Identification and structural characterisation of a novel mutation in the CNKSR2 gene associated with Houge-Type X-Linked Intellectual Developmental Disorder.

Neurological disorders with overlapping phenotypes pose significant diagnostic challenges, especially the ones that occur on account of rare genetic variants. We hereby report a case of a 16-year-old male with clinical symptoms of severe intellectual disability, excessive appetite, an elongated face with decreased body weight (<-2 SD) and microcephaly (<-3 SD). Behavioral manifestations like attention deficit, hyperactivity, and impulsivity were also observed in the patient. The Exome sequencing of affected child revealed a novel frameshift insertion in the CNKSR2 gene, or connector enhancer o

DIAPH1 regulates the Wnt/β-catenin pathway resulting in microcephaly and visual impairment.

BACKGROUND: Biallelic DIAPH1 mutations are linked to hereditary microcephaly syndrome, yet the underlying pathogenic mechanism remains unelucidated. This study aimed to clarify how DIAPH1 biallelic mutations cause microcephaly and visual impairment, focusing on the gene’s regulatory role in the Wnt/β-catenin signaling pathway. METHODS: Whole exome sequencing was performed on a patient’s peripheral blood to identify DIAPH1 mutations. A zebrafish model was established by microinjecting mutant human DIAPH1 cDNA into one-cell embryos (no zebrafish DIAPH1 homolog exists). Phenotypic analyses (morph

Beyond the genome: clinical challenges in diagnosing LONP1-related mitochondrial disorders.

BACKGROUND: LONP1 encodes an ATP-dependent protease essential for maintaining mitochondrial homeostasis. LONP1 variants have been associated with cerebral-ocular-dental-auricular-skeletal anomalies syndrome, pediatric cataract, congenital diaphragmatic hernia, and neurodevelopmental disorders; moreover, these variants can be inherited in both autosomal recessive and autosomal dominant modes. METHODS: We conducted a retrospective analysis of the clinical data and genetic test results of a Chinese boy diagnosed as having mitochondrial encephalopathy. Subsequently, we evaluated the pathogenicity

Expanding the Phenotypic Spectrum of Anauxetic Dysplasia Type 3: Reporting an Iranian Family With Unique Systemic Features and NEPRO Gene Variant.

The cartilage hair hypoplasia-anauxetic dysplasia (CHH-AD) spectrum represents a group of rare autosomal recessive skeletal dysplasias with significant phenotypic heterogeneity. These disorders are classified based on pathogenic variants in the RMRP, POP1, and NEPRO genes. Among these, anauxetic dysplasia type 3 (ANXD3), associated with NEPRO variants, manifests as severe skeletal dysplasia characterized by short stature, brachydactyly, skin laxity, and joint hypermobility, with distinct radiographic findings such as ovoid vertebrae, hypoplastic ilia, narrow acetabular angles, and irregular me

Deficiency of pnkp in zebrafish causes microcephaly, seizures, and developmental delay through mitochondrial dysfunction.

Variants in PNKP cause a severe neurodevelopmental disorder characterized by microcephaly, seizures, and developmental delay (MCSZ). Despite clear clinical significance, the pathological mechanisms underlying this condition remain incompletely understood. In this study, CRISPR/Cas9 genome editing was applied to generate a zebrafish pnkp knockout model ( pnkp -/- ), overcoming limitations associated with previously reported mouse models that exhibited postnatal lethality. The pnkp -/- zebrafish faithfully recapitulated key phenotypic traits observed in human PNKP deficiency, facilitating in-dep

Worm orthologues of cytokinesis-associated proteins CIT and ASPM regulate neuronal microtubule dynamics and polarity in C. elegans.

The polarized architecture of neurons is intricately associated with the modulation of microtubule dynamics. Over the years, several microtubule-associated factors that regulate neuronal polarity have been identified. However, the precise details of how microtubule arrangement and stability are established in axons and dendrites are not clearly understood. To uncover the relevant factors involved in the biological pathways governing microtubule regulation in neurons, we conducted a suppressor screen using the neuronal ectopic extension phenotype caused by the loss of the kinesin-13 family micr

ASPM promotes the progression of esophageal squamous cell carcinoma by upregulating the expression of HMMR.

OBJECTIVE: The ASPM gene, which is related to abnormal spindle-like microcephaly, is highly expressed in various human cancers. However, the expression of ASPM in human esophageal squamous cell carcinoma (ESCC) and its key role remain unclear. Therefore, this study conducted animal model experiments, cell experiments, and clinical experiments to evaluate the functional connection between ASPM and ESCC as well as the clinical significance of ASPM. METHODS: (1) A mouse esophageal cancer model was constructed using the 4NQO4-Nitroquinoline-1-oxide environmental mutagen. The mouse esophageal tiss

Early GH therapy and neurodevelopmental outcome in a child with compound heterozygous IGF1R variants.

Causal variants in the IGF 1 receptor (IGF1R) gene are associated with variable degrees of growth and neurodevelopmental impairment. While heterozygous variants often manifest as less severe phenotypes, homozygous loss-of-function mutations are widely regarded as incompatible with life. Biallelic hypomorphic variants are exceptionally rare and their clinical spectrum remains poorly defined. We report the case of a girl born at term after severe symmetric fetal growth restriction (FGR), identified prenatally through trio whole-exome sequencing as carrying compound heterozygous IGF1R variants-c.

The microcephaly protein Abnormal Spindle has an essential role in symmetrically dividing neural precursors to promote brain growth and development.

Neurogenesis must be coordinated in time and space to ensure proper neural development. Defects in this process can lead to a variety of brain malformations that affect tissue size and architecture. Here we examine the spatiotemporal requirements of the Abnormal Spindle (Asp) protein and its human ortholog Abnormal Spindle-Like, Microcephaly Associated (ASPM) in a fly model of human microcephaly (MCPH). By utilizing the fly optic lobe, whose neurogenic program parallels that of mammals, and the genetic tools available in this organism, we uncover the progenitor cell types and critical neurogen

Genetic variants in Rps4x cause intellectual disability with dysmorphic features, microcephaly, and autism.

X-linked intellectual disability (XLID) comprises a group of heterogeneous disorders associated with impaired cognitive function and developmental delays. It has been estimated that 10% of the genes on the X-chromosome are associated with at least one form of intellectual disability. To date, genetic variants in 172 genes have been associated with XLID. Clinically, these disorders are highly variable, with some exhibiting multi-system involvement and others limited only to intellectual impairment. Here we describe a new XLID condition caused by defects in RPS4X, which encodes a component of th

Rapgef1 paralog-mediated regulation of Wnt/β-catenin signaling orchestrates early embryo tissue patterning and morphogenesis.

RAPGEF1, a guanine nucleotide exchange factor, regulates signaling and cytoskeletal dynamics in mammalian cells, yet its role in development remains unclear as Rapgef1 null mouse embryos do not survive beyond implantation. We demonstrate that zebrafish rapgef1 is maternally expressed, and its paralogs, rapgef1a and rapgef1b, exhibit tissue and developmental stage-specific splicing. Disruption of rapgef1b caused brain and somite defects, impaired cranial neural crest specification, and microcephaly-like phenotypes, uncovering its previously uncharacterized functions in morphogenesis and tissue
