Changing Prevalence of Immunocompromising Conditions in Patients Hospitalized with Severe Acute Respiratory Infection in Europe: Insights from the COVIDRIVE Study.

BACKGROUND: Immunocompromised individuals are at substantially increased risk of severe acute respiratory infection (SARI), often compounded by inadequate response to vaccination. We assessed the prevalence of immunocompromising conditions (ICs) among hospitalized SARI patients, overall and by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) status, focusing on temporal trends during the coronavirus disease 2019 (COVID-19) pandemic. We hypothesized that IC prevalence among SARS-CoV-2-positive SARI patients may have increased over time due to increasing background rates of SARS-CoV-

Glycan-binding properties of SARS-CoV-2 spike proteins: interactions with aminoglycoside antibiotics.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infects host cells via its spike (S) protein, which binds to the angiotensin-converting enzyme 2 (ACE2) receptor. Glycans are thought to influence this interaction by modulating the binding affinity between the S protein and its receptor. In this study, we screened 300 carbohydrate species using a glycan array to identify potential ligands that interact with the S proteins of the Delta and Omicron variants. Among the identified candidates, two aminoglycoside antibiotics, tobramycin and sisomicin, exhibited notable binding to the S pr

Views on consent approaches used in emergency and critical care research: a rapid, systematic review.

BACKGROUND: Obtaining informed consent can be challenging in emergency and critical care research due to the acute and severe nature of the patient's condition. However, such research is urgently needed to inform practice and optimise patient outcomes. While alternative consent approaches have been commonly used, opinions may vary, particularly among diverse and underserved patient groups and in the context of the recent COVID-19 pandemic. The objective of this review was to assess views of alternative consent methods in emergency and critical care research. METHODS: We conducted a rapid syst

Preemptive rapid antigen test as an emergency measure during coronavirus disease 2019 outbreaks.

BACKGROUND: We acknowledge that the rapid antigen test (RAT) has several advantages, including faster results, cost-effectiveness, and suitability for on-site self-testing. However, there is still controversy regarding the performance of the RAT for coronavirus disease 2019 (COVID-19). The aim of this study was to evaluate RAT screening in a COVID-19 outbreak situation. METHODS: In this study, we developed a preemptive testing model in which a RAT was immediately followed by reverse transcriptase polymerase chain reaction (RT-PCR) during a single screening in a high-density community outbreak

PDCoV NSP5 cleaves the selective autophagy receptor CCDC50 to disrupt autophagic degradation of the viral envelope protein.

Selective autophagy is a critical host defense mechanism that eliminates viral components through lysosomal degradation during coronavirus infection. Coronaviruses (CoVs), however, deploy countermeasures that disrupt this process, and several underlying mechanisms remain unresolved. Here, we identify the autophagy receptor CCDC50 as a substrate of the coronavirus-encoded NSP5 protease. During porcine deltacoronavirus (PDCoV) infection, NSP5 cleaves CCDC50 at glutamine 171 (Q171), a conserved site also processed by NSP5 from PEDV, TGEV, and SARS-CoV-2. Functionally, CCDC50 restricts PDCoV repli

Antibody kinetics and shedding dynamics of MERS-CoV in dromedary camels from different production systems in Kenya: a longitudinal cohort study.

Understanding long-term antibody kinetics in different camel production systems is crucial for risk assessment for Middle East respiratory syndrome coronavirus (MERS-CoV), a zoonotic pathogen first detected in humans in Saudi Arabia. Though the virus is endemic in dromedary camels across Africa, the Middle East, and parts of South Asia, zoonotic transmission outside of the Arabian Peninsula is undocumented. A retrospective analysis of longitudinal surveillance data of MERS-CoV was conducted on 2460 serum samples collected from 174 camels in three high-risk counties of Kenya from April 2018 to

Epitope-focused discovery of SARS-CoV-2 antibodies that potently neutralize Omicron variants.

The emergence of SARS-CoV-2 Omicron variants has led to viral escape from many clinically approved monoclonal antibodies (mAbs) due to rapid evolution of the receptor-binding domain (RBD). Co-circulation of SARS-CoV-2 variants with unique sets of antigenic substitutions has further complicated therapeutic mAb discovery. New approaches are needed to rapidly discover and characterize mAbs with preferred specificity and functional characteristics. Here we describe and perform epitope-focused mAb discovery using glycan-masked antigens. We isolated and expressed a panel of 303 mAbs, some of which p

Estimating Changes in Facility Methicillin-Resistant Staphylococcus aureus (MRSA) Infection Rates Due to Changes in MRSA Precaution Policy.

BACKGROUND: The effectiveness of contact precautions (CP) and active surveillance (AS) for preventing methicillin-resistant Staphylococcus aureus (MRSA) in acute care remains uncertain. Some studies suggest CP reduces MRSA spread, while others report limited benefit. The COVID-19 pandemic disrupted MRSA prevention practices in the VA, creating an opportunity to assess their impact on MRSA healthcare-associated infections (HAIs). METHODS: We studied 121 VA acute care hospitals from July 2020-June 2022. Facility practices (AS, CP for colonized [CPC], CP for infected [CPI]) were assessed via nat

Hematologic and coagulation responses to Johnson & Johnson COVID-19 vaccination in the Amhara region, Ethiopia.

Coronavirus disease 2019 (COVID-19) is a pandemic disease that causes serious pulmonary complications. Vaccination is the most effective measure to prevent severe illness and reduce mortality. The Johnson & Johnson vaccine is an adenovirus vector-based vaccine designed to target the spike protein of the virus. The vaccine has its own side effects, such as thrombocytopenia and coagulation alteration. Therefore, this study aims to assess changes in hematological and coagulation parameters among individuals who received the Johnson & Johnson COVID-19 vaccine in the Amhara National Regional State,

Tracking viral variants by wastewater surveillance, from laboratory diagnosis towards on-site monitoring: lessons learned from the SARS-CoV-2 pandemic.

Wastewater-based epidemiology (WBE) provides a scalable, population-level biosurveillance layer that complements clinical testing for monitoring SARS-CoV-2 circulation, particularly when diagnostic participation, access, or representativeness is limited. As SARS-CoV-2 continues to evolve, wastewater surveillance has expanded beyond quantifying total viral RNA to also resolving signature mutations and mixed lineage compositions in complex matrices. This review synthesizes end-to-end workflows for variants-directed WBE, spanning sample collection and viral signal enrichment, sequencing-dependent

Life-long coping patterns and bio-psycho-social predictors of individual responses to the COVID-19 pandemic-protocol of the Rostock Longitudinal Study (ninth wave).

BACKGROUND: When faced with adversity and negative life events, such as infection with coronavirus or restrictive measures regarding COVID-19, individuals tend to rely on the same coping strategies they have already employed in the past. However, habitual coping measures vary greatly between people. Differences in dealing with challenging situations may depend on life-long coping experiences and a wide range of bio-psycho-social influences. The major goal of this study is to detect persisting patterns of coping which in turn may predict how individuals have handled the challenges of the COVID-
