Ginseng polysaccharides prevent mastitis through Lactobacillus murinus-derived deoxycholic acid and TGR5 signaling.

Mastitis, a common inflammatory disease of breast tissue, has been linked to gut microbiota dysbiosis; however, evidence establishing a causal relationship and the potential for microbiome-based therapies remains limited. This study investigated the gut-mammary gland axis and how the prebiotic candidate ginseng polysaccharide (GP) prevents mastitis. Using Lipopolysaccharide (LPS)-induced mouse models, we demonstrate that mastitis involves gut dysbiosis contributing to pathogenesis. GP prevented mastitis in a microbiota-dependent manner. Mechanistically, GP selectively enriches Lactobacillus (L

Surgical resection versus stereotactic body radiotherapy for oligometastatic breast cancer to the liver.

Comparative evidence for hepatic resection and stereotactic body radiotherapy (SBRT) in breast cancer liver oligometastases remains limited. We compared survival, progression, and recovery after these treatments. This three-center retrospective study included adults with breast cancer and ≤ 3 liver metastases undergoing hepatic resection or SBRT between 2010 and 2023. Patients with uncontrolled extrahepatic disease were excluded. Propensity score matching generated 1:1 treatment pairs. Overall survival (OS) was the primary endpoint. Adjusted Cox models evaluated OS and progression-free surviva

Psychological determinants of pain, fatigue, and symptom burden in breast cancer survivorship.

Breast cancer survivors frequently experience persistent pain, cancer-related fatigue, and interconnected symptom clusters that impair quality of life, functional status, and treatment adherence. Psychological factors, including pain catastrophizing, anxiety, depression, fear-avoidance behaviors, and maladaptive cognitive appraisals, are associated with the amplification and maintenance of these symptoms through proposed mechanisms involving central sensitization, HPA axis dysregulation, inflammatory signaling, and behavioral deconditioning. This narrative overview, rather than a systematic re

8th International consensus guidelines for the management of advanced breast cancer (ABC8 guidelines).

The Advanced Breast Cancer (ABC) 8th international consensus conference took place on 6-8 November 2025, in Lisbon, and updated the previous ABC international consensus guidelines that are described in this manuscript. These guidelines reflect the work of a large and global consensus panel, comprising healthcare professionals and patients, and provide recommendations on how to best manage patients with advanced breast cancer (inoperable locally advanced or metastatic), of all breast cancer subtypes. The guidelines are based on the best available evidence or on expert opinion when a higher leve

Targeting PRDX1-G6PD Interaction Promotes a Metabolic Vulnerability and Redox Imbalance in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited targeted options, and its progression is strongly driven by metabolic reprogramming and redox homeostasis. Here, we found that peroxiredoxin-1 (PRDX1) promotes TNBC progression by directly interacting with glucose-6-phosphate dehydrogenase (G6PD), the rate-limiting enzyme of the pentose phosphate pathway (PPP). Mechanistically, PRDX1 depletion reduced the formation of active G6PD dimers without affecting its expression, thereby impairing PPP metabolism and ultimately suppressing TNBC growth. Furthermore, w

Patterns of social determinants of health screening and health-related social needs among patients with newly diagnosed breast cancer in a safety-net system.

PURPOSE: Various social determinants of health (SDOH) have been shown to negatively impact breast cancer outcomes. Though there are recommendations for comprehensive SDOH assessment, little is known about the optimal frequency of reassessments and change in health-related social needs (HRSN) over time for specific patient populations. We sought to characterize SDOH screening and HRSN patterns among patients with breast cancer in a safety-net system. METHODS: A retrospective study was conducted to evaluate SDOH survey responses in the electronic medical records and screening frequency from pat

Surgical management in HER2 + de novo metastatic breast cancer: disease progression and survival.

PURPOSE: Advancements in human epidermal growth factor (HER)-targeted therapy have significantly improved survival in patients with de novo metastatic HER-2 positive (HER2+) breast cancer (dnMBC). The role of breast surgery is debated. We examined which HER2 + dnMBC patients undergo surgery and compared outcomes to systemic therapy alone. METHODS: We performed a single institution retrospective review of patients with HER2 + dnMBC diagnosed 2011-2023. Data included demographics, clinicopathologic details, treatment information, local and distant disease progression, and survival outcomes. Ana

National trends in dual de-escalation of axillary surgery and adjuvant radiation in early-stage HR+/HER2- breast cancer.

PURPOSE: De-escalation has become standard across breast cancer management. Many early-stage HR+/HER2-negative breast cancers are eligible for both SLNB omission and accelerated partial breast irradiation (APBI). However, the oncologic safety of dual de-escalation has not been evaluated except in patients ≥ 70 years. We sought to characterize national practice patterns and factors associated with adoption of this dual de-escalation approach. METHODS: Women aged 50-69 years with clinical T1-T2N0 breast cancer who underwent upfront breast-conserving surgery between (2012 - 2022) were identified

Ferroptosis in Breast Cancer: Linking Iron Metabolism, Intertumoral Heterogeneity, and Predictive Biomarkers of Clinical Response.

Breast cancer (BC) is a prevalent and heterogeneous neoplasm whose therapeutic resistance remains a major challenge. Ferroptosis, an iron-dependent form of regulated cell death that is characterized by lipid peroxidation, has emerged as a key mechanism linking iron metabolism, oxidative stress, and tumor aggressiveness. The objective for this narrative review was to summarize the principal mechanisms regulating ferroptosis in BC and discuss its potential as a therapeutic target. A literature review was conducted to investigate the mechanisms involved in ferroptosis in BC, with emphasis on iron

Delayed and selective bone health assessment following cancer diagnosis: a real-world gap in survivorship care.

BACKGROUND: Cancer treatment-induced bone loss (CTIBL) is well-recognized in hormone-sensitive malignancies such as breast and prostate cancer but is less consistently implemented in patients with other solid tumors and hematologic malignancies. Furthermore, the long-term skeletal effects of newer systemic therapies, including immune checkpoint inhibitors and targeted agents, remain incompletely characterized. Although bone mineral density (BMD) assessment by dual-energy X-ray absorptiometry (DXA) is recommended for patients at increased skeletal risk, real-world utilization patterns remain po

Integrated Spatial and Bulk Transcriptomic Analysis Identifies a Multicellular Ecosystem Involved in Progression of Young Breast Cancer.

Young breast cancer (YBC), defined as diagnosis at or before the age of 40 years, is characterized by an aggressive phenotype and poor prognosis. The tumor microenvironment (TME), comprising immune and stromal cells, plays a critical role in tumor initiation and progression and is closely associated with clinical outcomes. However, alterations in cell states and the organization of multicellular ecosystems within the YBC microenvironment remain poorly understood. We analyzed 7379 bulk transcriptome samples and spatial transcriptomic data from eight samples using EcoTyper and complementary spat

MFCAMNet: predicting miRNA-disease associations by multi-feature cascade attention mechanism network.

Drive by the rapid envolution of deep-learning techniques, a large body of biological experiments bas has uncovered extensive associations between microRNAs (miRNAs) and complex human diseases, hig- hlighting the pivotal roles of miRNAs in pathogenesis. Elucidating these associations is essential for understanding disease mechanisms and developing preventive strategies. Traditional wet-lab validati- on, however, is notoriously labor- and resource-intensive, creating an urgent demand for efficient computational tools that can prioritize the most promising miRNA-disease candidates. Association of edentulism with incident non-communicable diseases among middle-aged and older adults: a population-based cohort study.

BACKGROUND: Edentulism has been associated with multiple non-communicable diseases (NCDs). However, existing evidence from China is largely limited to specific high-risk regions or lacks national representativeness, and large-scale population-based prospective studies remain scarce. This study investigated the association between edentulism and the risk of developing NCDs in middle and older adults from China. METHODS: We examined the relationship between edentulism and NCDs in the CHARLS study, a large prospective cohort of 17,708 adults in China. Follow-up started from the baseline survey i
