BACKGROUND: The dynein cytoplasmic two heavy chain 1 (DYNC2H1) gene encodes a cytoplasmic dynein subunit. Cytoplasmic dyneins transport cargo towards the minus end of microtubules and are thus termed the "retrograde" cellular motor. Mutations in DYNC2H1 are the main causative mutations of short rib-thoracic dysplasia syndrome type III with or without polydactyly (SRTD3). Early diagnosis of SRTD3 prenatally by ultrasound alone is difficult. In this case, a couple who gave birth to three consecutive babies with SRTD3 requested fertility guidance to avoid having another baby with SRTD3.

BACKGROUND: Carriers of balanced reciprocal translocation are usually phenotypically normal; however, they have an increased risk of producing gametes with chromosomal imbalance through different types of meiotic segregation of the translocation quadrivalent. The genetically imbalanced gametes when they survive can result in embryos with chromosomal abnormalities. Here we report a family with two siblings inheriting partial trisomy for 9p and 18p concurrently resulting from a 3:1 meiotic segregation of a maternal balanced translocation involving chromosome 9q and 18p, and the associated phenot

Spontaneous pregnancy loss commonly occurs during the first trimester and can be caused by various factors including chromosomal abnormalities and submicroscopic aberrations. After the first trimester, the etiology of most pregnancy losses remains undetermined. This study aims to fill this gap by an in-depth investigation of the fetal genome and its effect on pregnancy outcome. Data from 1016 spontaneously aborted fetuses previously referred for genetic testing (2017-2023) were used for meta-analysis. Fetuses were categorized based on gestational age and genetic test result. Additionally, 35 s

OBJECTIVE: A case of prenatal diagnosis of familial 15q13.2q13.3 microdeletion is presented.
CASE REPORT: A 35-year-old, gravida 2, para 1, woman was referred for genetic counseling because of 15q13.2q13.3 microdeletion in the fetus and the mother. The carrier mother was asymptomatic and normal in phenotype. The woman underwent amniocentesis at 17 weeks of gestation because of short nasal bone on fetal ultrasound. Amniocentesis revealed a karyotype of 46,XX. Simultaneous array comparative genomic hybridization (aCGH) analysis on the DNA extracted from uncultured amniocytes revealed the result

BACKGROUND: Copy number variants (CNVs) are an important source of normal and pathogenic genome variations. Chromosomal microdeletions and microduplications have long been associated with abnormal developmental outcomes. Recently, the application of CNV sequencing (CNV-seq) is rapidly identifying new recurrent microdeletion and microduplication syndromes and a previously unsuspected level of copy number variation which needs to be distinguished from pathogenic change.
MATERIALS AND METHODS: In this research, a 26-year-old, gravida 1, para 0, woman underwent amniocentesis at 18 weeks of gestat

This study presents a case of autosomal dominant polycystic kidney disease (ADPKD) involving a mosaic microdeletion in the PKD1 gene and explores the application of long-read sequencing technologies for haplotype construction and preimplantation genetic testing (PGT). We report on a family where the proband was clinically diagnosed with PKD and found to have a partial deletion of the PKD1 gene because of the mosaic deletion mutation of PKD1 in the mother of the proband. Utilizing Oxford Nanopore long-read sequencing, we successfully constructed the haplotype of the deleted fragment region and

OBJECTIVE: 15q11-q13 microduplication syndrome, a genetic disorder caused by duplications in the 15q11-q13 region, has been associated with autism spectrum disorder (ASD); however, research on chromosome 15 microduplications is scarce. We explored the copy number variant (CNV) characteristics of 15q11-q13 microduplication syndrome, pathogenic mechanisms of ASD-related causative genes in this region, and factors affecting ASD development.
METHODS: We performed laboratory investigations and data collection on seven cases of 15q11-q13 microduplications, comprising 5 prenatal and 2 postnatal case

BACKGROUND: Non-invasive prenatal testing (NIPT), which made use of cell-free DNA (cffDNA) in maternal blood, was currently being applied all over the world for the detection of common chromosome abnormalities. It had relatively high sensitivity and specificity. Nevertheless, studies demonstrated that false positive results happened in 0.3% of the cases due to several factors. These factors included confined placental mosaicism, maternal mosaicism, maternal transfusions, maternal malignancy, vanishing twins and maternal chromosomal abnormalities.
CASE REPORT: We presented a case of a 27-year-

OBJECTIVES: In 2023 and 2024, our primary study group consisted of 39 fetuses with HLHS.
MATERIAL AND METHODS: From the final analysis, we excluded the following: 1 twin pair (twin A with normal heart anatomy and twin B with HLHS), 2 terminations of pregnancy, and 2 newborns without prenatal echocardiography (only ultrasound was performed, and HLHS was not detected prenatally). As a result, our study group comprised 34 fetuses. The mean gestational age at the detection of HLHS was 19 weeks (± 6 weeks). Extracardiac malformations were present in 7 fetuses out of 34. Fetal abnormalities are major issues in prenatal medicine. They affect the predicted pregnancy outcome and entail a risk of future recurrent adverse events in a particular couple. In order to clarify the possible outcomes of a specific pregnancy and subsequent ones, it is of the utmost importance to determine the causes of observed fetal abnormalities. Routine laboratory techniques sometimes fail to identify their cause because they are mainly intended for the detection of chromosomal disorders. Over recent years, single-gene disorders have increasingly been regarded as probable causes of feta

RATIONALE: Placental mesenchymal dysplasia (PMD) is a rare placental disorder that poses diagnostic challenges and is often misdiagnosed as a trophoblastic tumor. While PMD is associated with fetal growth restriction, intrauterine fetal death, and pregnancy-induced hypertension, it does not involve malignant trophoblastic disease. Accurate diagnosis is crucial for optimizing maternal and fetal outcomes.
PATIENT CONCERNS: A 30-year-old female, gravida 3, para 1, with an uncomplicated medical history, presented with early pregnancy bleeding, which was managed with oral progesterone. Ultrasound

Differentially methylated regions (DMRs) in certain areas of the genome are subject to genomic imprinting. DMRs at chromosome 11p15.5 are associated with Beckwith-Wiedemann syndrome (BWS) and Russell-Silver Syndrome (RSS), two growth disorders with opposite phenotypes. We identified a maternally inherited duplication containing part of the 11p15 DMR in a non-anomalous fetus in first trimester using genome sequencing (GS). The ∼281kb duplication at 11p15.5 contains the entire imprinting control region 1 (ICR1) and the H19 gene but lacks the IGF2 gene and the imprinting control region 2 (ICR2).

Rubinstein-Taybi syndrome (RSTS) is a rare genetic disorder characterized by distinctive craniofacial, limb, and developmental abnormalities, often identified postnatally. Prenatal diagnosis remains challenging due to a scarcity of ultrasound diagnostic markers and a wide range of phenotypic manifestations. We describe the case of a 28-year-old pregnant patient who presented to our center after fetal abnormalities such as aberrant cranial morphology, a shorter femur, and rocker-bottom feet were detected. A comprehensive ultrasound examination at 26 weeks revealed skeletal and craniofacial char

Renpenning syndrome is a rare X-linked genetic disorder caused by variants in the PQBP1 gene, but the information about its prenatal presentation is very limited. A 35-year-old woman experienced two male pregnancies with thickened nuchal translucency (NT) (5.5 mm and 5 mm). She went to our prenatal diagnosis center for the current natural conception during the second pregnancy. Trio-whole exome sequencing (TrioWES) of chorionic villus biopsy revealed a 666-bp genetic deletion (chrX:48755195-49760422) in the fetus, inherited from the mother, which included TIMM17B and PQBP1. The couple opted fo

The reversed cortico‑medullary differentiation in fetal kidneys on ultrasound has been described in the literature, but there have been no descriptions of such a finding on fetal magnetic resonance imaging (MRI) so far. The authors present three unrelated fetuses with hyperechoic kidneys on ultrasound (US) and reversed signal intensity of their cortex and pyramids on SSFSE/T2WI and FIESTA images on magnetic resonance imaging (MRI). All of them shared the same deletion of the long arm of chromosome 17 in the 17q12 region, responsible for the expression of clinical features of renal cysts and di

BACKGROUND: Chromosomes, cellular structures carrying genetic information, are fundamental to cellular genetics research, with abnormalities potentially causing serious genetic disorders. This makes chromosome screening an essential component in prenatal care for monitoring fetal development and detecting potential genetic abnormalities early in pregnancy. However, manual chromosome analysis is highly inefficient and increasingly burdensome for medical laboratories as prenatal examinations become more common in modern healthcare practices worldwide. Current karyotype analysis systems face sign

Genome-wide non-invasive prenatal testing (NIPT) is a powerful tool for prenatal detection of the common aneuploidies causing Down-, Edwards-, and Patau syndrome. Its genome-wide reach also enables the detection of unbalanced structural chromosomal abnormalities. We report a case where NIPT indicated a ~25 Mb terminal deletion of the long arm of chromosome 16, later confirmed to be mosaic in maternal blood and buccal cells (in 35%-45% of cells). Remarkably, the same mosaic deletion was seen in the child, who was born healthy at term after an uneventful pregnancy. Further analysis using bromode

BACKGROUND: Both balanced translocation and inversion belong to chromosome structural aberration, in which balanced translocation only changes the corresponding position of the translocation fragment on the chromosome. Inversion refers to the phenomenon that two breaks occur on one chromosome at the same time, and the fragments are reversed 180° and then reconnected. Chromosome balanced translocation and inversion usually have normal phenotype and intelligence because they retain the total amount of original genes. However, due to the reduction of the probability of producing normal gametes, p

BACKGROUND: This study aimed to determine how obstetricians' knowledge of T18 influences their attitudes toward the antepartum, intrapartum, and postpartum management of T18 cases and to compare these attitudes with their approaches to trisomy 21 (T21) cases. These attitudes were analysed within the framework of the medical ethics principles of beneficence and autonomy.
METHODS: This cross-sectional descriptive survey study was conducted between 1 February and 31 July 2021. Data from 194 of 388 participants who completed all questionnaire items were analysed.

BACKGROUND: Complex chromosomal rearrangement (CCR) refers to a structural rearrangement involving at least two chromosomes or a minimum of three breakpoints. CCR may lead to intellectual disability, structural anomalies, infertility, and recurrent miscarriages. Chromosome karyotyping and chromosomal microarray analysis (CMA) are unable to detect complex chromosomal rearrangements. As multiple diagnostic approaches are available in clinical practice for detecting chromosomal structural abnormalities and copy number variations-each with its own advantages and limitations-selecting the appropria
