Multidisciplinary management of agenesis of corpus callosum: a narrative review.

The corpus callosum (CC) is the most significant of the three cerebral commissures that join the human left and right hemispheres. Numerous vascular, chemical, genetic, and metabolic disturbances can impede the growth of the CC. Complete agenesis of the CC is the most prevalent abnormality. As a result of improvements in our knowledge and advancements in ultrasonography technology, an increasing number of congenital CC abnormalities have been identified. Prenatal magnetic resonance imaging is crucial to identify related central nervous system abnormalities. Counseling about the postnatal resul

Odor localization in structural interhemispheric deficits.

Contrary to all other sensory systems, olfactory information is processed predominantly ipsilaterally. Furthermore, odor localization, based on inter-nostril differences, is usually not possible under controlled conditions. These two observations suggest information exchange between both cerebral hemispheres in the olfactory system, although the exact anatomical substrate remains unknown. This study aimed to identify the anatomical structures necessary for odor localization, with a particular focus on the role of interhemispheric communication. We assessed the ability to localize pure olfactor

Sex- and age-dependent neurovascular abnormalities linked to neuroinflammation lead to exacerbated post-ischemic brain injury in Marfan syndrome mice.

Fibrillin 1 gene (Fbn1) mutations cause Marfan syndrome (MFS), triggering life-threatening aortic complications and multi-organ effects. MFS is increasingly linked to neurovascular complications, amplified by aortic surgery risks. However, the impact of MFS on the brain remains unclear, including the roles of sex, aging, and their contribution to cerebral injury. This study examines brain alterations and their role in cerebral ischemic injury in an MFS mouse model. RNA-seq analysis of young (3-month-old) and aged (13-month-old) male and female wild-type and MFS (Fbn1C1041G/+) mice revealed dis

Thick Corpus Callosum: An Unusual Finding of TUBGCP2-Related Tubulinopathy.

Tubulinopathies are a group of neurologic disorders caused by mutations in tubulin-related genes, characterized by a broad spectrum of cortical malformations. These disorders are typically associated with a thin or normal corpus callosum, while a thick corpus callosum has been rarely documented. This study reports the first known case of a homozygous insertion variant in the TUBGCP2 gene associated with a thick corpus callosum. A 2-year-old male of Turkish origin presenting with microcephaly, developmental delay, distinctive facial features, and hypertonia. Brain MRI revealed cortical malforma

White matter trajectories in Down syndrome and Alzheimer's disease: Insights from diffusion tensor-based morphometry.

INTRODUCTION: Diffusion magnetic resonance imaging studies investigating Down syndrome (DS) and Alzheimer's disease (AD) have mainly relied on white matter (WM) skeleton-based techniques, potentially overlooking broader WM architecture. METHOD: We applied diffusion tensor-based morphometry (D-TBM), a novel whole-volume WM registration technique, to characterize WM properties in DS. Between- and within-group analyses were conducted in 51 adults with DS and 35 controls, divided into two age groups, using diffusion tensor imaging (DTI)-derived metrics and local volumetric changes. RESULTS: DS p

Agenesis of Corpus Callosum: A Clinical Study of Complete Versus Partial Agenesis in a 20-Year Retrospective Cohort.

Agenesis of the corpus callosum (ACC) has a wide spectrum of anatomic manifestations associated with incompletely understood clinical variability. In this retrospective cohort study, 161 children with ACC were classified into complete ACC and partial ACC. Partial and complete ACC diagnoses were confirmed by postnatal magnetic resonance imaging (MRI) and further classified as isolated or complex (associated with other brain abnormalities or genetic diagnoses). Clinical characteristics, clinical outcomes, and co-occurring genetic variations were collected by chart review. Median age at last neur

Complete commissural agenesis in a child with Noonan-like syndrome with loose anagen hair 2.

RASopathies are a wide group of multisystemic disorders caused by pathogenic variants in genes belonging to the RAS/MAPK pathway. Among these, PPP1CB gene variants cause Noonan syndrome-like disorder with loose anagen hair 2 (NSLAH2), a rare condition with neuro-cardio-facio-skeletal involvement and the peculiar loose anagen hair. We report on a girl carrying the recurrent c.146 C > G (p.Pro49Arg) pathogenic variant, who presented the classical NSLH features associated with a previously unreported complete commissural agenesis, likely expanding the phenotype. The prominent role of Ras protein

Prenatally and postnatally sequential genetic etiology detection in corpus callosum abnormalities.

To evaluate the clinical utility of genetic testing via karyotyping, chromosomal microarray analysis (CMA), and exome sequencing in cases with corpus callosum abnormalities (CCA). Here, 65 prenatal and 12 postnatal cases diagnosed with CCA via ultrasound and magnetic resonance imaging examination were enrolled. All cases were divided into two groups: 21 (27.3%) isolated and 56 (72.7%) non-isolated CCA groups. Karyotyping and CMA were first performed in parallel, then followed by whole exome sequencing (WES) after negative karyotype and CMA results. Clinical outcomes were also followed up. Kary

Extremely rare case report of CEBALID syndrome presenting as congenital arthrogryposis.

BACKGROUND: CEBALID syndrome (Craniofacial defects, dysmorphic Ears, Brain Abnormalities, Language delay, and Intellectual Disability) is caused by C-terminal truncated transcriptional activator MN1. This novel syndrome characterized by neurodevelopmental delay, craniofacial anomalies, and partial rhombencephalosynapsis was recently identified in 2020. Since then, less than 30 cases have been reported worldwide. None of the reported cases described an association of arthrogryposis with CEBALID syndrome. Below we present the first reported case in Lebanon with an associated arthrogryposis at bi

Structural mediation of the default-mode network in children with callosal agenesis.

Agenesis of the corpus callosum is a neurodevelopmental condition characterized by the partial or complete absence of the corpus callosum, the largest white matter bundle connecting the cerebral hemispheres. The default-mode network comprises bilateral frontal, temporal, and parietal regions that exhibit correlated activity at rest. Previous studies show that individuals with agenesis of the corpus callosum show overall preserved default-mode network functional connectivity, suggesting compensatory mechanisms for maintaining bilaterally correlated activity. In this study, we aimed to explore w

Derivative Complex Small Supernumerary Marker Chromosomes (sSMC) Involving Chromosomes 2 and 15-A Novel Report.

Small supernumerary marker chromosomes (sSMC) constitute a rare group of structural chromosomal abnormalities characterized by additional genetic material that cannot be identified by conventional banding cytogenetics. The incidence of sSMC is extremely rare, and most are expected to have no clinical phenotypic abnormalities. Advanced cytogenetic modalities are crucial for sSMCs identification, characterization, and analysis of chromosomal structure. An 8-day-old neonate born to a G2P2 mother with gestational diabetes and a history of infertility was admitted for respiratory distress. Clinical

Longitudinal diffusion tensor imaging correlates with amyloid burden in Down syndrome.

INTRODUCTION: Adults with Down syndrome (DS) accumulate amyloid beta (Aβ) plaques faster and earlier on average than neurotypical adults with sporadic Alzheimer's disease (AD). White matter (WM) microstructure characterized with diffusion tensor imaging (DTI) can indicate underlying architectural changes in longitudinal studies, suggestive of neurodegeneration. This study investigated relationships between DTI and Aβ in DS along the AD continuum. METHODS: Using longitudinal amyloid Pittsburgh compound B positron emission tomography, Centiloid (CL) and DTI parameters were examined in 35 adults
