Development of Maternal Antibodies Post ZIKV in Pregnancy is Associated with Lower Risk of Microcephaly and Structural Brain Abnormalities in Exposed Infants.

BACKGROUND: We investigated the association between maternal neutralizing antibodies (nAb) to Zika virus (ZIKV) in pregnancy and neonatal outcomes. METHODS: In pregnant participants with confirmed ZIKV infection, we determined trimester of infection, collected sera longitudinally, and measured nAbs via plaque reduction. In neonates, adverse outcomes included microcephaly (MC), structural brain abnormalities (SBA), hearing, and eye abnormalities. Associations between trimester of infection, nAbs, and neonatal outcomes were analyzed with Cox regression. RESULTS: In total, 137 ZIKV-positive pre

Prolonged Zika Virus NS1 Protein Circulation in Patient Sera Impacts Clinical Outcome Before the Rise of a Specific IgM Response.

Zika virus (ZIKV) is a neurotropic virus that can be transmitted congenitally. In ZIKV-infected pregnant women, placental dysfunction is associated with the secretion of nonstructural protein 1 (NS1). In this study, the kinetics of NS1 secretion and antibody response were assessed and characterized in the serum of ZIKV-positive adult patients recruited in French Guiana. NS1 concentrations were quantified by a single molecule array (SiMoA) in 164 sequential serum samples collected from thirty patients during the first month after onset of symptoms. Serum NS1 concentrations in this cohort were u

Late-onset hearing loss was not observed among preschool- aged children with prenatal Zika virus exposure: An analysis of the Microcephaly Epidemic Research Group Pediatric Cohort (2015-2019).

Zika virus (ZIKV) infection during pregnancy can lead to Congenital Zika Syndrome (CZS), with significant impacts on early childhood development. This study aimed to analyze the natural history of hearing loss in children with CZS during their first four years of life. Data were collected from the Microcephaly Epidemic Research Group Pediatric Cohort in Pernambuco, Brazil. We investigated whether children with prenatal ZIKV exposure could develop late-onset hearing loss and whether those with preserved auditory thresholds at birth might exhibit deficits in functional hearing or primary languag

A humanized mouse model system mimics prenatal Zika infection and reveals premature differentiation of neural stem cells.

Zika, a mosquito-borne flavivirus, has been found in 87 countries and territories. Global outbreaks peaked in 2016. Prenatal infection of Zika virus was found to be associated with microcephaly, arthrogryposis, intracranial calcifications, fetal growth restriction, and fetal demise. The most severely affected children were diagnosed with congenital Zika syndrome, which impacts thousands worldwide. With no approved treatment or preventative measures for Zika, future viral outbreaks have the potential to cause epidemic levels of prenatal brain injury, as seen over the past 70 years. Therefore, t

Zika virus-induced fetal demise is triggered by strain- and dose-specific RLR-driven activation of the interferon response in the decidua, placenta, and fetus in Ifnar1-/- mice.

UNLABELLED: Congenital Zika syndrome (CZS), the set of fetal and neonatal complications associated with Zika virus (ZIKV) infection in pregnancy, was first noted during the outbreak in the Americas in 2015-2016. However, there was an unequal distribution of ZIKV cases and severe outcomes in all areas where ZIKV emerged in the Americas, demonstrating that the risk of CZS varied over space and time. Recently, we demonstrated that phenotypic heterogeneity existed between closely related ZIKV strains. All ZIKV strains tested infected the placenta but varied in their capacity to cause overt fetal h

Autophagy-mediated downregulation of AXL and TIM-1 promotes sustained Zika virus infection.

Zika virus (ZIKV) infection can lead to a variety of clinical outcomes, including severe congenital abnormalities. The phosphatidylserine receptors AXL and TIM-1 are recognized as critical entry factors for ZIKV in vitro. However, it remains unclear whether and how ZIKV regulates these receptors during infection. In this study, we investigated AXL and TIM-1 expression in human lung adenocarcinoma epithelial A549 cells, glioblastoma U87 cells, and embryonic stem cell-derived trophoblasts following ZIKV infection. We found that both the Asian strain FSS13025 and the African strain MR766 of ZIKV

Bioprofiling and vaccine development for Zika virus.

Zika virus (ZIKV) has emerged as a global health priority due to its association with severe congenital abnormalities, including microcephaly and neonatal mortality in infants born to infected mothers. In adults, ZIKV infection is epidemiologically linked to Guillain-Barré Syndrome (GBS). These clinical manifestations prompted the World Health Organization (WHO) to declare ZIKV a Public Health Emergency of International Concern (PHEIC). The antibody-dependent enhancement (ADE) phenomenon further complicates vaccine development and therapeutic strategies. While substantial progress has been mad

Autosomal recessive primary microcephaly in sibs in time of Zika epidemic: a Case Report.

Case report of two siblings, born to consanguineous parents, with congenital microcephaly secondary to a pathogenic homozygous ASPM gene variant. The proband was born during the Zika virus epidemic with a prenatal history of maternal exanthematous disease. Genetic diagnosis was made after the birth of the sibling, born with a similar condition. Next-generation sequencing enables a definitive diagnosis in cases of microcephaly, and genetic diagnosis should be pursued even when the patient history points to a possible, but not definite, environmental cause. Conclusive genetic diagnosis allows fo

Interleukin-27 is antiviral at the maternal-fetal interface.

Congenital viral infections can have severe consequences for pregnancy and fetal outcomes. Remarkably, the fetal-derived placenta serves as a robust barrier to infection through meticulous regulation by immune effectors and a diverse repertoire of cytokines. Yet, the regulatory roles of many cytokines remain undefined at the maternal-fetal interface. Interleukin 27 (IL-27) is a highly expressed cytokine in the placenta whose functional consequence during congenital infection is unknown. Here, we utilized trophoblast organoids (TO) derived from primary human placentas and a mouse model of conge

Harnessing endogenous miRNA targeting ZIKV: A cutting-edge strategy to inhibit virus infection.

Emerging RNA virus outbreaks, including Zika virus, highlight the urgent need for novel antiviral strategies. Zika virus, a positive-strand RNA virus, causes congenital Zika syndrome, and to date, there are no approved vaccines or antiviral treatments. In this context, microRNAs are small non-coding RNAs that regulate gene expression and show potential as antiviral agents due to their ability to target viral RNA, making them a promising therapeutic approach against Zika syndrome. In this study, we identified endogenous microRNAs that interact with the virus genome using computational algorithm

The human neural cell atlas of Zika virus infection in developing brain tissue.

Zika virus (ZIKV) infection during pregnancy can lead to fetal brain infection and developmental anomalies collectively known as congenital Zika syndrome (CZS). To define the molecular features underlying CZS in a relevant human cell model, we evaluate ZIKV infection in primary human fetal brain explants and human induced pluripotent stem cell-derived mixed neural cultures at single-cell resolution. We identify astrocytes as important innate immune sentinel cells detecting ZIKV and producing interferon-beta (IFN-β). In contrast, neural stem cells display impaired innate immunity and support hi

The unmothering of care: childhood disability, structural violence, and the politicisation of motherhood.

This article traces the intersection of gendered reproductive labour with disability discrimination, through an ethnographic exploration of families raising children with Congenital Zika Syndrome (CZS) in Barranquilla, Colombia. In this paper I will demonstrate how societal ableism intersects with the patriarchal institution of motherhood to shape the experiences of individual mothers raising disabled children. Children living with impairments caused by vertical transmission of Zika (CZS) experience structural violence due to the societal ableism, 'familism' (Gesser et al., 2022) and the neo-l

Quantification of Zika virus using a colloidal gold nanoparticle-based immunosensor and Fourier-transform infrared spectroscopy.

Zika virus (ZIKV) is an arthropod-borne virus that has spread worldwide and is the etiological agent of Zika fever, a disease of global concern distinguished by mild symptoms and recently linked to a congenital syndrome in neonates. Microcephaly, hydrocephalus, ocular deformations, and meningoencephalitis characterize this disease. The plaque assay is the gold standard used in scientific research laboratories to quantify the virus titer on a sample and assess their relationship with the infectious dosage. However, it requires costly infrastructure and specialized technical training. In this st

Comparative Analysis of Two Zika Virus Isolates in a Rhesus Macaque Pregnancy Model.

Zika virus (ZIKV) infection during pregnancy can cause a broad range of neurological birth defects, collectively named Congenital Zika Syndrome (CZS). We have previously shown that infection with the Puerto Rican isolate PRVABC59 (ZIKV-PR) results in abnormal oxygen transport in the placenta due to villous damage and uterine vasculitis in a nonhuman primate model. To investigate whether this type of damage occurs with endemically circulating strains in Thailand, we investigated a CZS case isolate, MU1-2017 (ZIKV-TH), in pregnant rhesus macaques. Pregnant animals (n = 3 per group) were infected

The Evolving Role of Zika Virus Envelope Protein in Viral Entry and Pathogenesis.

Zika virus (ZIKV) was first discovered in Uganda's Zika Forest in 1947. The early African viruses posed little or no health risk to humans. Since then, ZIKV has undergone extensive genetic evolution and adapted to humans, and it now causes a range of human diseases, including neurologically related diseases in adults and congenital malformations such as microcephaly in newborns. This raises a critical question as to why ZIKV has become pathogenic to humans, and what virological changes have taken place and enabled it to cause these diseases? This review aims to address these questions. Specifi

Inhibiting kinesin family member 20A disrupts Zika virus entry by blocking internalization.

Zika virus, a mosquito-borne virus, is associated with congenital birth defects and neurological complications. However, despite its significant public health threat, no approved vaccines or antiviral treatments are currently available. Therefore, this study aims to identify kinesin family member 20A as a key host factor promoting Zika virus life cycle. The elevated expression of kinesin family member 20A following Zika virus infection suggests its role in the viral life cycle. Suppressing its expression through gene silencing or inhibiting its function with a small-molecule inhibitor signific

Association of Toll-like receptor 3 rs3775291 with Zika virus infection in Brazil.

BACKGROUND: Zika virus (ZIKV) is an emergent pathogen known for the outbreak in early 2015, causing significant consequences, of which congenital ZIKV syndrome is the most severe manifestation. Toll-like receptors (TLRs) are important for the appropriate function of the innate immune response against pathogens. The presence of mutations in TLR genes can potentially result in altered susceptibility to diseases. Thus, this study aimed to investigate the influence of TLR2 rs5743708, TLR3 rs3775291, TLR4 rs4986790, TLR4 rs4986791, TLR6 rs5743810 and TLR7 rs179008 polymorphisms in ZIKV infection in
