Allergic load and symptom severity in children with tic disorders: a cross-sectional study.

UNLABELLED: Tic disorders (TD) are associated with allergies, but the immune mechanisms and their impact on tic severity remain unclear. This study examined the immune profile in children with comorbid TD and allergic diseases (AD) to explore the graded association between allergic load index and tic severity. A total of 136 children with TD (aged 4 - 13 years) were divided into TD (n = 76) and TD + AD (n = 60) groups. Serum IgE levels were measured, tic severity was assessed by the Yale Global Tic Severity Scale (YGTSS), and a composite allergic load index was constructed. Its graded associat

Paradoxical viper envenomation: minimal local manifestations with fatal intracranial hemorrhage- an autopsy case report.

Viper envenomation typically presents with prominent local manifestations, including pain, edema, ecchymosis, cellulitis, and tissue necrosis. Severe systemic toxicity in the absence of significant local signs is uncommon. We report an atypical case of a viper envenomation in a 50-year-old male who presented with minimal local manifestations but rapidly developed life-threatening systemic complications. The 20-min whole-blood clotting test was non-clotting on admission. Despite prompt administration of polyvalent anti-snake venom (ASV) and symptomatic management, his condition progressively de

Blood Glial Fibrillary Acidic Protein in MRI-Negative Attack-Like Episodes of Neuromyelitis Optica Spectrum Disorder.

OBJECTIVES: Magnetic resonance imaging (MRI) is widely used to support relapse diagnosis in neuromyelitis optica spectrum disorder (NMOSD), yet some patients present with attack-like symptoms without corresponding lesions. We assessed serum glial fibrillary acidic protein (sGFAP) levels in MRI-negative attack-like episodes. METHODS: We retrospectively analyzed aquaporin-4-IgG-positive NMOSD patients presenting with relapse-like symptoms but without corresponding new MRI lesions. sGFAP was measured from stored serum collected at presentation and converted to demographic-adjusted z-scores. Hypertensive disorders of pregnancy: epidemiology, pathophysiology, therapeutic strategies, and maternal-offspring outcomes.

Hypertensive disorders of pregnancy (HDP) encompass chronic hypertension, gestational hypertension, preeclampsia, and preeclampsia superimposed on chronic hypertension, affecting about 5% to 10% of pregnancies worldwide and remaining a major cause of maternal and perinatal morbidity and mortality. Their burden is increasing with rising obesity, advanced maternal age, evolving diagnostic criteria, and marked socioeconomic and racial-ethnic disparities. This review summarises current evidence on the epidemiology, pathophysiology, biomarkers, clinical management, and long-term outcomes of HDP, hi

Distinct insulin sensitivity profiles revealed by clustering based on continuous glucose monitoring data in people living with type 1 diabetes.

BACKGROUND: Continuous glucose monitoring (CGM) has improved glycemic management in Type 1 Diabetes Mellitus (T1DM). However, interpretation of CGM data remains challenging. CGM data clustering may support individualized management and identify insulin resistance. AIM: This study aimed to cluster CGM data based on interstitial glucose levels (2.2-27.8 mmol/L) and glucose variability, and to compare the identified clusters with biochemical and anthropometric markers of insulin sensitivity. METHODS: CGM data from 75 T1DM patients and 20 subjects without diabetes were analyzed using Uniform Man

Sex differences in a mouse model of radiation-induced cardiomyopathy.

BACKGROUND: Radiation-induced heart disease (RIHD) may develop months to years following radiation therapy for cancers near the heart such as lung or breast cancer. Several mouse models of RIHD exist to study the pathogenesis of disease, but to our knowledge no studies have examined whether sex differences exist in RIHD in mice. METHODS: Female and male BALB/cJ mice received a single whole-heart dose of 22 Gy (Gy) radiation via a C5 collimator vs. sham controls that received 0 Gy, anesthesia and were placed in the X-ray machine for the same length of time. Endpoints were examined at 24 h (inn

Sustained type I interferon activity and N-antigenemia form a vicious cycle during severe COVID-19.

BACKGROUND: Although antigenemia is associated with poor outcomes in respiratory viral infections, the underlying mechanisms driving viral bloodstream dissemination remain not fully understood. Herein, we aimed to characterise SARS-CoV-2 nucleocapsid (N)-antigenemia kinetics in patients with severe COVID-19. We also sought to identify host and viral determinants associated with viral dissemination through a multiparametric integrated analysis and assess the associated risk of death. METHODS: This observational study included hospitalised patients with severe COVID-19 across three independent

Loss of the human BRIP1 C terminus does not cause Fanconi anemia.

Fanconi anemia (FA) is a genetic disorder characterized by congenital anomalies, progressive bone marrow failure, and an increased cancer risk. FA patient cells are hypersensitive to DNA interstrand crosslinks (ICLs), which cause chromosomal and genomic instability if left unrepaired. We identified 11 individuals from seven families who are homozygous for one of two C-terminally truncating variants in BRIP1 (previously known as FANCJ), c.2947del (p.Ile983Leufs∗2) (n = 3) or c.2992_2995del (p.Lys998Glufs∗60) (n = 8). None of them exhibited hematologic manifestations of FA, including bone marrow
