectively analyzed, excluding those treated elsewhere. We assessed time intervals from initial disease appearance to referral across general practitioners (GP), urology or dermatology spec

CD47 as a prognostic biomarker and potential immunotherapy target in penile squamous cell carcinoma.

Penile squamous cell carcinoma (PSCC) remains a global healthcare concern, characterized by poor outcomes and limited therapeutic options. CD47, a “don’t-eat-me” signal that enables tumor immune evasion, has not been extensively evaluated in PSCC. This study aims to explore the expression and significance of CD47 in PSCC. RNA-sequencing was performed on eight paired PSCC and adjacent normal tissues. Protein expression was assessed by immunohistochemistry (IHC) in an independent of 131 PSCC cases, and associations with clinicopathological factors and prognosis was evaluated. In addition, the in

GPS1 Exon 9 Mutations Represent a Rare Genetic Event in Penile Squamous Cell Carcinoma Pathogenesis.

Penile squamous cell carcinoma (PSCC) is rare, but a biologically aggressive malignancy. Recent comprehensive genomic profiling (CPG) efforts revealed the underlying genomic landscape of PSCC, identifying TP53, TERT, CDKN2A, PIK3CA, NOTCH1, and FAT1 as frequently altered genes with potential roles in penile oncogenesis. In addition, recurrent mutations encoded in the GPS1 gene have been observed in 7.4% of cases in a particular PSCC cohort. Functional studies demonstrated loss of function due to GPS1 Exon 9 missense mutations, proposing a possible role for these alterations as oncogenic driver

A simple scoring system for predicting inguinal lymph node involvement and survival in patients with penile squamous cell carcinoma.

PURPOSE: Currently, no standardized predictive model is widely adopted in clinical practice for assessing outcome and lymph node metastasis (LNM). This study aimed at predicting inguinal lymph node involvement and mortality in patients suffering penile squamous cell carcinoma (PSCC) by developing a simple and clinically applicable scoring system. METHODS: Clinical and histopathological data from 11 centers, collected between 2000 and 2021, were reclassified (the Eighth Edition of the AJCC Cancer Staging Manual). Data from 8 centers (517 patients) were randomized into training and internal val

MicroRNAs in penile cancer: challenges, opportunities, and translational perspectives.

MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression, and dysregulated miRNA expression has been implicated in multiple cancer hallmarks. In penile cancer (PeCa), a rare but aggressive malignancy, evidence suggests that miRNAs contribute to tumor development and disease progression. This review aims to summarize the current understanding of miRNA involvement in PeCa, highlighting their potential as diagnostic, prognostic, and therapeutic biomarkers. A comprehensive review of published studies on miRNA expression in PeCa was performed, with an emphasis on molecular mechani

Association of CCL21+ CAFs with B-cell recruitment and TLS maturation in penile squamous cell carcinoma.

Penile squamous cell carcinoma (PSCC) is an aggressive malignancy with a poor prognosis and limited therapeutic options. Tertiary lymphoid structures (TLSs) can support antitumour immunity, yet TLS maturation and maturation-associated stromal determinants in PSCC remain unclear. Here, we integrated hematoxylin and eosin (H&E) staining, multiplex immunohistochemical (mIHC) colocalization, spatial transcriptomics, single-cell RNA sequencing (scRNA-seq), and bulk RNA sequencing (bulk RNA-seq) to profile TLS maturity and TLS-associated stromal-immune features in PSCC. TLS maturity was quantified u

Multidisciplinary Care of Patients With Penile Cancer.

Penile squamous cell carcinoma (PSCC) is a rare malignancy that displays profound disparities in incidence, access to care, and clinical outcomes rooted in economic, social differences, and cultural practices. PSCC has been linked to high treatment-related morbidity, social stigma, and psychological distress. This chapter provides an overview of this disease, integrating epidemiology, pathology, imaging, and multimodal management strategies. The principles of penile cancer staging, histopathologic features, and the role of imaging in local staging of the primary tumor and nodal disease are als

How do we counsel patients on short- and long-term complications after hypospadias repair? - A survey study.

INTRODUCTION: Hypospadias correction remains one of the most performed pediatric urologic procedures, affecting up to 1/150 males born in the United States. Current studies suggest that surgical counseling has a significant impact on shared decision making, decisional regret, and long-term follow-up. However, no set paradigm currently exists for long-term follow-up or counseling. We sought to obtain consensus from established pediatric urologists the optimal content and potential short, intermediate, and long-term complications to be considered when counseling patients and parents of patients

Angiomatous Subtype of Penile Kaposi Sarcoma With Concurrent HIV-Negative Multicentric Castleman Disease: A Case Report.

Kaposi sarcoma is a vascular neoplasm driven by human herpesvirus 8 (HHV-8), commonly seen in the setting of immunosuppression, such as HIV/AIDS or posttransplant therapy. The presentation of Kaposi sarcoma in HIV-negative patients with multicentric Castleman disease is exceedingly rare, particularly with uncommon histology subtypes and involvement of the penile shaft. A 58-year-old HIV-negative man with a history of HHV-8-associated multicentric Castleman disease presented with a violaceous lesion on the buttock, followed by a crusted ulcer on the penile shaft. Histopathologic evaluation of t

Topical steroids are effective even in severe phimosis: Evidence from a multicenter cohort.

Phimosis is commonly encountered in pediatric surgical practice. Despite evidence supporting topical corticosteroids for phimosis, many clinicians believe they are ineffective in certain subgroups such as severe phimosis or older children, leading to early circumcision. This study aimed to evaluate the effectiveness of topical corticosteroids across different severities and patient characteristics. We conducted a prospective multicenter cohort study across 12 Chilean hospitals between June 2023 and August 2024. Boys under 18 years old with phimosis grades 2 to 5 (Kikiros classification) were t

The identification of clinicopathological features predictive of recurrence following lymphadenectomy in penile cancer: A systematic review.

Recurrence following inguinal and/or pelvic lymph node dissection (I/PLND) for penile squamous cell carcinoma (PSCC) is associated with poor survival, yet predictors of post-I/PLND recurrence and optimal salvage strategies remain unclear. We conducted a systematic review to identify clinicopathological features predictive of recurrence after I/PLND. This systematic review followed PRISMA 2020 guidelines and was prospectively registered on PROSPERO. Major bibliographic databases were searched from inception to November 13th 2025. Eligible studies evaluated clinicopathological factors associated
