Distinct neural representations of hunger and thirst in neonatal mice before the emergence of food- and water-seeking behaviors.

Hunger and thirst are two fundamental drives for maintaining homeostasis and elicit distinct food- and water-seeking behaviors essential for survival. For neonatal mammals, however, both hunger and thirst are sated by consuming milk from their dams. While distinct neural circuits underlying hunger and thirst drives in the adult brain have been characterized, it is unclear when these distinctions emerge in neonates and what processes may affect their development. Here, we show that hypothalamic hunger and thirst regions already exhibit specific responses to starvation and dehydration well befor

Intraperitoneal fluid administration to neonatal calves.

OBJECTIVE: To determine if Intraperitoneal fluid therapy (IPFT) is an effective rehydration therapeutic option for dehydrated neonatal calves. Perceived risks and unknown efficacy have limited IPFT implementation into practice. It is hypothesized that IPFT will be an efficient and safe method of fluid administration to healthy neonatal calves. METHODS: 4 L of isotonic saline was administered IP to healthy calves (n = 4). Control animals received no treatment (n = 2). Clinical and blood parameters were collected before and after fluid administration in all animals. RESULTS: Girth diameter and

Complex Medium-Chain Triglycerides Mitigate Porcine Epidemic Diarrhea Virus Infection in Piglets by Enhancing Anti-Inflammation, Antioxidation, and Intestinal Barrier Function.

Porcine epidemic diarrhea (PED), a highly contagious enteric disease caused by the porcine epidemic diarrhea virus (PEDV), is characterized by vomiting, diarrhea, and dehydration, leading to high mortality in newborn piglets and significant economic losses in the swine industry. The shortage of effective PED vaccines emphasizes the need to explore potent natural compounds for therapeutic intervention. It has been shown that glycerol monolaurate (GML) effectively inhibits PEDV replication in vivo and in vitro. Further investigation is needed to assess whether complex medium-chain triglycerides

Call to action: recognize and prevent the effects of extreme heat on early childhood development and health.

The frequency and severity of heat waves are expected to worsen with climate change. Exposure to extreme heat, or prolonged unusually high temperatures, are associated with increased morbidity and mortality. The fetus, infant, and young child are more sensitive to higher temperatures than older children and most adults given that they are rapidly developing. During pregnancy, exposure to extreme heat may result in dehydration, inflammation, and reduced blood flow in the placenta potentially triggering preterm birth and increased rates of stillbirth and low birth weight infants. Young children

Development of a Safe and Effective mRNA Candidate Vaccine Against PEDV G2c Genotype Infection.

Porcine epidemic diarrhea virus (PEDV) is a highly contagious coronavirus that causes severe diarrhea, dehydration, and high mortality in piglets, leading to significant economic losses in the swine industry. The spike (S) protein of PEDV is the primary target for neutralizing antibodies and is critical for vaccine development. In this study, the pUC57-S01 and pUC57-S02 plasmids carrying the codon-optimized truncated S gene sequence were constructed. The mRNA S01 showed higher protein expression in vitro than mRNA S02, as confirmed by Western blotting. The safety and immunogenicity of mRNA S01

Study protocol of "A preterm infant life-saving package including a simple and affordable incubator and a portable flow generator mask-CPAP in resource-limited settings" SAVE preterm trial, saving preterm infants by adopting vital equipment.

INTRODUCTION: Despite a significant global decline in the mortality rate for children under the age of 5, the neonatal mortality rate (NMR) has not significantly improved over the last thirty years. Globally, 250 million newborn babies die each year, and 98% of the neonatal deaths occur in middle- and low-income countries. The leading cause of death is preterm birth; and complications due to the prematurity of the infant include respiratory failure, hypothermia, infections, and dehydration. Treatment for these complications may require respiratory support devices, temperature management using

Awareness of neonatal heat illness: a fatal outcome in a late preterm infant.

Extreme heat impacts on child health and well-being due to immature thermoregulation, less sweating and a high surface-to-body mass ratio that make them more vulnerable to heat-related illness. Severe dehydration, especially if struggling with breastfeeding, renal failure and electrolyte imbalance, and neurological and cardiorespiratory failure may result. This late preterm was born healthy during a heatwave and developed lethargy and minimal responsiveness from day of life (DOL) 5, developed seizures at DOL 12, then demised at DOL 20. All hospitals involved in the diagnostic workup and treatm

De Novo KCNJ11 Mutation in an Infant With Neonatal Diabetes Mellitus Presenting as Diabetic Ketoacidosis: A Case Report and Literature Review.

Neonatal diabetes mellitus (NDM) is a rare metabolic disorder that develops within the first 6 months of life and can have a wide clinical presentation which includes diabetic ketoacidosis (DKA). We describe a 56-day-old female with permanent NDM whose clinical presentation included polyuria, fever, vomiting, and dehydration. Laboratory workup indicated DKA. The patient started her treatment with subcutaneous insulin, but her blood sugar level was poorly controlled. She was later found to have a KCNJ11 mutation and was subsequently switched to sulfonylurea, which offered better control of bloo

Neonatal Hypernatremic Dehydration.

Newborn infants experience significant fluid shifts following delivery as a result of the contraction of extracellular water. Without prompt identification and treatment, significant weight loss can result in neonatal hypernatremic dehydration. This case study presents an infant with significant weight loss, dehydration, and electrolyte imbalances. The differential diagnosis for hypernatremia is presented along with clinical presentation, treatment, and discharge education recommendations.

Effect of pre-weaning glycyl-glutamine feeding on diarrhea rate, hindgut fermentation, antioxidant stress capability, and fecal microbiota in calves.

Glycyl-glutamine (Gly-Gln, GQ) is formed by the dehydration-condensation reaction of glycine and glutamine. Different studies have shown a positive effect of GQ on gut health and growth performance in early neonatal animals. We evaluated oral GQ effects in Holstein calves on growth (body weight, average dairy gain), fecal score, diarrhea rate, serum biomarkers (antioxidants/inflammation/biochemistry/amino acids), and hindgut parameters (fermentation and microbiota). Twenty-two neonatal Holstein heifer calves (1-5 d of age; 36.93 ± 1.052 kg) from multiparous dams were randomly divided into cont

The integrin protein ITGβ1 effectively suppresses porcine epidemic diarrhea virus replication through facilitating MDA5 oligomerization and subsequent activation of the type I interferon signaling pathway.

Integrins are cell surface adhesion molecules. They bridge the intracellular and extracellular environments, enabling bidirectional transmembrane signaling and regulating immune responses. However, it remains unclear whether integrin protein β1 (ITGβ1) is involved in innate immune responses. In our previous study, we demonstrated that porcine epidemic diarrhea virus (PEDV) can induce type I interferon (IFN-I) production. In this study, we observed that ITGβ1 expression is rapidly induced following PEDV infection and further established that PEDV infection primarily promoted ITGβ1 expression th

Human induced pluripotent stem cells for in vitro modeling of impaired mucociliary clearance in cystic fibrosis lung disease.

Severely impaired mucociliary airway function is the primary pathomechanism in Cystic Fibrosis (CF) lung disease. Despite significant advances in CF therapy, there is still a critical need for alternative, individualized treatment options, especially for patients with untreatable CFTR mutations. Although intestinal organoids and primary airway cells are widely used as preclinical models of CF, both systems exhibit limitations with regard to the proper modelling of mucociliary clearance or the availability of sufficient cell quantities. Patient-specific human induced pluripotent stem cells (hiP

Incidence and predictors of dehydration among preterm neonates admitted to the neonatal intensive care unit of Southern Ethiopia hospitals: A prospective follow-up study.

BACKGROUND: Dehydration is one of the causes of preterm morbidity and mortality. Many efforts are being implemented to decrease the cause of preterm mortality, but there is a gap regarding dehydration in generating research-supported evidence. Therefore, this study aimed to investigate the incidence and predictors of dehydration among preterm neonates admitted to the neonatal intensive care units of Southern Ethiopia hospitals. METHODOLOGY: A prospective, institution-based follow-up study was conducted among 363 maternal-preterm neonatal pairs. Data were collected using a checklist and entere

Evaluation of diarrhea management appropriateness and its determinants among children under five years of age in regional and national referral hospitals in Eritrea: a cross-sectional study.

BACKGROUND: Diarrhea is a major global public health concern, accounting for 18% of deaths in children under five. Timely and appropriate management at both household and healthcare levels is crucial for reducing mortality associated with childhood diarrhea. However, adherence to recommended guidelines for managing childhood diarrhea is often suboptimal worldwide. This study aimed to assess the overall appropriateness of diarrhea management and its associated factors in children under five, with or without co-morbid illnesses. METHODS: Hospital-based cross-sectional study was conducted. Data
