Microcephaly with Simplified Gyral Pattern in Children: Quantitative Morphometric Assessment on Brain MRI and Correlation with Clinical Outcome.

BACKGROUND AND PURPOSE: Microcephaly with simplified gyral pattern (SGP) represents an often subtle subtype of malformation of cortical development that is commonly accompanied by epilepsy and neurodevelopmental delay. However, whether gyral simplification scales to reduced brain volume and whether MRI features predict clinical outcome still remain unclear. The aims of this study are therefore to apply a quantitative morphometric approach to distinguish a simplified gyral pattern from microcephaly-related effects and to correlate quantitative MRI-derived features with clinical outcome. Fatty acid transporter protein isoform 2 (FATP2) inhibitor lipofermata inhibits Zika virus replication and blocks fatty acid uptake in SH-SY5Y neuroblastoma cell line.

Zika virus (ZIKV) infection in adults is typically mild and self-limiting, but infection during pregnancy can cause fetal and neonatal microcephaly. No approved drugs or vaccines currently exist. ZIKV hijacks host lipid homeostasis to facilitate replication, and inhibitors of fatty acid synthesis decrease its production. However, cells obtain lipids from both de novo synthesis and uptake. Neurons, a primary target of ZIKV pathogenesis, have limited capacity for de novo lipid synthesis and rely more on lipid uptake. We previously demonstrated that lipid uptake inhibitors block influenza virus r

The people behind the papers - Alexander Phillips and David Keays.

Certain mutations in end-binding (EB) protein 2, a member of the EB family of conserved microtubule plus-end tracking proteins, are implicated in a rare syndromic form of microcephaly. While the two other EB family members - EB1 and EB3 - have been the subject of intense study, the cellular role of EB2 is not clearly understood. In their work, Alexander (Alex) Phillips, David Keays and colleagues show that EB2 is necessary for maintaining mitotic fidelity during embryonic brain development in mouse and human. To learn more about their work, we spoke to the first author, Alex Phillips, and the

Proteostasis failure and mitochondrial dysfunction contribute to chromosomal instability-induced microcephaly.

Mosaic variegated aneuploidy (MVA), a rare human congenital disorder that causes microcephaly, is characterized by extensive abnormalities in chromosome number and results from mutations in genes involved in accurate mitotic chromosome segregation. To characterize the cellular mechanisms underlying this disease, here we generated a Drosophila model of microcephaly caused by the depletion of a single spindle assembly checkpoint (SAC) gene in the neural stem cell (NSC) compartment. We present evidence that loss of stemness - compromised identity and proliferative capacity of NSCs- plays an impor

Ultra-rare biallelic THAP12 variants cause loss of function and underlie severe epileptic encephalopathy.

Developmental and epileptic encephalopathies (DEEs) are a group of severe childhood-onset neurological disorders, often caused by rare genetic variants affecting brain development and excitability. Despite advances in genomic sequencing, a substantial proportion of DEE cases remain unsolved. Here, we identify THAP12 as a novel disease-causing gene associated with autosomal recessive DEE. Whole-genome sequencing in two siblings who presented with infantile spasms and progressed to Lennox-Gastaut syndrome revealed compound heterozygous variants in THAP12 , leading to a reduction in protein abund

Biallelic XPR1 Variants Are Linked to Brain Calcifications, Developmental Delay, Hypophosphatemia, and Cardiopulmonary Phenotype.

XPR1 encodes the only known phosphate exporter in human cells and regulates the export of inorganic phosphate (Pi). To date, heterozygous pathogenic variants have been reported to be associated with autosomal-dominant idiopathic basal ganglia calcification-6 (IBGC6; MIM: 616413). We collate clinical information on individuals with biallelic XPR1 variants and use in silico and cell-based studies to evaluate pathogenicity. Four consanguineous families have been documented, with a severe neonatal phenotype presenting with persistent pulmonary hypertension, chronic lung disease, cardiomyopathy, an

Intellectual disability-causing mutations in KIF11 impair microtubule dynamics and dendritic arborization.

Microcephaly with or without chorioretinopathy, lymphedema, or intellectual disabilities (MCLID) is a rare disease caused by mutations in the mitotic motor KIF11. However, the specific neuronal functions of KIF11, its mechanisms of microtubule (MT) regulation, and the impact of MCLID mutations on KIF11 function remain underexplored. Here, using live-imaging, we find that KIF11 depletion in postmitotic neurons increases minus-end-out MT dynamics in both axons and dendrites. Introducing MCLID-associated KIF11 mutations, KIF11Y82F and KIF11ΔCterm, significantly reduces MT dynamics, impairs dendri

CDK4 and CDK6 variants in patients with primary microcephaly lead to cell cycle defects and mitochondria-induced apoptosis.

BACKGROUND: The disruption of neural progenitor proliferation is a key mechanism underlying primary microcephaly, yet how cell cycle arrest leads to progenitor loss remains only partially understood. Cyclin-dependent kinases, CDK4 and CDK6, are central regulators of the G₁/S transition, but their role in cellular stress responses during neurodevelopment remains unclear. METHODS: We studied fibroblasts from affected individuals in two families: siblings homozygous for a CDK4 frameshift (p.Glu94Argfs*65) presenting with microcephaly and pontine hypoplasiaand a child homozygous for a CDK6 missen

Functional rescue of a disease-linked ERAD pathway mutation via alternative splicing.

ER-associated degradation (ERAD) targets misfolded proteins in the endoplasmic reticulum (ER) for proteasomal degradation. Mutations in its most conserved branch involving the SEL1L-HRD1 complex cause ERAD-associated neurodevelopmental disorders with onset in infancy (ENDI), characterized by developmental delay, microcephaly, and locomotor dysfunction. Its most severe form, ENDI with agammaglobulinemia (ENDI-A), results from a bi-allelic SEL1L-Cys141Tyr (C141Y) mutation within its fibronectin II (FNII) domain and currently lacks effective treatment. Here, we find that knock-in mouse models car

Novel PCDH12 pathogenic missense variants cause neurodevelopmental disorders with ocular malformation.

Protocadherin-12 (PCDH12), a cell-adhesion protein belonging to the non-clustered protocadherin family, plays a crucial role in the establishment and regulation of neuronal connections and communication. Bi-allelic loss-of-function (LoF) variants in the PCDH12 gene have been associated with several neurodevelopmental disorders (NDDs) such as diencephalic-mesencephalic junction dysplasia syndrome, cerebral palsy, and cerebellar ataxia, often accompanied by ocular abnormalities. However, genotypes exhibit variable expressivity. Affected individuals sharing the same PCDH12 variant presenting diff

CEP170 as a novel molecular link between centrosomal function and cerebral cortical development.

BACKGROUND: The centrosome is a critical regulator of cortical development, orchestrating microtubule dynamics, cell cycle progression, and neuronal migration. Disruptions in centrosome-associated proteins have been associated with a range of neurodevelopmental disorders. CEP170, a microtubule-binding protein localized to the subdistal appendages (SDA) of centrioles, has been implicated in centrosome function, yet its role in corticogenesis remains poorly defined. METHODS: We analyzed CEP170 expression in mouse cortex using western blotting, qPCR, scRNA-seq, and spatial transcriptomics, and e

Architects of the Developing Brain: Cytoskeleton-Organizing Molecules in Neurodevelopmental Disorders.

Different components of the cytoskeleton are very important determinants of brain development. They orchestrate multiple cellular processes involved in all phases of cerebral cortex development. In this review, we summarize current knowledge on the components of the cytoskeleton-microtubules, actin filaments, and intermediate filaments-and their roles in cortical development. We provide a detailed analysis of how cytoskeleton molecules control neuronal progenitor proliferation, neuronal migration, polarization, axon and dendrite specification and outgrowth, and synaptogenesis. We further exami

The global outbreak of Oropouche virus: surveillance of global trends.

Oropouche virus (OROV) is a re-emerging arthropod-borne virus of the genus Orthobunyavirus (family Peribunyaviridae) that poses an increasing public health concern in South and Central America and the Caribbean. First identified in Trinidad in 1955, OROV has caused recurrent outbreaks, with recent resurgence associated with geographic expansion and increased urban transmission. In 2024 alone, more than 11,600 confirmed cases were reported in the Region of the Americas, with continued transmission documented in early 2025. The virus is primarily transmitted in urban settings by the biting midge
