The dual role of vulvovaginal candidiasis in HPV infection: implications for vaginal microecology and cervical lesions.

Persistent infection with human papillomavirus (HPV) is a major pathogenic factor in the development of cervical lesions and cervical cancer. Its occurrence is not only related to the virus itself but also closely associated with the stability of the host vaginal microecological environment. In particular, microbial dysbiosis caused by vaginal infections such as bacterial vaginosis (BV), trichomoniasis (TV), and vulvovaginal candidiasis (VVC) may facilitate HPV acquisition and persistence. Among these factors, VVC has drawn special attention due to its unique bidirectional role: it may promote

Meteorin-like is associated with poor outcome in invasive candidiasis in mouse models and in humans.

Invasive candidiasis is a leading cause of nosocomial bloodstream infection associated with high mortality, and there is a pressing need to develop biomarker-guided antifungal therapy to improve clinical outcomes. Meteorin-like (METRNL) is a cytokine that can act as a high-affinity ligand for the stem cell factor receptor KIT; however, the functional role of METRNL in fungal infection remains unclear. Here, we found that METRNL acts as a disease-promoting immune checkpoint to facilitate invasive Candida albicans (C. albicans) infection. Mice deficient in METRNL were refractory to a lethal syst

Vacuole and mitochondria patch (vCLAMP) protein Mcp1 reduces the pathogenesis of Candida albicans by impeding the function of Cap1-mediated oxidative stress response.

BACKGROUND: Vacuole and mitochondria patch (vCLAMP) is a novel organelle membrane contact site. The vCLAMP protein Mcp1 plays a crucial role in cellular signaling and homeostasis. However, its role in the virulence of Candida albicans remains to be elucidated. RESULTS: This study explored the function of Mcp1 in the oxidative stress response of C. albicans. Fluorescence co-localization and immune transmission electron microscopy results suggested that Mcp1 was located on vacuolar and mitochondrial membranes. RNA sequencing and Gene Ontology enrichment analyses showed that the deletion of MCP1

Case Report: Diagnosis of bronchopulmonary candidiasis-refractory airway hyperresponsiveness, severe pneumonia, and normal serological markers.

BACKGROUND: Bronchopulmonary candidiasis is a clinically common type of pulmonary fungal disease, primarily caused by infection with Candida species (mostly Candida albicans). It typically manifests as a secondary infection when the body's resistance is compromised. Clinically, bronchopulmonary candidiasis is primarily classified into three types: (1) bronchitic, (2) pneumonic, and (3) hypersensitivity. METHODS: This paper reports two cases of bronchopulmonary candidiasis. The first case presented with bronchitis-type manifestations. Imaging studies showed no evidence of severe pneumonia, but

Antifungal and molecular analysis of gene expression caused by haloperidol in Candida spp.

Candidiasis, caused by yeasts of the Candida genus, is increasingly characterized by a high prevalence of clinical isolates resistant to conventional antifungals, rendering the development of novel therapeutic strategies paramount. Drug repurposing has emerged as a key strategy, utilizing established pharmaceuticals for indications beyond their original design; notably, haloperidol (HAL) has shown promising antimicrobial potential. In this context, the present study evaluates the activity of haloperidol, both as a monotherapy and in combination with conventional antifungals, against fluconazol

Oligonucleotide-Driven Nanoparticle Biosensor for Candida albicans Detection in Vaginal Swabs.

Vulvovaginal candidiasis (VVC), caused by Candida albicans ( C.albicans ), is a common gynecological infection. VVC is traditionally diagnosed using microbiological culture and microscopy by immunofluorescence, which suffer from limitations such as time-consuming procedures and subjectivity. This study developed a colorimetric biosensor based on gold nanoparticles functionalized with oligonucleotides for the rapid detection of C.albicans. To target the specific hypha wall protein 1 gene (HWP1), thiol-modified oligonucleotide probes were designed and conjugated to the surface of 60 nm citrate-s

Recurrent mutations in the stress regulator Cap1 reveal a trade-off between azole resistance and oxidative stress response in Candida albicans.

Drug resistance is a critical challenge in treating life-threatening fungal infections. Here, we uncover a mechanism of acquired azole resistance in Candida albicans through mutations in CAP1, encoding a conserved fungal transcription factor that mediates the oxidative stress response. We analyzed 300 clinical isolates and identified 25 distinct CAP1 missense or nonsense mutations, with many occurring within the DNA-binding domain. We identified two nearly identical CAP1 heterozygous nonsense mutations, one in an isolate obtained from a bloodstream infection and one in a population of cells un

Impact of extracellular vesicles from various species of Candida fungi on the cross-species interactions with human epithelial cells.

Candida species are members of the human microbiota but can become pathogenic when host homeostasis is disrupted. Among the species responsible for human fungal infections are C. albicans, C. tropicalis, and Nakaseomyces glabratus (formerly classified as C. glabrata). These fungi employ multiple strategies to colonize host niches, including phenotypic and morphological plasticity, biofilm formation in mono- or multispecies communities, secretion of virulence factors, and release of extracellular vesicles (EVs). Adhesion of fungal cells to host tissues is a critical step in the establishment an

Fungal infection drives metabolic reprogramming in epithelial cells via aerobic glycolysis and an alternative TCA cycle shunt.

Candida albicans-induced immunometabolic changes drive complex responses in immune cells. However, whether and how C. albicans causes remodeling of oral epithelial cell (OEC) metabolism is unclear. Here, we use in vitro experiments and patient biopsies to demonstrate that OECs undergo metabolic reprogramming when infected by C. albicans independently of candidalysin secretion, increasing glycolysis and decreasing tricarboxylic acid (TCA) cycle activity. Glycolysis and glucose transport inhibition show that these pathways support OEC cytokine release, highlighting the partial control of antifun

Maternal and demographic factors influencing oral Candida albicans in infants: A stratified analysis using a novel partial linear semiparametric mixed-effects model.

Candida albicans, a pathogenic fungus implicated in early childhood caries (ECC), plays a crucial role in oral health. While its colonization usually begins at birth, the extent of maternal involvement in yeast transmission to their offspring, particularly across different racial groups, remains unclear. Studies have shown elevated levels of C. albicans in both mothers and children, with genetically related fungal strains, suggesting maternal transmission, but the racial component, notably higher levels in Black children, lacks thorough investigation of underlying factors. Our research aimed t

Elevated vaginal heparan sulfate correlates with impaired neutrophil killing of Candida albicans in women with vulvovaginal candidiasis.

Recurrent vulvovaginal candidiasis (RVVC), primarily caused by the fungal pathogen Candida albicans, is a common infection affecting a significant number of women worldwide. Despite a robust inflammatory response by polymorphonuclear neutrophils (PMNs) with potent antifungal properties during symptomatic episodes, fungal clearance often fails, leading to persistent overgrowth and PMN-associated immunopathology. Studies in an established animal model demonstrated that elevated vaginal heparan sulfate (HS) interferes with PMN-C. albicans interactions, thereby impairing fungal clearance. This stu
