Mitochondria as a therapeutic target in neurodegeneration caused by hypoxia and ischemia during the perinatal period.

Perinatal hypoxia, also known as neonatal hypoxia-ischemia (HI), can pose a significant threat to the life and health of newborns, leading to hypoxic-ischemic encephalopathy (HIE) and numerous organ dysfunctions, including the nervous system. Mitochondria, which are key organelles in maintaining cellular homeostasis, adenosine triphosphate (ATP) production, regulation of apoptosis, and the cell's response to oxidative stress, appear to be particularly vulnerable to hypoxic damage. In the course of HIE, a number of mitochondrial functions may be impaired, including inhibition of the respiratory

MicroRNA210 regulated brain pericyte dysfunction exacerbates hypoxic-ischemic brain injury in neonatal mice.

BACKGROUND: While the role of pericytes in blood-brain barrier (BBB) disruption and neuroinflammation is well-established in adult neurological disorders, their contribution to neonatal brain injury is largely unexplored. Here, we investigated the role of brain pericytes in hypoxic-ischemic (HI) brain injury in the developing brain, with a particular focus on the regulatory role of pericyte-derived microRNA210 (miR210) in pericyte dysfunction. METHODS: HI brain injury was induced on postnatal day 9 transgenic mice, including Atp13a5-tdTomato brain pericyte reporter mice, pericyte-specific diph

Melatonin attenuates neurovascular unit dysfunction via AKT/GSK-3β/CREB pathway in hypoxic-ischemic neonatal rats.

OBJECTIVES: Previous research has shown that melatonin (Mel) can alleviate hypoxic-ischemic brain damage (HIBD) in newborns. The roles of Mel on neurovascular unit (NVU) after HIBD and the mechanisms were explored in this study. Methods: The combination of multiparametric magnetic resonance imaging (MRI) techniques and various histology experiments was utilized to observe the impacts of Mel on the NVU in the damaged cerebral cortex of neonatal rats. Especially, water extraction with phase contrast arterial spin tagging (WEPCAST)-MRI was used to monitor the changes in blood-brain barrier (BBB)

Association between hydrogen gas inhalation and cardiac output in an asphyxiated piglet model.

About 80% of neonates with hypoxic-ischaemic encephalopathy (HIE) have cardiovascular dysfunction and are vulnerable to late-onset injury due to disrupted cerebral blood flow and metabolism. We previously reported the neuroprotective effect of hydrogen (H2) gas. This study investigated the effects of H2 gas inhalation on cardiac function in piglets after hypoxia-ischaemia (HI). Seventeen piglets ≤ 24 h old were subjected to HI insult for approximately 40 min and, after 10 min of resuscitation, were divided into an HI group (n = 10) and an HI-H2 gas inhalation group (2.2%–2.7% H2 gas for 6 h; n

PKM2 preconditioning protects endothelial cells from pyroptosis and BBB disruption via NRF2/TRX/TXNIP signaling in neonatal hypoxic-ischemic brain injury.

BACKGROUND: Neonatal hypoxic-ischemic brain damage (HIBD) is a leading cause of neurological deficits and death in neonates. In HIBD, the death of endothelial cells and disruption of the blood-brain barrier (BBB) are closely related to the severity of brain damage and long-term clinical outcomes. There is increasing evidence that a glycolytic enzyme, pyruvate kinase M2 (PKM2), is essential for managing metabolic processes in endothelial cells, but its role (and underlying molecular mechanism) in hypoxic-ischemic (HI)-associated endothelial cell metabolism, cell survival, and BBB function remai

Purα overexpression mitigates neonatal hypoxic-ischemic brain damage by inhibiting neuronal pyroptosis via the P2X7R/NLRP3/Caspase-1 pathway.

Hypoxic-ischemic brain damage (HIBD) is a common neonatal disorder that often leads to permanent neurological sequelae. Neuronal pyroptosis, a pro-inflammatory form of programmed cell death, has been increasingly recognized as a key contributor to neuronal damage and subsequent neurological deficits in HIBD. However, effective therapeutic strategies for HIBD remain limited, highlighting the urgent need to identify novel therapeutic targets. Purine-rich element-binding protein α (Purα) is a multifunctional DNA/RNA-binding protein involved in gene transcription and nervous system regulation. How

Quantitative interpretation using ADC values for subjective MRI classification of neonatal hypoxic-ischemic encephalopathy.

BACKGROUND: MRI-based classification of hypoxic-ischemic encephalopathy (HIE), particularly using diffusion-weighted imaging (DWI), remains susceptible to subjective bias in determining the presence or absence of diffusion restriction. Quantitative measures such as apparent diffusion coefficient (ADC) values may provide a more objective assessment of injury severity. PURPOSE: To evaluate the depth and distribution of brain injury in neonatal HIE by quantitatively assessing regional ADC values across MRI pattern classifications, independent of subjective visual interpretation of DWI signal cha

EVADR lncRNA Mitigates Neonatal Hypoxic-Ischemic Brain Injury via the miR-145→WNT/β-Catenin Axis and Exhibits Biomarker Potential.

Perinatal hypoxic-ischemic injury (HI) upregulated an endogenous retrovirus-derived lncRNA, EVADR, via HIF-1α/NF-κB. EVADR bound and repressed miR-145, thereby activating WNT/β-catenin signaling. Binding specificity was confirmed by biotin-miRNA pull-down and mutant-seed luciferase reporters. Genetic (si-CTNNB1/si-WNT3A) and pharmacologic (XAV939/CHIR99021) cross-validation demonstrated pathway necessity and rescue. EVADR gain-of-function reduced neuronal death and inflammation and improved behavioral outcomes following HI. Circulating EVADR levels in plasma/CSF correlated with injury severity

Activation of the serotonin-1A receptor promotes neural regeneration in the rat model of neonatal hypoxic-ischemic brain damage.

Perinatal hypoxic-ischemic brain damage (HIBD) is a leading cause of lifelong neurodevelopmental disability, and effective therapies remain limited. Activation of the postsynaptic serotonin-1A receptor (5-HT1A-R) has neuroprotective potential against acute and chronic brain injuries. The present study evaluated whether activation of postsynaptic 5-HT1A-R enhances neural regeneration and improves long-term outcomes after neonatal HIBD. Seven-day-old rats were subjected to left carotid artery ligation followed by 2 h of hypoxia (8.0 % O2). NLX-101 (0.16 mg/kg), a selective postsynaptic 5-HT1A-R

Ferroptosis under fire: cannabidiol mitigates iron-dependent injury in differentiated human neuroblastoma cells following oxygen-glucose deprivation.

BACKGROUND: Perinatal hypoxia-ischemia is a major cause of long-term neurological impairments in newborns, with ferroptosis recognized as a key mechanism of injury. Cannabidiol (CBD) is a non-psychoactive phytocannabinoid with antioxidant and neuroprotective properties. CBD is a potential modulator of hypoxic-ischemic brain damage, however its effects on ferroptosis-related pathways remain unclear. PURPOSE: In this study, we examined whether CBD can alleviate ferroptosis-associated damage in differentiated human neuroblastoma (neuron-like SH-SY5Y) cell model of hypoxic-ischemic injury. STUDY

Therapeutic Hypothermia for Neonatal Hypoxic-Ischemic Encephalopathy: Clinical Report.

Therapeutic hypothermia to a temperature of 33.5 to 34.5 °C initiated within 6 hours of birth and continued for 72 hours reduces the risk of death or moderate-to-severe neurodevelopmental impairments in neonates with moderate-to-severe hypoxic-ischemic encephalopathy (HIE) born at ≥36 0/7 weeks of gestation. This specialized therapy requires neuromonitoring, neuroimaging, and plans for follow-up of neurodevelopmental outcomes. Any center or practitioner involved in newborn deliveries should have action plans for prompt recognition and initiation of therapeutic hypothermia or transfer of infant

Human chorionic gonadotropin decreases cerebral cystic encephalomalacia and parvalbumin interneuron degeneration in a pro-inflammatory model of mouse neonatal hypoxia-ischemia.

Human chorionic gonadotropin (hCG) is an immunoregulatory and neurotrophic glycoprotein of potential clinical utility in the neonate at risk for cerebral injury. However, hCG has not been demonstrated to affect the pro-degenerative actions of inflammation in neonatal brain injury. Here we utilize a neonatal mouse model of mild hypoxia-ischemia (HI) combined with intraperitoneal administration of lipopolysaccharide (LPS) to evaluate the neuroprotective actions of hCG in the setting of systemic inflammation. Intraperitoneal treatment with hCG significantly decreased tissue loss and cystic degene
