Use of implementation science methods to design Wellness Hub, a responsive program to address long-term care and retirement homes' challenges during the COVID-19 pandemic.

BACKGROUND: Long-term care and retirement homes (LTCH/RH) faced systemic challenges that were exacerbated by the COVID-19 pandemic. Homes faced three major challenges over the course of the pandemic: implementing infection prevention and control (IPAC) practices, facilitating COVID-19 vaccine uptake and confidence, and addressing staff well-being and burnout. This manuscript describes the use of implementation science methods to design an evidence-based, theoretically-rooted program titled the Wellness Hub to support LTCH and RH to navigate real-time COVID-19 challenges. METHODS: Challenges fa

RNF115-Tollip coordination mediates selective autophagic degradation of N protein during PEDV infection.

Porcine epidemic diarrhea virus (PEDV) is a highly lethal swine coronavirus that poses a persistent threat to animal health due to its adaptability and frequent mutations. Despite available vaccines, control remains challenging because of immune evasion and complex host responses. Understanding virus-host interactions, particularly selective degradation pathways, is crucial for improving control strategies. This study systematically reveals the role of E3 ubiquitin ligase RNF115 (also known as BCA2 or Rabring7) during PEDV infection, identifying it as a key regulator of selective autophagy. PE

Identification of two immunodominant and neutralizing linear B-cell epitopes exposed on the surface of the porcine deltacoronavirus spike protein.

Porcine deltacoronavirus (PDCoV) is an enteropathogenic virus that causes severe diarrhea in pigs, particularly suckling piglets, and exhibits cross-species transmission with zoonotic potential. The S1 subunit of the viral spike (S) protein mediates cell entry and elicits neutralizing antibodies, making it an ideal target for diagnostics, prophylaxis, and therapeutics. Here, we produced two monoclonal antibodies (mAbs), B8F10 and G10C2, by immunizing BALB/c mice with a recombinant PDCoV S1 protein fused to a human IgG Fc fragment, expressed in an insect baculovirus system and purified using Pr

Levels and correlates of pandemic anxiety in people living with rare diseases: a cross-sectional analysis using a structural equation model.

BACKGROUND: Within the context of the COVID-19 pandemic and beyond, pandemic anxiety (PA) is of high social and psychopathological relevance. Compared to the general population and people living with common diseases, people living with rare diseases suffer more from the effects of a pandemic in various areas. To date, there are almost no systematic studies on PA in this subpopulation. Therefore, the current study examines the levels and factors associated with PA, as well as the relationship of PA with clinical measures of anxiety and depression disorders in people living with a wide range of

Olfactory Dysfunction and Cognitive Deterioration in Long COVID: Pathomechanisms and Clinical Implications in Development of Alzheimer's Disease.

Complete or partial loss of smell (anosmia), sometimes in association with distorted olfactory perceptions (parosmia), is a common neurological symptom affecting nearly 60% of patients suffering from post-acute neurological sequelae of COronaVIrus Disease of 2019 (COVID-19) syndrome, called long COVID. Severe Acute Respiratory Syndrome CoronaVirus 2 (SARS-CoV-2) may gain access from the nasal cavity to the brain (neurotropism), and the olfactory route has been proposed as a peripheral site of virus entry. COVID-19 is a risk factor for developing Alzheimer's Disease (AD), an age-dependent and p

Healthcare-Associated Infections in Critically Ill COVID-19 Patients Across Evolving Pandemic Waves: A Retrospective ICU Study.

Background and Objectives: Healthcare-associated infections (HAIs) significantly increase morbidity and mortality in critically ill patients, and their burden became more pronounced during the COVID-19 pandemic. However, data describing the temporal evolution of HAIs, pathogen distribution, and associated risk factors across consecutive pandemic waves remain limited. This study aimed to characterize the epidemiology, microbiology, and outcomes of HAIs in COVID-19 intensive care units (ICU) patients and to identify clinical and laboratory predictors of mortality. Materials and Methods: This ret

Bayesian Mixed Models Approach to Exploring Resilience: Impact of Stress on Subjective Health and Affects Over Time During the COVID-19 Pandemic.

BACKGROUND: Profound stressors such as the COVID-19 pandemic have highlighted the importance of understanding resilience mechanisms and approaches for quantifying them in longitudinal studies. METHODS: We used Bayesian mixed models to analyze resilience dynamics with ordinal dependent variables: subjective physical and mental health, and fear, sadness, and anger. The models included fixed effects for individual stressors and random intercepts for participants, applied to the Gutenberg-COVID-19 cohort study. RESULTS: There were 206,912 responses from 7386 participants (mean age 55.09 years, 5

Research Progress of Coronavirus Reverse Genetics Technology.

In recent years, coronavirus, a kind of virus with a wide host range and high variability, has a large and complex genome. Research on the reverse genetics of coronaviruses has always been a hot spot. Coronavirus cannot only infect mammals, but also infect humans, showing its wide host adaptability. The mutation ability of the coronavirus is extremely strong. Recently, the rapid mutation rate of SARS-CoV-2 has made the development of vaccines and therapeutic methods face great challenges. At present, reverse genetics technology is a molecular biology tool. This process primarily involves cloni

Eu-MOF@Ce-MOF heterostructure and molecularly imprinted polymer composite system for SARS-CoV-2 point-of-care detection.

Metal-organic frameworks (MOFs) have attracted growing interest as advanced sensing materials due to their structural tunability and functional versatility. The responsiveness of MOF-based sensors is typically governed by changes in either the metal centers or organic linkers, making it essential to engineer novel architectures that enhance sensing performance. In this work, we present a MOF-on-MOF heterostructure (Eu-MOF@Ce-MOF), composed of a europium-based framework (Eu-MOF) and a cerium-based counterpart (Ce-MOF), synthesized via a two-step hydrothermal strategy. The Eu-MOF was first prepa

[Management of Community-Acquired Pneumonia in Adults - Argentine Society of Infectious Diseases].

Community-acquired pneumonia (CAP) is responsible for substantial morbidity and mortality worldwide. Epidemiological surveillance indicates that Streptococcus pneumoniae remains the most frequent etiological agent and the leading cause of mortality. However, with the advent of new diagnostic techniques, viral etiology has gained priority. Chest X-ray is considered mandatory to confirm the diagnosis and establish the spread. Microbiological, antigen, molecular, biomarker, and carriage tests have specific indications and a role to play in reconsidering empirical treatments. Severity scales are u

Defining the mechanism of cross-reactivity for a SARS-CoV-2 Beta-elicited antibody toward omicron sub-lineages.

Despite the continual emergence of SARS-CoV-2 variants and increasing diversity within the receptor binding domain (RBD), some antibody responses that are directed to conserved regions can display cross-reactivity against variants. We previously isolated an RBD-directed monoclonal antibody (084-7D) from a Beta-infected donor that neutralized Beta and emerging Omicron variants. Here, we solved a high-resolution crystal structure of the 084-7D Fab in complex with the Beta RBD. These data revealed an epitope overlapping both the ACE2 binding site and those of other class 1 antibodies. Furthermore

Circulating mitochondrial and cellular damage markers in long COVID: Links to cognitive function, psychological distress, and inflammation.

Persistent mitochondrial inflexibility and mitochondrial damage may contribute to Post-Acute Sequelae of COVID-19 (PASC). However, data linking mitochondrial biomarkers, such as circulating cell-free mitochondrial DNA (ccf-mtDNA) to long-COVID symptoms remain limited. We analyzed ccf-mtDNA relative to glycerinaldehyd-3-phosphat-dehydrogenase and total cell-free DNA (ccf-DNA) in a nested case-control study of 228 adults (PASC: n = 128, recovered controls: n = 100). Possible associations between these markers and general cognition, verbal memory, psychological distress, and inflammation were als
