Extremely rare case report of CEBALID syndrome presenting as congenital arthrogryposis.

BACKGROUND: CEBALID syndrome (Craniofacial defects, dysmorphic Ears, Brain Abnormalities, Language delay, and Intellectual Disability) is caused by C-terminal truncated transcriptional activator MN1. This novel syndrome characterized by neurodevelopmental delay, craniofacial anomalies, and partial rhombencephalosynapsis was recently identified in 2020. Since then, less than 30 cases have been reported worldwide. None of the reported cases described an association of arthrogryposis with CEBALID syndrome. Below we present the first reported case in Lebanon with an associated arthrogryposis at bi

Structural mediation of the default-mode network in children with callosal agenesis.

Agenesis of the corpus callosum is a neurodevelopmental condition characterized by the partial or complete absence of the corpus callosum, the largest white matter bundle connecting the cerebral hemispheres. The default-mode network comprises bilateral frontal, temporal, and parietal regions that exhibit correlated activity at rest. Previous studies show that individuals with agenesis of the corpus callosum show overall preserved default-mode network functional connectivity, suggesting compensatory mechanisms for maintaining bilaterally correlated activity. In this study, we aimed to explore w

Derivative Complex Small Supernumerary Marker Chromosomes (sSMC) Involving Chromosomes 2 and 15-A Novel Report.

Small supernumerary marker chromosomes (sSMC) constitute a rare group of structural chromosomal abnormalities characterized by additional genetic material that cannot be identified by conventional banding cytogenetics. The incidence of sSMC is extremely rare, and most are expected to have no clinical phenotypic abnormalities. Advanced cytogenetic modalities are crucial for sSMCs identification, characterization, and analysis of chromosomal structure. An 8-day-old neonate born to a G2P2 mother with gestational diabetes and a history of infertility was admitted for respiratory distress. Clinical

Longitudinal diffusion tensor imaging correlates with amyloid burden in Down syndrome.

INTRODUCTION: Adults with Down syndrome (DS) accumulate amyloid beta (Aβ) plaques faster and earlier on average than neurotypical adults with sporadic Alzheimer's disease (AD). White matter (WM) microstructure characterized with diffusion tensor imaging (DTI) can indicate underlying architectural changes in longitudinal studies, suggestive of neurodegeneration. This study investigated relationships between DTI and Aβ in DS along the AD continuum. METHODS: Using longitudinal amyloid Pittsburgh compound B positron emission tomography, Centiloid (CL) and DTI parameters were examined in 35 adults

Pathogenic variants in TMEM184B cause a neurodevelopmental syndrome associated with alteration of metabolic signaling.

Transmembrane protein 184B (TMEM184B) is an endosomal 7-pass transmembrane protein with evolutionarily conserved roles in synaptic structure and axon degeneration. We report six pediatric cases who have de novo heterozygous variants in TMEM184B; five individuals harbor a rare missense variant, and one individual has an mRNA splice site change. This cohort is unified by overlapping neurodevelopmental deficits including developmental delay, corpus callosum hypoplasia, seizures, and/or microcephaly. TMEM184B is predicted to contain a pore domain wherein four of five human disease-associated misse

White matter microstructure in school-age children with down syndrome.

Down syndrome (DS) is the most common genetic cause of intellectual disability, but our understanding of white matter microstructure in children with DS remains limited. Previous studies have reported reductions in white matter integrity, but nearly all studies to date have been conducted in adults or relied solely on diffusion tensor imaging (DTI), which lacks the ability to disentangle underlying properties of white matter organization. This study examined white matter microstructural differences in 7- to 12-year-old children with DS (n = 23), autism (n = 27), and typical development (n = 50

A large cohort study of prenatal exome sequencing redefines diagnosis in fetal corpus callosum anomalies.

Anomalies of the corpus callosum (AnCC) are congenital malformations associated with highly variable neurodevelopmental outcomes. We performed prenatal exome sequencing (pES) on a cohort of 352 fetuses diagnosed with AnCC, analysing the diagnostic yield, the implicated genes based on the type of anomaly (partial or complete agenesis, short corpus callosum, or callosal dysgenesis) and assessing the impact on pregnancy outcomes. The overall diagnostic yield of pES was 23%, with pathogenic or likely pathogenic variants identified in 49 different genes, most of which linked to intellectual develop

Early Magnetic Resonance Imaging Predicts 12-Month Outcome in Neonates with Congenital Diaphragmatic Hernia.

INTRODUCTION: Advances in neonatology, neonatal surgery, and extracorporeal membrane oxygenation have improved the prognosis of congenital diaphragmatic hernia (CDH). However, CDH survivors are at considerable risk of long-term neurological morbidity. Magnetic resonance imaging (MRI) abnormalities are reported in up to 84% of CDH survivors but have only been rarely compared with neurodevelopmental outcomes. This study aims to investigate whether assessment of postnatal MRI in CDH survivors allowed association with and prediction of long-term outcome. METHODS: Brain MRI was performed in 36 neo

Comprehensive genotypic, phenotypic, and biochemical characterization of GOT2 deficiency: A progressive neurodevelopmental disorder with epilepsy and abnormal movements.

PURPOSE: Glutamic-oxaloacetic transaminase (GOT), also known as aspartate aminotransferase, catalyzes the reversible transamination of oxaloacetate and glutamate to aspartate and α-ketoglutarate. Two isoforms, cytosolic (GOT1) and mitochondrial (GOT2), are integral to the malate-aspartate shuttle, a key regulator of intracellular redox homeostasis. Recently, 5 patients with biallelic variants in GOT2 were described, presenting with developmental and epileptic encephalopathy. METHODS: We report 11 additional patients with homozygous GOT2 variants, along with additional data from 4 previously r

Insights into ALG3-CDG: A case study combining glycan profiling and genetic analysis.

Congenital disorders of glycosylation (CDG) are a group of rare metabolic disorders caused by the defects in the glycosylation pathways of biomacromolecules leading to altered glycoprofiles in affected individuals. In this case study, we present a 3-year-old Slovak male patient with developmental delay, hearing impairment, epilepsy, microcephaly, facial dysmorphism, corpus callosum dysgenesis, and cardiac abnormalities. To elucidate the underlying cause, we performed LC-ESI-MS analysis of RapiFluor-labelled N-glycans released from blood serum glycoproteins. The results revealed an abnormal N-g

Expanding genetic and clinical spectra of β-tubulinopathies: A Korean study.

Tubulin proteins form microtubules, which are critical for neuronal cell migration. In humans, there are at least 27 tubulin genes. Pathogenic variants in these genes cause tubulinopathies, which are characterized by diverse neurodevelopmental symptoms and brain malformations. This study analyzed the genetic variants and clinical characteristics of 12 patients (3 males, 9 females) with confirmed β-tubulinopathies. A retrospective chart review indicated that diagnoses were made via exome, genome, or targeted next-generation panel sequencing. Most patients (11/12) showed significant developmenta

Cardiac Involvement and TBCK -Related Neurodevelopmental Disorder: Is It a New Feature of This Condition?

TBCK (TBC1 Domain-Containing Kinase) encodes a protein playing a role in actin organization and cell growth/proliferation via the mTOR signaling pathway. Deleterious biallelic TBCK variants cause Hypotonia, infantile, with psychomotor retardation and characteristic facies 3. We report on three affected sibs, also displaying cardiac malformations. The parents, a consanguineous couple of first cousins, were referred to schedule invasive diagnosis for their sixth pregnancy. They were known to carry the pathogenic c.1532G>A TBCK variant. The variant was originally identified in homozygosity in the

A novel DCC truncating mutation leads to rare congenital mirror movements and corpus callosum agenesis: A case report.

ABSTRACTRATIONALE: Mirror movements (MRMVs) are quite common in young children and typically diminish before the age of 10. However, congenital MRMVs often persist into adulthood. MRMV1 is a rare neurodevelopmental disorder characterized by involuntary movements mirroring intentional actions on the opposite side of the body. Recent research has confirmed an intrinsic connection between congenital agenesis of the corpus callosum (ACC) and MRMV1, which is associated with mutations in the deleted in colorectal carcinoma (DCC) gene. PATIENT CONCERNS: The proband was a fetus, which was prenatally

Corpus callosum agenesis: does sleep matter?

Agenesis of the corpus callosum is a brain malformation that can lead to intellectual, behavioural, social and emotional impairments, with outcomes varying based on severity. Sleep disturbances are common in children with this disorder, and some studies have evidenced the role of corpus callosum in interhemispheric synchronisation that may influence sleep patterns. This narrative review attempted to intercept available information on sleep in patients with isolated Agenesis of the corpus callosum, partial and complete, across different ages. Since literature about this issue is underdeveloped,

Transcription factor 4 regulates the interhemispheric midline remodeling through neuron-astroglia communications during corpus callosum formation.

Agenesis of the corpus callosum (CC), the largest axon tract bridging the two hemispheres of the brain to support higher cognitive functions, is linked to various neurological disorders, including intellectual disability and autism spectrum disorders. Mutations of the transcription factor 4 (TCF4) cause Pitt-Hopkins Syndrome (PTHS), an autism spectrum brain disorder characterized by severe intellectual disability. Through ex vivo transplantation assays and single-cell RNA sequencing of the interhemispheric midline, we found that Tcf4-mediated neuron-astroglia communication is essential for est
