Diagnostic evaluation of a recombinant chimeric Sporothrix brasiliensis protein-based ELISA for serological detection of feline sporotrichosis.

Sporotrichosis is a zoonotic infection caused by fungi of the Sporothrix schenkii complex, with S. brasiliensis being the most prevalent etiological agent in animals, particularly cats. Felines are highly susceptible to infection and may develop severe clinical forms characterized by extensive cutaneous lesions and systemic dissemination. In addition, infected cats play a major role in zoonotic transmission, as Sporothrix spp. can be transmitted to humans through bites and scratches. The gold standard for sporotrichosis diagnosis remains the isolation and identification of Sporothrix species f

Unusual Sporotrichosis: A New Concept Proposal on the Unexpected Faces of Sporothrix spp. Infection.

"Unusual sporotrichosis", a concept proposed in this review, refers to severe, extracutaneous, or anatomically atypical manifestations of sporotrichosis occurring in immunocompetent hosts and represents an underrecognized clinical subset associated with important diagnostic and therapeutic challenges. This systematic review aimed to characterize unusual sporotrichosis worldwide and to clarify its epidemiological, clinical, diagnostic, and therapeutic patterns. Following a registered protocol and PRISMA guidelines, PubMed, Scopus, and BVS/LILACS were searched up to November 2025 using a PICO-ba

Silver(I) and Copper(II) complexes of 1,10-phenanthroline-5,6-dione inhibit Sporothrix brasiliensis azole-resistant clinical isolates.

BACKGROUND: Sporothrix brasiliensis is the principal etiological agent responsible for cat-transmitted zoonotic sporotrichosis, which is currently the most prevalent mycosis in South America-especially in Brazil-and is increasingly being reported in other regions. While itraconazole is the standard treatment, its use is limited by hepatotoxicity, high cost, and emerging reports of reduced susceptibility. Complexes of 1,10-phenanthroline-5,6-dione (phendione) with transition metals have previously shown promise as antifungal agents against bacteria, parasites, yeasts and filamentous fungi. This

A Presentation of Necrotizing Disseminated Sporotrichosis.

We report a biopsy-proven case of disseminated sporotrichosis in a 42-year-old immunocompetent male, highlighting an uncommon presentation with rapid clinical deterioration. The patient initially presented with cutaneous lesions involving the face, thorax, and extremities that were suspected to represent a delayed hypersensitivity reaction to trimethoprim-sulfamethoxazole, and he was treated with systemic corticosteroids. He returned 10 days later with worsening necrotic lesions and constitutional symptoms. Otolaryngology and dermatology services obtained multiple skin biopsies, which demonstr

Unique Genetic Clades and Frequent Zoonotic Transmission of Sporothrix brasiliensis with Reduced Itraconazole Susceptibility.

Sporothrix brasiliensis is driving a major zoonotic epidemic in Brazil, predominantly via cat-to-human transmission. In the late 1990s major outbreaks were reported in Rio de Janeiro (RJ) and Rio Grande do Sul (RS), the southernmost state. We characterized the antifungal susceptibility and genetic diversity of 450 S. brasiliensis isolates collected between 2009 and 2024 in RS to better understand treatment response, emergence and transmission.A bimodal distribution of the itraconazole minimum inhibitory concentration (MIC) was observed with 24% non-wild-type (NWT) isolates. Short tandem repeat

Cryotherapy for treatment of sporotrichosis-rapid cure with adjuvant cryotherapy: case report.

BACKGROUND: Sporotrichosis is a subcutaneous mycosis caused by various sporothrix fungus species. This ubiquitous fungal infection is more commonly seen in field/farm workers in tropical and subtropical regions. This prevalence could largely be attributed to the climatic condition favorable for harboring fungi in these regions. Cutaneous infection often occurs via inoculation of fungus into the intact skin owing to traumatic skin injuries. Clinically apparent lesions can occur anywhere in exposed part of the body and mostly these occur over upper extremities, which are prone to trauma during f

In Vitro Evidence for the Efficacy of Manuka Honey and Its Components Against the Major Human Pathogenic Sporothrix Species.

Background/Objectives: While various clinical manifestations occur in sporotrichosis, cutaneous forms predominate. The recommended sporotrichosis treatment is itraconazole, an antifungal with certain restrictions. In recent years, the observation of reduced treatment effectiveness in some patients has arisen, possibly due to Sporothrix spp. resistance mechanisms. Consequently, there is a growing need for alternative therapeutic approaches. This study investigates the antifungal activity of manuka honey (MH) against pathogenic species of the genus Sporothrix. Methods: In this study, we assessed

Virulence assessment of Sporothrix species from clinical and environmental clades using Tenebrio molitor as an experimental model.

Sporotrichosis is a neglected subcutaneous mycosis of global distribution caused by thermally dimorphic Sporothrix species. Despite their pathogenic potential in vertebrates, the virulence mechanisms underlying their interactions with other organisms remain poorly understood. This study employed the coleopteran insect Tenebrio molitor as an experimental model to evaluate the virulence of seven Sporothrix species from clinical (S. brasiliensis, S. schenckii, S. globosa, S. luriei) and environmental (S. pallida, S. mexicana, S. chilensis) clades. Larvae were inoculated with conidia or yeast cell

Sporothrix brasiliensis Atypical Dimorphism in Tissue.

Sporotrichosis, caused by the thermodimorphic fungus Sporothrix brasiliensis, is an emerging zoonotic infection in Brazil and other Latin American countries. Typically, the parasitic form in host tissue is yeast; however, we report three cases in immunocompromised patients exhibiting simultaneous yeast and mycelial structures in biopsies. Identification of S. brasiliensis was confirmed through calmodulin gene sequencing, with phylogenetic analysis supporting species-level classification. Direct examination and histopathology revealed both budding yeast cells (3-8 μm) and hyphae, a rare morphol

Extracutaneous Sporotrichosis With Dissemination to the Brain in a Patient Taking Biologic Medications: A Case Report.

Sporotrichosis is a fungal infection caused by Sporothrix species, primarily Sporothrix schenkii and Sporothrix brasiliensis. Disease manifestation varies from localized cutaneous form to disseminated cutaneous and disseminated extra-cutaneous forms. We present a case of sporotrichosis with widespread skin lesions and fungal dissemination to the brain in a patient taking biologic medications to treat psoriatic arthritis. Her diagnosis was made via positive skin tissue culture, and her symptoms responded well to amphotericin B. With this case report, we would like to highlight the risk of rare

In vitro activity of the anthelmintic drug niclosamide against Sporothrix spp. strains with distinct genetic and antifungal susceptibility backgrounds.

The drugs available to treat sporotrichosis, an important yet neglected fungal infection, are limited. Some Sporothrix spp. strains present reduced susceptibility to these antifungals. Furthermore, some patients may not be indicated to use these drugs, while others may not respond to the therapy. The anthelmintic drug niclosamide is fungicidal against the Sporothrix brasiliensis type strain. This study aimed to evaluate whether niclosamide also has antifungal activity against Sporothrix globosa, Sporothrix schenckii and other S. brasiliensis strains with distinct genotypes and antifungal susce

Combination therapy of itraconazole and an acylhydrazone derivative (D13) for the treatment of sporotrichosis in cats.

UNLABELLED: Acylhydrazone (AH) derivatives represent a novel category of anti-fungal medications that exhibit potent activity against Sporothrix sp., both in vitro and in a murine model of sporotrichosis. In this study, we demonstrated the anti-fungal efficacy of the AH derivative D13 [4-bromo-N'-(3,5-dibromo-2-hydroxybenzylidene)-benzohydrazide] against both planktonic cells and biofilms formed by Sporothrix brasiliensis. In a clinical study, the effect of D13 was then tested in combination with itraconazole (ITC), with or without potassium iodide, in 10 cats with sporotrichosis refractory to

Non-Zoonotic Transmission of Sporotrichosis: A Translational Study of Forty-Three Cases in a Zoonotic Hyperendemic Area.

Over the past two decades, zoonotic sporotrichosis transmitted by naturally infected cats has become hyperendemic in Rio de Janeiro, Brazil. Sporothrix brasiliensis is the main agent involved. However, there are other forms of transmission of sporotrichosis. The aim of this study was to evaluate and associate the epidemiological, clinical and therapeutic data and the susceptibility of Sporothrix spp. to antifungal drugs in 43 non-zoonotic sporotrichosis cases. Forty-three clinical strains of Sporothrix were identified by partial sequencing of the calmodulin gene. An antifungal susceptibility t

First Reported Case of Breast Sporotrichosis in Bahrain.

Granulomatous mastitis is a chronic inflammation of the breast, mostly of unknown etiology. The treatment would be definitive if the causative organism were isolated. It is characterized histologically by granulomas, formed mostly by polymorph nuclear neutrophils and central necrosis. Herein, we report a 46-year-old woman who presented with a non-responding to treatment right breast abscess of six months duration. The abscess was associated with multiple fistula formation. Radiological investigations were inconclusive. She needed incision and drainage many times. The diagnosis of granulomatous

Sporotrichoid Nocardiosis in an Immunocompetent Patient: A Case Report.

Nocardia spp. rarely cause infection in humans and are most common in the immunocompromised population. Pulmonary nocardiosis is the most common presentation. Cutaneous involvement is usually acquired after direct contact. A 71-year-old healthy male patient presented with a non-healing wound in his left leg that had appeared two weeks before. The patient had been engaged in yard work after a hurricane. He developed pain in the left calf, later noticing a small wound attributed to an insect bite or a thorn prick. The lesion exhibited increasing surrounding erythema, purulent discharge, and esca

Laser therapy in the treatment of feline sporotrichosis: A case series.

Sporotrichosis is the most prevalent subcutaneous mycosis in Latin America and is an important zoonosis in expansion throughout all the brazilian territory. Domestic cats are highly susceptible to the disease and play an important role in the spread of the agent to both other animals and humans. Sporothrix brasiliensis, the predominant species in the country, has greater virulence and some isolates also showed resistance to azoles, the class of antifungals of choice for treatment. Because it is a long-duration treatment, of high cost, and oral use, sick animals are often abandoned, which contr

Evaluation of Five Non-Culture-Based Methods for the Diagnosis of Meningeal Sporotrichosis.

Sporotrichosis is the main subcutaneous mycosis worldwide. Several complications, including meningeal forms, can be observed in immunocompromised individuals. The sporotrichosis diagnosis is time-consuming due to the culture's limitations. The low fungal burden in cerebrospinal fluid (CSF) samples is another important drawback in the diagnosis of meningeal sporotrichosis. Molecular and immunological tests can improve the detection of Sporothrix spp. in clinical specimens. Therefore, the following five non-culture-based methods were evaluated for the detection of Sporothrix spp. in 30 CSF sampl
