Prenatal initiation of elexacaftor/tezacaftor/ivacaftor via carrier mother prevents congenital bilateral absence of vas deferens in a male infant with cystic fibrosis.

BACKGROUND: Prenatal therapy with elexacaftor/tezacaftor/ivacaftor (ETI) has recently emerged as a potential strategy to modify the early natural history of cystic fibrosis (CF). While case reports have described prevention of meconium ileus and pancreatic insufficiency, data on male reproductive outcomes remain extremely limited. CASE PRESENTATION: We report a male infant with CF (homozygous F508del) whose heterozygous carrier mother initiated ETI at 27+4 weeks of gestation after prenatal diagnosis. Pregnancy was uneventful until preterm delivery at 35+2 weeks. The neonate presented with nor

Adverse Pregnancy Outcomes and Prevalence Associated With Arboviral Infections (Zika, Dengue, and Chikungunya): An Umbrella Review of Systematic Reviews and Meta-Analyses.

Maternal infection with Zika virus (ZIKV), dengue virus (DENV), or chikungunya virus (CHIKV) may increase the risk of adverse pregnancy and foetal outcomes, but evidence credibility varies. The aim of this study is to evaluate and compare the strength of evidence from systematic reviews and meta-analyses on these arboviruses and pregnancy outcomes. PubMed, Web of Science, and Cochrane Library from inception to December 31, 2025 were searched. Two reviewers independently screened studies, extracted data, and assessed methodological quality using AMSTAR 2. Evidence credibility was graded as high

Evaluation of the relationship between worry and anxiety with the general health status of pregnant women at risk of diagnosing abnormalities.

BACKGROUND: Today, the screening of fetal abnormalities during pregnancy is used as one of the components of the prenatal care worldwide, and many abnormalities are detected by ultrasound during pregnancy. On the other hand, the possibility of an abnormality in the fetus causes worry and anxiety in pregnant women. Therefore, the present study was conducted with the aim of determining the relationship between worry and anxiety with the general health status of pregnant women at risk of diagnosing fetal abnormalities. METHODS: This descriptive-analytical cross-sectional study was conducted on 2

A novel compound heterozygous mutation in the DYNC2H1 gene in a Chinese family with Jeune syndrome.

BACKGROUND: The dynein cytoplasmic two heavy chain 1 (DYNC2H1) gene encodes a cytoplasmic dynein subunit. Cytoplasmic dyneins transport cargo towards the minus end of microtubules and are thus termed the "retrograde" cellular motor. Mutations in DYNC2H1 are the main causative mutations of short rib-thoracic dysplasia syndrome type III with or without polydactyly (SRTD3). Early diagnosis of SRTD3 prenatally by ultrasound alone is difficult. In this case, a couple who gave birth to three consecutive babies with SRTD3 requested fertility guidance to avoid having another baby with SRTD3. METHODS:

Analysis of pulmonary sequestration cases, based on the 11-year birth defects surveillance data of Changsha hospitals.

Pulmonary sequestration (PS) is a rare congenital malformation that is characterized by the absence of a connection between a portion of the lung tissue and the tracheobronchial tree, with blood supply from arteries throughout the body. Abnormal lung tissue cannot perform the normal gas exchange function. In the absence of timely diagnosis and early intervention, some cases may need labor induction, and some of the infants who survive may develop symptoms in childhood. The manifestations are repeated lung infections, dry cough, chest pain, chest tightness, and hemoptysis. Some cases have no sy

Phenotypic and genotypic insights into concurrent tertiary trisomy for 9p and 18p.

BACKGROUND: Carriers of balanced reciprocal translocation are usually phenotypically normal; however, they have an increased risk of producing gametes with chromosomal imbalance through different types of meiotic segregation of the translocation quadrivalent. The genetically imbalanced gametes when they survive can result in embryos with chromosomal abnormalities. Here we report a family with two siblings inheriting partial trisomy for 9p and 18p concurrently resulting from a 3:1 meiotic segregation of a maternal balanced translocation involving chromosome 9q and 18p, and the associated phenot

Fetal genetic factors in pregnancy loss: Insights from a meta-analysis and effectiveness of whole exome sequencing.

Spontaneous pregnancy loss commonly occurs during the first trimester and can be caused by various factors including chromosomal abnormalities and submicroscopic aberrations. After the first trimester, the etiology of most pregnancy losses remains undetermined. This study aims to fill this gap by an in-depth investigation of the fetal genome and its effect on pregnancy outcome. Data from 1016 spontaneously aborted fetuses previously referred for genetic testing (2017-2023) were used for meta-analysis. Fetuses were categorized based on gestational age and genetic test result. Additionally, 35 s

Reproductive Implications and Management of Congenital Uterine Anomalies: Scientific Impact Paper No. 62.

Congenital uterine anomalies (CUAs) are malformations of the uterus (womb) that develop during fetal life. When a female baby is in her mother's uterus, her uterus develops as two separate halves from two tubular structures called Müllerian ducts, which fuse together before she is born. Anomalies that occur during the baby's development can be variable, from complete absence of the uterus through to more subtle anomalies, which are classified into specific categories. While conventional ultrasound is good at detecting CUAs, 3D ultrasound is used to confirm a diagnosis. If a complex uterine ano

Prenatal diagnosis and genetic counseling of a Chinese family with inherited multiple chromosomal microduplications.

BACKGROUND: Copy number variants (CNVs) are an important source of normal and pathogenic genome variations. Chromosomal microdeletions and microduplications have long been associated with abnormal developmental outcomes. Recently, the application of CNV sequencing (CNV-seq) is rapidly identifying new recurrent microdeletion and microduplication syndromes and a previously unsuspected level of copy number variation which needs to be distinguished from pathogenic change. MATERIALS AND METHODS: In this research, a 26-year-old, gravida 1, para 0, woman underwent amniocentesis at 18 weeks of gestat

Intrauterine Herpes Simplex Virus Infection: Insights Into a Silent Threat.

We report a rare case of congenital herpes simplex virus type 2 (HSV-2) infection in a neonate born to a mother with no clinical evidence of genital HSV infection either before or during pregnancy. Routine prenatal ultrasound assessments in the first two trimesters were unremarkable, and infectious disease screening results were negative. The pregnancy, however, was complicated by premature rupture of membranes (PROM) at 24 weeks of gestation. An ultrasound at that time revealed severe fetal malformations, including brain abnormalities and organomegaly. At 31 weeks, an emergency cesarean secti

Utilizing Long-Read Sequencing for Haplotype Construction and Prevention of Autosomal Dominant Polycystic Kidney Disease Transmission in Mosaicism Family.

This study presents a case of autosomal dominant polycystic kidney disease (ADPKD) involving a mosaic microdeletion in the PKD1 gene and explores the application of long-read sequencing technologies for haplotype construction and preimplantation genetic testing (PGT). We report on a family where the proband was clinically diagnosed with PKD and found to have a partial deletion of the PKD1 gene because of the mosaic deletion mutation of PKD1 in the mother of the proband. Utilizing Oxford Nanopore long-read sequencing, we successfully constructed the haplotype of the deleted fragment region and
