The influence of maternal gut and vaginal microbiota on gastrointestinal colonization of neonates born vaginally and per caesarean section.

BACKGROUND: Early life microbial colonization of the neonatal gastrointestinal tract is crucial for imprinting of the immune system. Vertical transmission of maternal microbes is considered the key source of initial neonatal microbial colonization. We aimed to evaluate the role of the maternal vaginal and rectal microbiota in early neonatal gastrointestinal colonization in vaginally- and caesarean section-born neonates. METHODS: Maternal vaginal and rectal swabs were collected shortly before delivery. Neonatal fecal samples were collected at day 0, 7 and 28 postnatally in both vaginally-born

Genome-wide mutagenesis identifies factors involved in MRSA vaginal colonization.

Methicillin-resistant Staphylococcus aureus (MRSA) is an opportunistic pathogen that colonizes various body sites, including the nares, skin, and vagina. During pregnancy, colonization can lead to dysbiosis, adverse pregnancy outcomes, and invasive disease. To identify genes contributing to MRSA vaginal fitness, we performed transposon sequencing (Tn-seq) using a murine model of vaginal colonization, identifying over 250 conditionally essential genes. Five genes were validated in our murine model, including those encoding the aerobic respiration protein QoxB, bacillithiol biosynthesis component

Characterizing the prevalence of Fusobacterium necrophorum subsp. necrophorum, Fusobacterium necrophorum subsp. funduliforme, and Fusobacterium varium in bovine and ovine semen, bovine gut, and vagino-uterine and fetal microbiota using targeted culturing and qPCR.

Fusobacterium necrophorum is an important pathogen associated with several infectious diseases in cattle. However, recent sequencing-based studies reported that F. necrophorum may be positively associated with pregnancy in beef cows and that Fusobacterium is highly abundant in bull seminal microbiota with potential involvement in reproductive health and fertility. Here, we performed a comprehensive screening to (i) determine the prevalence of Fusobacterium necrophorum (subspecies necrophorum [FNN] and funduliforme [FNF]) and Fusobacterium varium (FV) in the reproductive microbiota of cattle an

Asymptomatic Leukocytospermia and Assisted Reproductive Technology Outcomes: Reason for concern?

Leukocytospermia, defined as ≥1×10⁶ white blood cells (WBC)/ml of semen, is a condition frequently observed in infertile men. While symptomatic leukocytospermia is often associated with genital tract infections and managed accordingly, the clinical significance of asymptomatic leukocytospermia remains uncertain-particularly in the setting of Assisted Reproductive Technology (ART). Seminal leukocytes, primarily neutrophils, play a physiological role in immune surveillance and tissue homeostasis. However, when excessively activated, they may generate high levels of reactive oxygen species (ROS),

Patterns of care-seeking for postpartum symptoms in urban Karachi, Pakistan: implications for intervention design.

BACKGROUND: In Pakistan, the maternal mortality rate is 186/100,000 live births, with postpartum (PP) or maternal sepsis being the third leading cause of maternal deaths. Delays in early identification and timely management of PP sepsis are associated with mortality and severe maternal outcomes, including septicemia, neonatal deaths, infertility, etc. In this study, we aim to explore patterns of care-seeking of maternal health services by recently delivered women (RDW) in semi-urban Karachi, Pakistan. Insights of this study will help in identifying and addressing the barriers in care-seeking t

Antimicrobial Resistance in Maternal Infections During Pregnancy.

Background: An imbalance in the vaginal microbiota, often characterized by reduced lactobacilli, paves the way forth for opportunistic bacteria from the gastrointestinal tract. The presence of aerobic bacteria in the genital tract during pregnancy can have negative outcomes on the pregnancy. Peripartum infections, when not adequately managed, can significantly impact maternal and neonatal health. Antimicrobial resistance poses an escalating global health threat, with newborns particularly vulnerable. Methods: This study constitutes a retrospective observational analysis, encompassing all micro

The vaginal microbiota and preterm birth: A systematic review.

Vaginal microbiota is a dynamic community of bacteria, fungi, and viruses that colonize the lower genital tract. Changes in vaginal microbiota can result in obstetric complication. For example, a reduction in Lactobacilli can lead to bacterial vaginosis, which is associated with preterm birth. We performed a systematic review to investigate the association between vaginal microbiota composition during pregnancy and preterm birth risk. Scopus, Web of Science, PubMed databases were used to find published studies on the topic. The search period was between January 2014 and June 2024. A total of 2

Social, microbial, and immune factors linking bacterial vaginosis and infectious diseases.

Bacterial vaginosis (BV) is a polymicrobial condition of the vaginal microbiota associated with a variety of sexually transmitted infections, infections of maternal and fetal tissues during pregnancy, and even some infections outside of the reproductive tract, including the urinary tract and mouth. BV has also been associated with conditions in which the body generates prominent inflammatory reactions to microbes, including infections of the cervix and other upper genital tract tissues. For reasons still not understood, BV is a highly recurrent and often difficult-to-treat condition, complicat

Genomic insights into the potency and functional roles of Lactobacillus species in term and preterm births.

The vaginal microbiome, typically dominated by Lactobacillus species, plays a key role in reducing the risk of preterm birth (PTB) by protecting against infections. In this study, we sequenced 133 Lactobacillus genomes isolated from the reproductive tracts of pregnant Indian women. Genomic analysis identified genes linked to pathogen resistance and anti-inflammatory functions. Further proteome analysis of cell-free supernatant revealed antimicrobial properties, including lysin and bacteriocin, while antibacterial tests confirmed their ability to inhibit reproductive tract pathogens. These find

Drug and therapeutic intravaginal delivery targeting diseases in the female reproductive tract: A mathematical modeling perspective.

Intravaginal drug and therapeutic delivery targeting diseases in the female reproductive tract is advantageous, yet presents significant challenges due to unique anatomical features, cyclic variations, and a complex microbial ecosystem. Disruption of the intricate vaginal environment due to dysbiosis or infection can decrease immune protection and lead to infertility and pregnancy complications. A variety of intravaginal drug delivery systems (DDS) including creams, gels, suppositories, tablets, rings, and films have been developed to address these conditions. However, relying solely on empiri

Identification and characterisation of vaginal bacteria-glycan interactions implicated in reproductive tract health and pregnancy outcomes.

Lactobacillus displacement from the vaginal microbiome associates with adverse health outcomes and is linked to increased risk of preterm birth. Glycans mediate bacterial adhesion events involved in colonisation and infection. Using customised glycan microarrays, we establish glycan interaction profiles of vaginal bacteria implicated in reproductive health. Glycan binding signatures of the opportunistic pathogens Escherichia coli, Fusobacterium nucleatum and Streptococcus agalactiae to oligomannose N-glycans, galactose-terminating glycans and hyaluronic acid, respectively are highly distinct f

The association between dietary fiber intake and pelvic inflammatory disease in women: findings from the NHANES 2015-2018.

BACKGROUND AND AIM: Pelvic inflammatory disease (PID) is a common and serious gynecological condition caused by ascending infections of the female genital tract. It affects approximately one million women annually in the United States and is a leading cause of infertility, ectopic pregnancy, and chronic pelvic pain. While well-established risk factors include multiple sexual partners and sexually transmitted infections, emerging evidence suggests that dietary factors may also influence systemic and reproductive tract inflammation. In particular, dietary fiber has been shown to exert anti-infla
