A decade later, what have we learned from the Zika epidemic in children with intrauterine exposure?

Since the emergence of the Zika virus epidemic in 2014 and the associated novel sequalae that emerged, much has been learned about the effects of antenatal exposure to Zika virus. Zika virus in pregnancy carries severe teratogenic potential to the fetus, ranging from congenital Zika syndrome to milder neurodevelopmental sequelae. Congenital Zika syndrome is associated with a spectrum of alterations that can affect cognitive, language, and motor development. Among children with congenital Zika syndrome, dysphagia and seizures are common, as are hospitalisations for pneumonia and urinary tract i

Travel-related congenital Zika infection associated with Asian lineage virus acquired in Thailand.

Congenital Zika infection is rarely reported in travellers to East Asia. We describe a case of a traveller infected during her ninth week of pregnancy in Thailand. Post-termination analysis revealed a significantly higher viral load in the fetal brain than in maternal blood, highlighting Zika's strong neurotropism and the Thai strain's ability to cause congenital infection.

Adverse fetal and perinatal outcomes associated with Zika virus infection during pregnancy: an individual participant data meta-analysis.

BACKGROUND: Zika virus (ZIKV) infection during pregnancy is associated with an increased risk of congenital malformations. The prevalence of short and long-term consequences, however, remains uncertain due to heterogeneity across studies. Individual Participant Data Meta-Analysis (IPD-MA) offers an alternative approach to provide more precise and generalisable estimates through data harmonisation across studies, allowing for standardised definitions and exploration of heterogeneity. This project was undertaken to estimate absolute and relative risks of adverse outcomes for individuals with ZIK

Building a growing genomic repository for maternal and fetal health through the PING Consortium.

BACKGROUND: Prenatally transmitted viruses can cause severe damage to the developing brain. There is unexplained variability in prenatal brain injury and postnatal neurodevelopmental outcomes, suggesting disease modifiers. Of note, prenatal Zika infection can cause a spectrum of neurodevelopmental disorders, including congenital Zika syndrome. Currently, there is no preventative treatment or cure. The Prenatal Infection and Neurodevelopmental Genetics (PING) Consortium aims to identify modulators of brain injury and adverse neurodevelopmental outcomes for Zika and other prenatal viral infectio

Does gastrostomy make a difference in children with congenital Zika syndrome growth?

INTRODUCTION: Children diagnosed with congenital Zika syndrome often experience stomatognathic disorders that may compromise caloric intake via oral feeding. OBJECTIVE: The aim of this study was to assess the effects of gastrostomy on the growth and nutritional parameters of children with congenital Zika syndrome. METHODS: This prospective cohort study included children with congenital Zika syndrome who underwent gastrostomy and were followed up by the Center of Support for Children with Microcephaly in the state of Paraiba, in northeast Brazil. The study was conducted between April 2019 and

Microcephaly protein ANKLE2 promotes Zika virus replication.

Orthoflaviviruses are positive-sense single-stranded RNA viruses that hijack host proteins to promote their own replication. Zika virus (ZIKV) is infamous among orthoflaviviruses for its association with severe congenital birth defects, notably microcephaly. We previously mapped ZIKV-host protein interactions and identified the interaction between ZIKV non-structural protein 4A (NS4A) and host microcephaly protein ankyrin repeat and LEM domain-containing 2 (ANKLE2). Using a fruit fly model, we showed that NS4A induced microcephaly in an ANKLE2-dependent manner. Here, we explore the role of ANK

Induction of systemic and mucosal immune response against Zika virus by vaccination with non-infectious chimeric VLPs.

The Zika virus (ZIKV) causes acute febrile illness and can lead to complications such as Guillain-Barré syndrome and congenital disorders. As arbovirus outbreaks increase, vaccination becomes a crucial preventive strategy. Currently, no commercial vaccines are available for ZIKV, which is transmitted by mosquitoes and bodily fluids, underscoring the need for a safe vaccine that induces both systemic and mucosal immune responses. In this study, we present a ZIKV vaccine candidate utilizing virus-like particles (VLPs) technology combined with variant-specific surface proteins (VSP) from Giardia

A case-control neuroimaging investigation of chronic Zika virus-infected adults.

Systemic viral infections with neurotropic potential pose significant global health challenges. The Zika virus (ZIKV) is known for its pronounced neurotropism, with recent infectious clusters raising renewed public health concerns. While research has predominantly focused on congenital populations, growing evidence suggests that the mature central nervous system (CNS) is also vulnerable. However, no study has examined the long-term impact of ZIKV infection on the adult human brain. To address this gap, we studied a rare group of adult ZIKV patients presenting with both peripheral (Guillain-Bar

Discovery of orally bioavailable Zika and West Nile Virus antiviral compounds targeting the NS2B-NS3 protease.

Flaviviruses are a class of pathogenic viruses with pandemic potential which are typically transmitted via infected arthropods. In particular, Zika virus is sexually transmissible and causes congenital malformations if infection occurs during pregnancy. Although over 1.5 million people were infected during the 2015-2016 Zika outbreak, to date, there are no clinical-stage vaccines or antivirals. Herein, we report the discovery of potent inhibitors of the Zika virus NS2B-NS3 protease that also show activity against the West Nile virus NS2B-NS3 protease. Starting from a crystallographic fragment

A Review of the Recent Epidemiology of Zika Virus Infection.

Zika virus (ZIKV) is a flavivirus transmitted primarily by the bite of infected Aedes species mosquitoes. Although typically asymptomatic or causing mild symptoms and infrequent neurological disease in older children and adults, infection during pregnancy can result in severe congenital malformations and neurodevelopmental deficits. We conducted a review of published literature and official data sources to describe recent Zika epidemiological trends, building on WHO updates posted in 2019 and 2022. Globally, cases declined after the height of ZIKV transmission in the Americas in 2015-2016; how

Unraveling Zika virus-induced cell death pathways in eradicating embryonal central nervous system tumors.

The Zika virus (ZIKV) has gained attention due to its link with causing congenital syndrome, which is characterized by microcephaly as a significant symptom. The neurotropism of the virus is not limited to neural progenitor cells; ZIKV has also been tested as an oncolytic virus, given its affinity for tumor cells in the central nervous system. Understanding the cell death pathway involved in eliminating tumor cells is crucial to determining whether ZIKV triggers a silent or immunogenic response in the nervous tissue, which could potentially harm the surrounding healthy tissue. Hence, this stud

Disruption of functional network development in children with prenatal Zika virus exposure revealed by resting-state EEG.

Children born to mothers infected by Zika virus (ZIKV) during pregnancy are at increased risk of adverse neurodevelopmental outcomes including microcephaly, epilepsy, and neurocognitive deficits, collectively known as Congenital Zika Virus Syndrome. To study the impact of ZIKV on infant brain development, we collected resting-state electroencephalography (EEG) recordings from 28 normocephalic ZIKV-exposed children and 16 socio-demographically similar but unexposed children at 23-27 months of age. We assessed group differences in frequency band power and brain synchrony, as well as the relation

Role of non-human primate models in accelerating research and developing countermeasures against Zika virus infection.

Zika virus, a mosquito-transmitted orthoflavivirus, has become a pathogen of global health concern ever since the virus caused an epidemic in Brazil in 2015 associated with approximately 700 000 laboratory-confirmed cases of congenital microcephaly. The subsequent spread of the epidemic in 2016 resulted in a wide spectrum of congenital neurological, ophthalmological, and developmental abnormalities across the Americas, Africa, and Asia. In this context, non-human primate models have become essential tools for Zika virus research to understand the pathogenesis of congenital brain injury and per

Zika virus: advancing a priority research agenda for preparedness and response.

The 2015-16 Zika virus epidemic emerged in the Americas and rapidly spread throughout the region and beyond, showing the epidemic potential of this mosquito-borne Orthoflavivirus and its capacity to cause severe congenital malformations and neurological sequelae. WHO declared the Zika virus epidemic a public health emergency of international concern in 2016. Despite this declaration, there are no licensed Zika virus vaccines, therapeutics, or diagnostic tests appropriate for routine antenatal screening. To address this absence of essential tools to detect and mitigate the threat of future Zika

Neferine inhibits Zika virus replication by targeting viral protease.

BACKGROUND: Zika virus (ZIKV) belonging to the Flaviviridae family causes critical neurological abnormalities, including congenital microcephaly and Guillain-Barré syndrome. Despite being a major public health concern, effective therapeutic interventions against ZIKV remain unavailable. NS2B-NS3 (ZIKVpro), the viral serine protease critical for polyprotein processing as well as viral replication, is a potential target for developing antiviral agents PURPOSE: This study aims to develop ZIKV inhibitors targeting the ZIKVpro from natural products. METHODS: Surface plasmon resonance (SPR) based a

Relative and attributable risks of neurological and perinatal adverse outcomes among children with and without prenatal Zika virus exposure in Northeast Brazil: A prospective cohort study (2015-2018).

BACKGROUND: Although there has been substantial progress in the characterization of Congenital Zika Syndrome, the lack of a control group in the majority of published studies on Zika virus (ZIKV) infections during pregnancy limits our understanding of, first, the magnitude by which prenatal ZIKV exposure may increase risks of adverse outcomes for offspring and, second, the fraction of abnormalities that are attributable to this exposure. METHODS: To overcome this limitation, this study harmonized and integrated data collected prospectively in Recife, Pernambuco, Brazil, from offspring of ZIKV

Structured and specialized physical therapy: Does it make a difference for children with congenital Zika syndrome? A pilot randomized controlled trial.

Interventions for improving motor function of children with congenital Zika syndrome (CZS) are still underexplored. This study aimed to evaluate the feasibility of a structured and specialized 12-week intervention based on the Bobath concept and Pediasuit protocol compared to no change in therapeutic routine on the motor function of children with CZS. This pilot randomized controlled trial was conducted in a support center for children with microcephaly in Brazil between February and May 2018. Sixteen children were included. Anthropometric variables and motor functions (GMFM-88) were assessed.

Congenital Zika virus infection in laboratory animals: a comparative review highlights translational studies on the maternal-foetal interface.

The 2015-16 Zika virus (ZIKV) epidemic has posed unprecedented concern for maternal-infant health, mainly due to the substantial risk of microcephaly and other neurological birth abnormalities associated with congenital ZIKV syndrome (CZS). As licenced vaccines and effective antivirals are still unavailable, attention has been focused on post-delivery in vitro or translational in vivo studies to understand the impact of maternal ZIKV infection on placentation and neurodevelopmental consequences for the foetus. Here, we review clinical and translational studies highlighting ZIKV-induced materna
