In vitro inhibitory activity of indole alkaloid derivatives against porcine epidemic diarrhea virus.

Porcine epidemic diarrhea virus (PEDV) is an enteric coronavirus that can cause acute diarrhea, vomiting, dehydration, and high mortality of newborn piglets, leading to huge economic losses to the world pig industry. Given the limited efficacy of current PEDV vaccines, there is an urgent need for the development of antiviral drugs. In this study, the antiviral effects of 17 synthesized indole alkaloid derivatives against PEDV were investigated. It was observed that indole alkaloid derivative no. 14 exhibited significant inhibition of PEDV replication in a dose-dependent manner. Furthermore, ti

Prenatal Renal Vein Thrombosis.

Background: The neonatal period has a number of characteristics leading to an increased risk of severe and, in many cases, life-threatening complications. Renal venous thrombosis is one of them. It accounts for 16-20% of all thromboembolisms in the neonatal period. Due to the delicate balance in coagulation status in the first days after birth, conditions such as infections, hypoxia, hypotension, and dehydration can lead to the occurrence of this complication. The incidence of renal thrombosis is 2.2/100,000 live births, with cases of intrauterine renal thrombosis being even rarer (7% of cases

Nursing Management of Humidification Levels for the Extremely Low Birth Weight Infant: A Process Improvement Protocol to Reduce Hypernatremia in the First 2 Weeks of Life.

The use of a humidified environment has long been established as a method of maintaining a thermoneutral environment and minimizing evaporative insensible water losses in the extremely low birth weight (ELBW) population. The ELBW infant is at high risk for insensible water losses leading to dehydration, excessive weight loss, temperature instability, and electrolyte abnormalities, thereby increasing the ELBW infant's risks of morbidity and mortality. Literature supports the use of a humidified environment; however, a lack of consensus has led to widespread variation in the implementation of hu

Clinico-Epidemiological Characteristics of Symptomatic and Asymptomatic Enterotoxigenic and Enteropathogenic Escherichia Coli Diarrhea and Impact on Child Growth.

Enterotoxigenic Escherichia (E.) coli (ETEC) and enteropathogenic E. coli (EPEC) are major bacterial causes of childhood diarrhea. We explored the clinico-epidemiological characteristics of children aged <5 years associated with moderate-to-severe diarrhea (MSD), asymptomatic ETEC or EPEC infections, and subsequent impact on growth reflected by z-score. Data from 9,439 MSD and 13,128 asymptomatic children were extracted from Global Enteric Multicenter Study, conducted between 2007 and 2011. Epidemiological risk factors and clinical characteristics of ETEC and EPEC infection were explored using

Extreme heat and pediatric health in a warming world: a space-time stratified case-crossover investigation in Ontario, Canada.

BACKGROUND: Globally, climate change is causing frequent and severe extreme heat events (EHEs). A large body of literature links EHEs to multiple health endpoints. While children's physiology and activity patterns differ from those of adults in ways that are hypothesized to increase susceptibility to such endpoints, research gaps remain regarding the specific impacts of EHEs on child health. This study evaluated pediatric emergency healthcare utilizations associated with EHEs in Ontario. METHODS: Applying a space-time stratified case-crossover design, associations between EHEs (same-day or la

Distinct neural representations of hunger and thirst in neonatal mice before the emergence of food- and water-seeking behaviors.

Hunger and thirst are two fundamental drives for maintaining homeostasis and elicit distinct food- and water-seeking behaviors essential for survival. For neonatal mammals, however, both hunger and thirst are sated by consuming milk from their dams. While distinct neural circuits underlying hunger and thirst drives in the adult brain have been characterized, it is unclear when these distinctions emerge in neonates and what processes may affect their development. Here, we show that hypothalamic hunger and thirst regions already exhibit specific responses to starvation and dehydration well befor

Complex Medium-Chain Triglycerides Mitigate Porcine Epidemic Diarrhea Virus Infection in Piglets by Enhancing Anti-Inflammation, Antioxidation, and Intestinal Barrier Function.

Porcine epidemic diarrhea (PED), a highly contagious enteric disease caused by the porcine epidemic diarrhea virus (PEDV), is characterized by vomiting, diarrhea, and dehydration, leading to high mortality in newborn piglets and significant economic losses in the swine industry. The shortage of effective PED vaccines emphasizes the need to explore potent natural compounds for therapeutic intervention. It has been shown that glycerol monolaurate (GML) effectively inhibits PEDV replication in vivo and in vitro. Further investigation is needed to assess whether complex medium-chain triglycerides

Call to action: recognize and prevent the effects of extreme heat on early childhood development and health.

The frequency and severity of heat waves are expected to worsen with climate change. Exposure to extreme heat, or prolonged unusually high temperatures, are associated with increased morbidity and mortality. The fetus, infant, and young child are more sensitive to higher temperatures than older children and most adults given that they are rapidly developing. During pregnancy, exposure to extreme heat may result in dehydration, inflammation, and reduced blood flow in the placenta potentially triggering preterm birth and increased rates of stillbirth and low birth weight infants. Young children

Development of a Safe and Effective mRNA Candidate Vaccine Against PEDV G2c Genotype Infection.

Porcine epidemic diarrhea virus (PEDV) is a highly contagious coronavirus that causes severe diarrhea, dehydration, and high mortality in piglets, leading to significant economic losses in the swine industry. The spike (S) protein of PEDV is the primary target for neutralizing antibodies and is critical for vaccine development. In this study, the pUC57-S01 and pUC57-S02 plasmids carrying the codon-optimized truncated S gene sequence were constructed. The mRNA S01 showed higher protein expression in vitro than mRNA S02, as confirmed by Western blotting. The safety and immunogenicity of mRNA S01

Study protocol of "A preterm infant life-saving package including a simple and affordable incubator and a portable flow generator mask-CPAP in resource-limited settings" SAVE preterm trial, saving preterm infants by adopting vital equipment.

INTRODUCTION: Despite a significant global decline in the mortality rate for children under the age of 5, the neonatal mortality rate (NMR) has not significantly improved over the last thirty years. Globally, 250 million newborn babies die each year, and 98% of the neonatal deaths occur in middle- and low-income countries. The leading cause of death is preterm birth; and complications due to the prematurity of the infant include respiratory failure, hypothermia, infections, and dehydration. Treatment for these complications may require respiratory support devices, temperature management using

Awareness of neonatal heat illness: a fatal outcome in a late preterm infant.

Extreme heat impacts on child health and well-being due to immature thermoregulation, less sweating and a high surface-to-body mass ratio that make them more vulnerable to heat-related illness. Severe dehydration, especially if struggling with breastfeeding, renal failure and electrolyte imbalance, and neurological and cardiorespiratory failure may result. This late preterm was born healthy during a heatwave and developed lethargy and minimal responsiveness from day of life (DOL) 5, developed seizures at DOL 12, then demised at DOL 20. All hospitals involved in the diagnostic workup and treatm

De Novo KCNJ11 Mutation in an Infant With Neonatal Diabetes Mellitus Presenting as Diabetic Ketoacidosis: A Case Report and Literature Review.

Neonatal diabetes mellitus (NDM) is a rare metabolic disorder that develops within the first 6 months of life and can have a wide clinical presentation which includes diabetic ketoacidosis (DKA). We describe a 56-day-old female with permanent NDM whose clinical presentation included polyuria, fever, vomiting, and dehydration. Laboratory workup indicated DKA. The patient started her treatment with subcutaneous insulin, but her blood sugar level was poorly controlled. She was later found to have a KCNJ11 mutation and was subsequently switched to sulfonylurea, which offered better control of bloo
