Congenital toxoplasmosis is a significant public health issue caused by the transplacental passage of Toxoplasma gondii to the embryo/fetus. The standard treatment involves a combination of sulfadiazine and pyrimethamine, drugs often associated with adverse effects and high toxicity. The current study aimed to investigate the potential of prenylated chalcones (C2, C4 and C9) in controlling T. gondii infection in human trophoblast cells (BeWo) and human placental explants. As results, non-cytotoxic doses of C2, C4 and C9 impaired parasite invasion and subsequent intracellular proliferation in B

Acute infection with Toxoplasma gondii in pregnant people can lead to vertical transmission to the foetus and congenital toxoplasmosis. As part of risk assessment, the epidemiology of toxoplasmosis among pregnant people must be quantitatively elucidated. Herein, we investigated the risk of primary T. gondii infection during pregnancy in Japan, estimating the incidence of T. gondii infection among pregnant people as well as that of congenital toxoplasmosis. We used a compartment model that captured the infection dynamics in pregnant people, analysing prescription data for spiramycin in Japan, t

BACKGROUND AND AIMS: Congenital toxoplasmosis can cause severe diseases to the fetus and newborns or abortion, and is associated with significant morbidity and mortality. In Burkina Faso, while seroprevalence data for toxoplasmosis are available for urban areas, such data are scarce for rural areas. This study aims to determine the seroprevalence of toxoplasmosis in pregnant women attending antenatal services in primary health facilities in rural and urban areas.
METHODS: A cross-sectional study was conducted between July 2022 and June 2023 in health facilities located in the Cascades (n = 2)

Congenital toxoplasmosis presents with a wide clinical spectrum, ranging from asymptomatic infection to severe disease with multi-organ failure. We report a rare fatal case of disseminated congenital toxoplasmosis in a human neonate. The infant initially had thrombocytopenia and mild hepatitis, which rapidly progressed to fulminant liver failure. Despite initiation of standard therapy with pyrimethamine, sulfadiazine, and folinic acid on postnatal day 25, the infant died two days later. Autopsy revealed widespread involvement of the liver, spleen, brain, heart, lungs, urinary bladder, and skel

To estimate the performance of anti-Toxoplasma IgA assay for the diagnosis of congenital toxoplasmosis, a retrospective monocenter study was conducted comparing serological results obtained in the framework of routine diagnosis workup. All infants born to mothers infected with Toxoplasma gondii during pregnancy from 2010 to 2023 with at least 6 months of serological follow-up were included. Four hundred and eighty-three cases (1,171 sera) were included, of which 56 infants (11.6%) were infected. Twenty out of 60 infants (33.3%) with positive IgA were not infected. The sensitivity to detect IgA

Toxoplasma gondii is a globally distributed protozoan parasite and a major cause of congenital infections, particularly in South America. Current therapies for congenital toxoplasmosis are limited by toxicity, long treatment regimens, and suboptimal efficacy, highlighting the urgent need for safer and more effective alternatives. In this study, we evaluated the antiparasitic effects of crude ethanolic extract of Brazilian Red Propolis (BRP) and its isolated compounds, focusing on 7-O-methylvestitol, in human trophoblast (BeWo) cells and third-trimester placental explants. Both BRP and 7-O-meth

Childbearing-aged and pregnant women represent a risk group for Toxoplasma gondii infection due to possible transplacental transmission resulting in congenital toxoplasmosis. We analyzed the seroepidemiological trends of toxoplasmosis in Croatia over ten years (2015-2024). A total of 2791 childbearing-aged and pregnant women were included. Toxoplasma gondii-specific IgM/IgG antibodies were detected using an enzyme-linked fluorescence assay. Samples with positive IgM and IgG antibodies were tested for IgG avidity. IgG antibodies were detected in 695 (24.9%) participants, while acute toxoplasmos

Background:Toxoplasma gondii (T. gondii) infection causes serious diseases in immunocompromised patients and causes congenital toxoplasmosis in infants. T. gondii microneme protein 8 (MIC8) and apical membrane antigen 1 (AMA1) are essential proteins involved in parasitic invasion. Methods: In this study, we generated virus-like particles (VLPs) and recombinant vaccinia virus (rVV) containing MIC8 or AMA1 proteins. Vaccine efficacy was evaluated in mice (BALB/c) upon challenge infection with T. gondii ME49. Results: Intramuscular immunization with heterologous vaccines (rVV + VLPs; rVV for prim

The protozoan Toxoplasma gondii is a ubiquitous and highly prevalent parasite that can theoretically infect all warm-blooded vertebrates. In humans, toxoplasmosis causes infections in both immunodeficient and immunocompetent patients, congenital toxoplasmosis, and ocular lesions. These manifestations have different degrees of severity. Clinical severity is determined by multiple factors, including the genotype of the T. gondii strain involved in the infection. T. gondii exhibits remarkable genetic diversity, which varies according to geography and ecotype (domestic or wild). Previous studies h

OBJECTIVE: To describe two severe cases of congenital toxoplasmosis in infants born to chronically infected mothers who did not receive education or information on the prevention of gestational toxoplasmosis during prenatal care.
CASE DESCRIPTION: The mothers had a previous serological diagnosis of toxoplasmosis conducted during prenatal care, with non-reactive (<10 IU/mL) IgM and reactive IgG (>10 IU/mL), and were considered "immune" to the infection. Both infants were born with sequelae of the congenital infection, including neurological and ocular alterations.

Toxoplasma gondii, one of the most prevalent zoonotic parasites globally, may be transmitted to the fetus if a primary infection occurs during pregnancy. The earlier the gestational period, the lower the probability of transmission, but if it occurs, the higher the probability of pregnancy loss or severe fetal neurological or ocular damage. Primary prevention measures to pregnant women include avoiding eating or handling raw or undercooked meat, washing hands thoroughly after gardening, avoid handling contaminated soil or water or come in contact with cat feces. The infected fetus may exhibit

Congenital toxoplasmosis (CT) due to Toxoplasma gondii, an apicomplexan parasite, leads to significant sequelae in children, foetal losses and neonatal deaths worldwide. This study aimed to assess the burden of CT in Burundi for the year 2020. We used epidemiological and economic data collected in major hospitals in Burundi, the Ministry of Public Health in Burundi and international peer-reviewed literature to estimate the disability-adjusted life years (DALYs) and economic costs spent on prenatal consultations, diagnosis and treatment of toxoplasmosis for pregnant women. In Burundi, the incid

Toxoplasmosis during pregnancy can cause congenital malformations and fetal death. This study aimed to estimate the Toxoplasma gondii seroprevalence among pregnant women participating in the 2021 French national perinatal survey and identify associated factors. All women giving birth in France during the study period were invited to participate. Data collected included demographic information, nationality, socio-economic status, education level, and Toxoplasma gondii serological status. Women were classified as seropositive if IgG antibodies were present or if seroconversion occurred during pr
