Nomogram for predicting successful ear molding in infants with congenital auricular deformities: A retrospective cohort study.

BACKGROUND: Nonsurgical ear molding is effective for congenital auricular deformities, but treatment response varies by patient- and ear-specific characteristics. This study developed and internally validated an ear-level prediction model for successful correction. METHODS: This single-center retrospective cohort included infants undergoing nonsurgical ear molding between September 2022 and December 2025. The individual ear was the analytical unit. Successful correction was defined as an excellent or good outcome based on blinded photographic assessment. Six prespecified pretreatment predicto

Generation of two CASK patient-derived human induced pluripotent stem cell lines to study CASK-related disorders.

In this study, we report generation of human induced pluripotent stem cells (hiPSCs) from two calcium/calmodulin dependent serine protein kinase (CASK)-related disorder patients with intronic mutations in the CASK gene. Reprogramming of hiPSCs was done using peripheral blood mononuclear cells (PBMCs) as a cell source and Sendai virus for delivery of reprogramming factors. Generated hiPSC lines are a valuable tool to study neurodevelopmental deficits associated with CASK mutant variants.

Reproducible Bradycardia After Flumazenil Reversal of Midazolam-Propofol Intravenous Sedation in a Patient With Klinefelter Syndrome: A Case Report.

Klinefelter syndrome (KS) is the most common sex chromosome aneuploidy in males and is associated with metabolic, cardiovascular, and neurodevelopmental features. Some individuals with KS have also been reported to exhibit altered heart rate (HR) regulation, although cardiovascular responses associated with intravenous sedation (IV-S) have not been well documented. We report a 14-year-old boy with KS, intellectual disability, and autism spectrum disorder who underwent multiple dental treatments under IV-S with midazolam and propofol. During three consecutive sessions at approximately two-week

Intra-Abdominal Bowel Dilation in Experimental Gastroschisis is Associated with a Modifiable Transcriptomic Program of Intestinal Dysfunction.

OBJECTIVE: To characterize intestinal transcriptional profiles in gastroschisis, their temporal evolution, and response to fetal intervention. SUMMARY BACKGROUND DATA: Gastroschisis causes significant intestinal dysfunction, with intra-abdominal bowel dilation clinically shown to correlate with worse outcomes. While inflammation and neurovascular impairment have been implicated, genome-wide transcriptional characterization of disease severity remains lacking. METHODS: Using a fetal ovine model of complex gastroschisis, in which all gastroschisis animals demonstrated significant intra-abdomin

Postnatal weight trajectories drives beter head circumference growth in preterm infants ≤32 weeks: a multicenter Latin American cohort study.

BACKGROUND: Postnatal weight loss is nearly universal in preterm infants, and its relationship to head circumference (HC) growth remains incompletely characterized. Growth standards were recently updated with the Fenton 2025 charts, which modify Z-score parameters compared to the 2013 version. OBJECTIVE: To evaluate the association between postnatal weight trajectory and HC growth from birth to discharge in preterm infants ≤32 weeks of gestation across a large multicenter Latin American cohort, using Fenton 2025 growth standards. METHODS: This observational cohort study by secondary analysis

Kidney Organoids: Disease Modeling and Clinical Implementations for Patient Benefit.

BACKGROUND: Progress in nephrology is still constrained by the limited ability of conventional experimental models to faithfully recapitulate the complex structure and function of the human kidney. Human pluripotent stem cell-derived kidney organoids provide a three-dimensional in vitro experimental system that allows investigation of kidney development and disease-related mechanisms in a human cellular context, although still limited in their ability to reproduce the full structural and physiological complexity of the native kidney. SUMMARY: Kidney organoids have emerged as a powerful system

Tibialis Anterior Tendon Transfer in an Arthrogrypotic Clubfoot: A Case Report.

CASE: A 30-month-old boy with arthrogryposis multiplex congenita presented with severe, recurrent teratological clubfoot refractory to serial casting, Achilles tenotomy, and plantar fascia release. Because of repeated cast slippage and patient distress, comprehensive posterior, plantar, and medial releases were performed, with cast exchange 2 weeks thereafter. A robust, taut tibialis anterior prompted addition of tibialis anterior tendon transfer to the lateral cuneiform. Postoperatively, plantigrade alignment was achieved both clinically and radiographically and maintained at the 2-year follo

Deep learning for multimodal physiological signal based assessment of sleep disordered breathing.

Sleep disordered breathing (SDB) is commonly assessed using polysomnography (PSG), which records multiple physiological signals during sleep. Accurate automated assessment of SDB related breathing abnormalities remains challenging because respiratory events occur within complex and stage dependent sleep dynamics. To address this problem, this study proposes a multimodal physiological signal based deep learning framework for SDB oriented sleep assessment. The proposed framework treats SDB related respiratory event detection as the clinically targeted task and incorporates sleep stage classifica

Sono-embryonic detection of monochorionic, diamniotic triplets with conjoined twins after in vitro fertilization: A case report and review of the literature.

Monochorionic diamniotic triplets with a conjoined pair are exceedingly rare and arise from multiple splitting events of a single zygote. This report presents the earliest documented ultrasound diagnosis of MCDA triplets with a conjoined pair, at 8 weeks and 5 days of gestation, in a patient who underwent in-vitro fertilization with a single embryo transfer. High-resolution transvaginal ultrasound revealed a monochorionic diamniotic configuration with two amniotic sacs: one containing a structurally normal embryo and the other containing di-thoracic parapagus conjoined twins. This case highlig

Genotype and Ultrastructure Correlation in Primary Ciliary Dyskinesia Among Saudi Children: A Case Series.

Background: Primary ciliary dyskinesia (PCD) is a rare inherited disorder of motile cilia causing chronic oto-sinopulmonary disease. Transmission electron microscopy (TEM) remains essential for confirming ultrastructural defects, especially in resource-limited settings. This study characterizes the clinicopathologic and molecular spectrum of Saudi children with PCD. Methods: Ten Saudi children with confirmed PCD (2018-2025) were retrospectively reviewed. Clinical, radiological, ultrastructural, and whole exome sequencing (WES) data were analyzed to assess genotype-phenotype correlation. Result

Endolymphatic sac tumors in a tertiary referral center: long-term outcomes and management of primary and salvage cases.

Endolymphatic sac tumors are rare neoplasms arising from the posterior aspect of the petrous temporal bone and are characterized by locally aggressive behavior with significant bone destruction. Although they may occur sporadically, a well-established association with von Hippel-Lindau (VHL) disease has been reported. This study aimed to characterize the clinical features, management, and long-term outcomes of a consecutive series of seven patients treated at a tertiary referral center, including cases associated with VHL. A retrospective analysis of institutional cases was performed and compl

HIF-Regulated Pannexin-1 Channel Drives Luminal ATP Accumulation in Kidney Cysts.

Autosomal dominant polycystic kidney disease (ADPKD) is marked by the progressive development of bilateral kidney cysts, leading to compression of intact surrounding tissue and a subsequent decline in kidney function. Accumulation of ATP within the cyst fluid significantly drives cyst expansion by activating purinergic receptors, thereby promoting calcium-dependent chloride secretion. Moreover, the induction of hypoxia-inducible factor 1α (HIF-1α) in cyst-lining epithelial cells further enhances chloride conductance. However, the mechanisms underlying ATP release into the cyst fluid and the ro
