Automated Counting of Chikungunya Virus-Formed Plaques.

The cytopathic effect (CPE) is a key to understanding the pathogenicity of the chikungunya virus (CHIKV) and is useful for evaluating viral infectivity in vitro. The plaque assay, based on the CPE induced by viral infection, provides an infectious virus titer for test samples. Additionally, the assay is simple, quantitative, and cost-effective. Thus, the plaque assay remains the gold standard for determining the infectivity of CHIKV. However, manually counting of virus-induced plaques with the naked eye or under the microscope is often time-consuming and labor-intensive. Therefore, automated p

Early discontinuation vs. continuation of empirical antibiotics in patients with cancer and febrile neutropenia: a systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Empirical broad-spectrum antibiotics are standard care for febrile neutropenia, but prolonged treatment until neutrophil recovery may increase antimicrobial resistance, toxicity, Clostridioides difficile infection, fungal superinfection, and healthcare burden. Whether antibiotics can be safely discontinued before neutrophil recovery remains uncertain. METHODS: We systematically reviewed randomized controlled trials comparing early or short-course discontinuation of empirical antibacterial therapy with continuation until neutrophil recovery or an extended fixed course in patients w

Phospholipid codes at virus-host membrane interfaces: mechanisms and therapeutic opportunities in animal viral diseases.

Animal viruses depend on host membranes at every stage of infection, yet membrane lipid states are often treated as background rather than as causal determinants of virus-host interactions. This Review develops a phospholipid-code framework in which headgroup identity, acyl-chain chemistry, leaflet topology, and lipid flux define infection-stage-specific membrane states. Evidence from Newcastle disease virus, infectious bronchitis virus, avian influenza virus, porcine reproductive and respiratory syndrome virus, African swine fever virus, Senecavirus A, Japanese encephalitis virus, and porcine

Epidemiological update on spatial and space-time clusters of African swine fever in the Philippines.

African swine fever (ASF), particularly the highly contagious Genotype II strain, is a transboundary viral disease causing up to 100% mortality in swine and threatening food security across the Asia-Pacific region. Since 2019, control strategies in the Philippines have relied on depopulation and strict administrative zoning to limit contiguous, land-based spread. However, the long-term effectiveness of these approaches within the country's fragmented archipelagic landscape remains poorly understood, complicating evidence-based resource allocation and preparedness for regional vaccination campa

Comparative evaluation of real-time PCR and droplet digital PCR targeting the B646L (p72) and CP204L (p30) genes for early detection of African swine fever virus.

This study compared the analytical performance, diagnostic accuracy, and early detection capability of four nucleic acid amplification assays for African swine fever virus (ASFV), including two real-time PCR (qPCR) assays and two droplet digital PCR (ddPCR) assays targeting the B646L (p72) and CP204L (p30) genes. Analytical performance was evaluated using plasmid standards, whereas diagnostic performance was assessed using 133 samples comprising 73 specimens from experimentally infected pigs and 60 ASFV-negative controls. Both qPCR assays exhibited excellent linearity (R² > 0.99) and acceptabl

Comparing two case definitions for including severe acute respiratory infection patients in influenza and COVID-19 vaccine effectiveness studies among older adults using the test-negative design: VEBIS hospital network, seven European countries, 2023/24.

INTRODUCTION: Two severe acute respiratory infection (SARI) case definitions (CDs) are often used in test-negative design (TND) hospital-based vaccine effectiveness (VE) studies. SARI-CD1 requires ≥1 symptom (cough, fever, shortness of breath, anosmia, ageusia, or dysgeusia); SARI-CD2 requires both fever and cough. Using VEBIS hospital network end-of-season 2023/24 data, we compared the effect of these CDs on influenza and COVID-19 VE estimates in Europe. METHODS: Using the TND, we restricted analysis to seven countries recruiting patients using SARI-CD1; we created a subset of SARI patients

Clinical characteristics, pathogen detection, and severity markers among children with acute respiratory infections during the COVID-19 restriction period: a retrospective hospital-based study.

BACKGROUND: The epidemiology of pediatric acute respiratory infections (ARIs) changed substantially during the COVID-19 (coronavirus disease 2019) pandemic, largely because of school closures, masking, reduced travel, and other non-pharmaceutical interventions (NPIs). However, hospital-based data from the restriction period remain useful for describing pathogen detection patterns, clinical severity, and markers of adverse clinical course. METHODS: We conducted a retrospective hospital-based study of 62 children aged <18 years who were evaluated for ARI symptoms and underwent real-time fluores

When Fever Hides Fire: A Systematic Review of Clinical and Digital Approaches to Early Recognition of Pediatric Sepsis.

Early recognition of pediatric sepsis remains challenging because clinical presentation, sepsis definitions, and recognition approaches vary across healthcare settings. This systematic review evaluated clinical and digital strategies for the early recognition of pediatric sepsis, including structured screening tools, electronic alerts, prediction models, machine-learning approaches, clinical decision support, and recognition pathways. Diagnostic criteria were considered separately as reference frameworks for classifying established sepsis and interpreting the performance of recognition tools.

Endovascular retrieval of bone cement entrapped in an inferior vena cava filter after percutaneous vertebroplasty: a case report.

Percutaneous vertebroplasty (PVP) is widely used to treat osteoporotic vertebral compression fractures, however, bone cement leakage may enter the venous system and cause potentially serious embolic complications. Entrapment of migrated bone cement within an inferior vena cava filter (IVCF) is rare and may pose substantial technical challenges during filter retrieval. We report the case of a 67-year-old woman with bilateral deep vein thrombosis who underwent placement of a Denali retrievable IVCF after thrombus progression while receiving a nonstandard initial rivaroxaban regimen selected beca

Disparate tetraparvovirus species linked to hepatitis.

The role of human tetraparvoviruses (PARV4) in cases of acute illness is poorly characterized, and the contribution of animal-associated tetraparvoviruses to human disease is unknown. In this study, we applied metagenomic and viral-enrichment next-generation sequencing to plasma from patients with diagnostically unresolved acute illness in Georgia (non-A-C hepatitis) and Bangladesh (acute febrile illness), and two tetraparvovirus genomes were recovered. Specimen HEP004 from a Georgian patient with acute hepatitis contained a virus clustering within Tetraparvovirus ungulate4, a species associat

Confirmation of gamma irradiation-mediated inactivation of a vero-adapted African swine fever virus Lisbon 60 strain for molecular assays.

African swine fever virus (ASFV), the etiologic agent of African swine fever (ASF), is a high-consequence pathogen requiring experiments to be conducted in containment in non-endemic countries, thereby restricting diagnostic development, the creation of reference standards, and proficiency testing (PT). Safe and reliable inactivation methods are essential to expand diagnostic capacity while preserving nucleic acid integrity for molecular assays in unaffected countries. This study employed gamma irradiation to achieve complete inactivation of ASFV without compromising downstream molecular detec
